We have used fluorescent 7-nitrobenz-2-oxa-1,3-diazol-4-yl (NBD)-tagged phospholipid analogues, NBD-PE (phosphatidylethanolamine), NBD-PC (phosphatidylcholine) and NBD-PS (phosphatidylserine), to demonstrate that Cdr1p and its other homologues, Cdr2p and Cdr3p, belonging to the ATP-binding cassette (ABC) superfamily behave as general phospholipid translocators. Interestingly, CDR1 and CDR2, whose overexpression leads to azole resistance in C. albicans, elicit in-to-out transbilayer phospholipid movement, while CDR3, which is not involved in drug resistance, carries out-to-in translocation of phospholipids between the two monolayers of plasma membrane. Cdr1p, Cdr2p and Cdr3p could be further distinguished on the basis of their sensitivities to different inhibitors. For example, the in-to-out activity associated with Cdr1p and Cdr2p is energy-dependent and sensitive to sulphydryl blocking agents such as N-ethylmaleimide (NEM) and cytoskeleton disrupting agent cytochalasin E, while Cdr3p-associated out-to-in activity is energy-dependent but insensitive to NEM and cytochalasin E. We found that certain drugs, such as fluconazole, cycloheximide and miconazole, to which Cdr1p confers resistance could also affect in-to-out transbilayer movement of NBD-PE, while the same drugs had no effect on Cdr3p-mediated out-to-in translocation of NBD-PE. The ineffectiveness of these drugs to affect Cdr3p mediated out-to-in phospholipid translocation further confirms the inherent difference in the directionality of phospholipid translocation between these pumps. Notwithstanding the role of some of the Cdrps in drug resistance, this study clearly demonstrates that these ABC transporters of C. albicans are phospholipid translocators and this function could represent one of the physiological functions of such large family of proteins. 相似文献

Curcumin, the principal curcuminoid of turmeric, is responsible for its yellow colour and serves as a measure of turmeric quality. The Prevention of Food Adulteration (PFA) Act of India allows only Curcuma longa L. for the production of turmeric powder and prohibits addition of any foreign matter/artificial colour, but it does not specify a minimum curcumin content. The present surveillance was undertaken to study the quality of loose versus branded turmeric powders vis-à-vis curcumin content and the presence of unwarranted extraneous colours from city markets in India using a newly developed two-dimensional high-performance thin-layer chromatography (2D-HPTLC) method. The results show that curcumin content in branded samples ranged from 2.2% to 3.7%, while non-branded samples had from 0.3% to 2.6%. Though none of the branded turmeric powders contained artificial colours, 17% of loose powders showed the presence of extraneous colour metanil yellow, in the range 1.0–8.5 mg g?1, which may pose health threats. Low curcumin content in the analysed samples may be due to mixing of other curcuma species or their curcumin-depleted matrices and foreign starches as cheaper alternatives. This is supported by the fact that major Indian turmeric trade types are known to possess curcumin contents ranging from 2.1% to 8.6%, with an average of 4.8%. There is thus an urgent need to prescribe realistic curcumin limits for turmeric powder, otherwise there is no obligation on the part of traders to stick to any minimum levels and consumers will keep on getting this nutrient-depleted household spice. 相似文献

ABSTRACT Supply chains are becoming increasingly complex, and many companies and researchers are looking to nature for finding solutions. In this paper, we provide a narrative synthesis and systematic review to explore state-of-the-art, and identify key areas for exploration of nature inspired solutions in the supply chain area. We consider strategic, tactical, and operational decisions within supply chains and identify analogies between biological systems and supply chains to create a new research agenda. Our findings show that the majority of the research in nature inspired supply chains from the systemic perspective focuses on closed loop supply chains. In contrast, similar systemic solutions such as industrial symbiosis, circular economy, and industrial ecology have not been explored as frequently. Reducing production waste (industrial symbiosis) in addition to end-of-life product waste (closed loop supply chains), optimising raw material use and extending the life of used resources (circular economy), investigating the interactions between biosphere and technosphere (industrial ecology), and seeking solutions to industrial problems from nature (biomimicry) are all possible paths for reaching a zero-waste economy. These systemic solutions have a considerable impact on the supply chains at the strategic, tactical, and operational levels, which need to be explored to identify efficient and effective solutions. 相似文献

Displaying information on transparent screens offers new opportunities in next‐generation electronics, such as augmented reality devices, smart surgical glasses, and smart windows. Outstanding luminance and transparency are essential for such “see‐through” displays to show vivid images over clear background view. Here transparent quantum dot light‐emitting diodes (Tr‐QLEDs) are reported with high brightness (bottom: ≈43 000 cd m?2, top: ≈30 000 cd m?2, total: ≈73 000 cd m?2 at 9 V), excellent transmittance (90% at 550 nm, 84% over visible range), and an ultrathin form factor (≈2.7 µm thickness). These superb characteristics are accomplished by novel electron transport layers (ETLs) and engineered quantum dots (QDs). The ETLs, ZnO nanoparticle assemblies with ultrathin alumina overlayers, dramatically enhance durability of active layers, and balance electron/hole injection into QDs, which prevents nonradiative recombination processes. In addition, the QD structure is further optimized to fully exploit the device architecture. The ultrathin nature of Tr‐QLEDs allows their conformal integration on various shaped objects. Finally, the high resolution patterning of red, green, and blue Tr‐QLEDs (513 pixels in.?1) shows the potential of the full‐color transparent display. 相似文献

Present investigations deal with development of a novel variable compression ratio (VCR) mechanism and its implementation in a small and relatively large size single-cylinder engines. Operation of this mechanism is found to be smooth and effective in the running condition of the engine as well. This mechanism, when incorporated in the small size spark ignition HONDA engine, portrayed improvement in engine performance with increment in compression ratio (CR) for petrol and kerosene. Their respective optimum CRs 5.02 (petrol) and 5.27 (kerosene) are higher than the base value 4.8. In case of large size KIRLOSKAR engine, the present VCR mechanism is found to be useful while operating with liquefied petroleum gas (LPG), where measurements showed that combustion duration is lower with LPG for CR 9.79 as compared with base value 9.0. The present experiments clearly demonstrate the usefulness of VCR mechanism in improving engine performance for a given fuel and broadening the range of alternative fuels burnt in the engine. Ease of fabrication, simplicity in installation, accessibility in troubleshooting and smooth run-time alterations are the advantages with the current novel mechanism. 相似文献

Black phosphorus (BP) with unique 2D structure enables the intercalation of foreign elements or molecules, which makes BP directly relevant to high‐capacity rechargeable batteries and also opens a promising strategy for tunable electronic transport and superconductivity. However, the underlying intercalation mechanism is not fully understood. Here, a comparative investigation on the electrochemically driven intercalation of lithium and sodium using in situ transmission electron microscopy is presented. Despite the same preferable intercalation channels along [100] (zigzag) direction, distinct anisotropic intercalation behaviors are observed, i.e., Li ions activate lateral intercalation along [010] (armchair) direction to form an overall uniform propagation, whereas Na diffusion is limited in the zigzag channels to cause the columnar intercalation. First‐principles calculations indicate that the diffusion of both Li and Na ions along the zigzag direction is energetically favorable, while Li/Na diffusion long the armchair direction encounters an increased energy barrier, but that of Na is significantly larger and insurmountable, which accounts for the orientation‐dependent intercalation channels. The evolution of chemical states during phase transformations (from LixP/NaxP to Li3P/Na3P) is identified by analytical electron diffraction and energy‐loss spectroscopy. The findings elucidate atomistic Li/Na intercalation mechanisms in BP and show potential implications for other similar 2D materials. 相似文献

Traditional relational database systems handle data by dividing their memory into sections such as a buffer cache and working memory, assigning a memory budget to each section to efficiently manage a limited amount of overall memory. They also assign memory budgets to memory-intensive operators such as sorts and joins and control the allocation of memory to these operators; each memory-intensive operator attempts to maximize its memory usage to reduce disk I/O cost. Implementing such memory-intensive operators requires a careful design and application of appropriate algorithms that properly utilize memory. Today's Big Data management systems need the ability to handle large amounts of data similarly, as it is unrealistic to assume that truly big data will fit into memory. In this article, we share our memory management experiences in Apache AsterixDB, an open-source Big Data management software platform that scales out horizontally on shared-nothing commodity computing clusters. We describe the implementation of AsterixDB's memory-intensive operators and their designs related to memory management. We also discuss memory management at the global (cluster) level. We conducted an experimental study using several synthetic and real datasets to explore the impact of this work. We believe that future Big Data management system builders can benefit from these experiences. 相似文献

Nanomaterials have potential medical applications, for example in the area of drug delivery, and their possible adverse effects and cytotoxicity are curently receiving attention. Inhalation of nanoparticles is of great concern, because nanoparticles can be easily aerosolized. Imaging techniques that can visualize local populations of nanoparticles at nanometre resolution within the structures of cells are therefore important. Here we show that cells obtained from mice exposed to single-walled carbon nanohorns can be probed using a scanning probe microscopy technique called scanning near field ultrasonic holography. The nanohorns were observed inside the cells, and this was further confirmed using micro Raman spectroscopy. Scanning near field ultrasonic holography is a useful technique for probing the interactions of engineered nanomaterials in biological systems, which will greatly benefit areas in drug delivery and nanotoxicology. 相似文献
