Lithium containing silicon carbonitride ceramics (SiCN:Li) were synthesized via precursor-to-ceramic-transformation of Li-containing (poly)silazanes. The precursors were obtained by lithiation of the model compound 2,4,6-trimethyl-2,4,6-trivinylcyclotrisilazane and of a commercial poly(organosilazane) VL20® with n-butyllithium in different molar ratios. According to Raman spectroscopic measurements, lithiation takes place at the NH groups of the molecular organosilazane structure. If the amount of n-BuLi exceeds the stoichiometric amount of NH groups, addition of n-BuLi at the vinyl groups (attached as substituents at the Si atoms of the silazane) occurs. Thermal analysis coupled with in situ mass spectrometry evidenced the loss of methane and hydrogen as the main gaseous by-products formed during the precursor-to-ceramic-transformation. The ceramization process is completed at 1100 °C in argon and yielded Li-containing silicon carbonitride, SiCN:Li. X-ray powder diffraction revealed that the resulting SiCN:Li ceramics were basically amorphous and contained LiSi2N3 as a crystalline phase with increasing amount of Li. Possible applications of the new SiCN:Li ceramics are seen in the field of Li-ion batteries as alternative anode or solid electrolyte material.  相似文献

Resting lymphocyte survival is dependent upon the expression of Bcl-2, yet the factors responsible for maintaining lymphocyte Bcl-2 protein expression in vivo are largely unknown. Natural killer (NK) cells are bone marrow-derived lymphocytes that constitutively express the beta and common gamma(c) subunits of the IL-2 receptor (R) as a heterodimer with intermediate affinity for IL-2. IL-15 also binds to IL-2Rbeta gamma(c) and is much more abundant in normal tissues than IL-2. Mice that lack the IL-2 gene have NK cells, whereas mice and humans that lack IL-2R gamma(c) do not have NK cells. Further, treatment of mice with an antibody directed against IL-2Rbeta results in a loss of the NK cell compartment. These data suggest that a cytokine other than IL-2, which binds to IL-2Rbeta gamma(c), is important for NK cell development and survival in vivo. In the current report, we show that the recently described IL-15R(alpha) subunit cooperates with IL-2Rbeta gamma(c) to transduce an intracellular signal at picomolar concentrations of IL-15. We demonstrate that resting human NK cells express IL-15R(alpha) mRNA and further, that picomolar amounts of IL-15 can sustain NK cell survival for up to 8 d in the absence of serum. NK cell survival was not sustained by other monocyte-derived factors (i.e., TNF-alpha, IL-1beta, IL-10, IL-12) nor by cytokines known to use gamma(c) for signaling (i.e., IL-4, IL-7, IL-9, IL- 13). One mechanism by which IL-15 promotes NK cell survival may involve the maintenance of Bcl-2 protein expression. Considering these functional properties of IL-15 and the fact that it is produced by bone marrow stromal cells and activated monocytes, we propose that IL-15 may function as an NK cell survival factor in vivo.  相似文献

While there is growing experimental evidence that cerebrospinal fluid (CSF) flow induced by the beating of ependymal cilia is an important factor for neuronal guidance, the respective contribution of vascular pulsation-driven macroscale oscillatory CSF flow remains unclear. This work uses computational fluid dynamics to elucidate the interplay between macroscale and cilia-induced CSF flows and their relative impact on near-wall dynamics. Physiological macroscale CSF dynamics are simulated in the ventricular space using subject-specific anatomy, wall motion and choroid plexus pulsations derived from magnetic resonance imaging. Near-wall flow is quantified in two subdomains selected from the right lateral ventricle, for which dynamic boundary conditions are extracted from the macroscale simulations. When cilia are neglected, CSF pulsation leads to periodic flow reversals along the ventricular surface, resulting in close to zero time-averaged force on the ventricle wall. The cilia promote more aligned wall shear stresses that are on average two orders of magnitude larger compared with those produced by macroscopic pulsatile flow. These findings indicate that CSF flow-mediated neuronal guidance is likely to be dominated by the action of the ependymal cilia in the lateral ventricles, whereas CSF dynamics in the centre regions of the ventricles is driven predominantly by wall motion and choroid plexus pulsation.  相似文献

Estuarine sediments are often highly enriched in particle-reactive metal contaminants and because aquatic animals have often been shown to acquire metals predominantly from their diet, benthic animals feeding on deposited or resuspended sediments may also accumulate metals through this uptake pathway. Laboratory experiments were performed in which the surface deposit-feeding polychaete, Nereis succinea, was exposed to As(+ 5), Cd, and Cr(+ 3) in pore water or in estuarine sediments with and without enrichment with algal debris. These experiments generated metal uptake parameters (assimilation efficiency of ingested metal [AE], uptake rate constant of dissolved metal, efflux rate constants following dietary or aqueous metal exposures) used in a kinetic model of metal bioaccumulation. The model showed that > 97% of the body burden of these metals is accumulated through ingested sediment. The kinetic model was further modified to consider the geochemical fractionation of the metals in the sediments because metals bound to some fractions were shown to be unavailable to these polychaetes. The modified model substituted the AE term for each metal by the percentage of metal extracted in neutral and weak acid exchangeable fractions (termed “carbonex” fraction) multiplied by the slope of the regression between the metal AE and its fractionation in carbonex. The modified model generated predictions of As, Cd, and Cr body burdens in polychaetes at three different estuarine sites that matched independent field observations at these sites (r2 = 0.84 for sediments without organic enrichment, r2 = 0.87 with organic enrichment). Model predictions that relied on total metal concentrations showed weaker relationships (r2 = 0.11-0.50). This study adds to the evidence for the dominance of dietary uptake of metals in aquatic animals and identifies a key sedimentary fraction of metals that can account for bioavailability of sediment-bound metals.  相似文献

Materials libraries of binary alloy nanoparticles (NPs) are synthesized by combinatorial co‐sputter deposition of Cu and Au into the ionic liquid (IL) 1‐butyl‐3‐methylimidazolium bis(trifluoromethylsulfonyl)imide ([C1C4im][Tf2N]), which is contained in a micromachined cavity array substrate. The resulting NPs and NP‐suspensions are investigated by transmission electron microscopy (TEM), X‐ray diffraction (XRD), UV‐Vis measurements (UV‐Vis), and attenuated total reflection Fourier transformed infrared (ATR‐FTIR) spectroscopy. Whereas the NPs can be directly observed in the IL using TEM, for XRD measurements the NP concentration is too low to lead to satisfactory results. Thus, a new NP isolation process involving capping agents is developed which enables separation of NPs from the IL without changing their size, morphology, composition, and state of aggregation. The results of the NP characterization show that next to the unary Cu and Au NPs, both stoichiometric and non‐stoichiometric Cu–Au NPs smaller than 7 nm can be readily obtained. Whereas the size and shape of the alloy NPs change with alloy composition, for a fixed composition the NPs have a small size distribution. The measured lattice constants of all capped NPs show unexpected increased values, which could be related to the NP/surfactant interactions.  相似文献

Precision cut liver slices from humans and rats were used to investigate the covalent binding of xenobiotics to the DNA by means of the (32)P-postlabeling technique. Human liver slices were incubated with the structurally related steroid hormones chlormadinone acetate (5 mu g/ml), cyproterone acetate (0.01-5 mu g/ml), megestrol acetate (5 mu g/ml), and the positive control 2-aminofluorene (0.01-5 mu g/ml), which is known for its marked ability to form DNA-adducts in vivo. Rat liver slices were incubated with cyproterone acetate in concentrations of 0.1, 1, and 5 mu g/ml. The functional viability and metabolic activity of the slices were shown to be sufficiently maintained during the incubation time by measurement of the intracellular K(+)-content and the metabolic turnover of the model substrate 7-ethoxycoumarin, respectively. All three test substances and the control induced DNA-adducts in human liver slices, however, with a different adduct pattern. While the total DNA-adduct levels obtained with cyproterone acetate and megestrol acetate were in the same order of magnitude (on average 1000 DNA-adducts/10(9) nucleotides after incubation with 5 mu g /ml), the relative adduct labeling calculated for chlormadinone acetate was about 400. Following in vitro incubation of rat liver slices with cyproterone acetate, the relative adduct labeling values increased proportionally with increasing concentrations and added linearily to in vivo generated DNA-adducts. At the level of liver slices, different DNA-adduct patterns were induced by cyproterone acetate in rat and man. In contrast to the finding of others, using rat hepatocytes, the relative adduct labeling values of cyproterone acetate and megestrol acetate were in the same order of magnitude after incubation with human liver slices. The present study indicates that liver slices are a useful tool to investigate the in vitro DNA-adduct inducing potential of xenobiotics.  相似文献

Microsensors provide instruments particularly suited for the rapid, noninvasive and on-line analysis of cell and tissue cultures. The microsensor system presented in this paper is a modular arrangement of various planar and nonplanar sensor elements for the measurement of physiological parameters of cell cultures. An optic access to the cultures (e.g. for light microscopy and spectrophotometric techniques) is also provided for a parallel and comparative data acquisition. The system was originally designed for biomedical research in chemotherapy (predicative chemotherapy assays) and pharmacology but it turned out to be also an effective tool for toxicological and environmental research.  相似文献
