Doxorubicin (Dox) is one of the most widely used treatments for breast cancer, although limited by the well-documented cardiotoxicity and other off-target effects. Mesenchymal stem cell (MSC) secretome has shown immunomodulatory and regenerative properties, further potentiated under 3D conditions. This work aimed to uncover the effect of the MSC-derived secretome from 3D (CM3D) or 2D (CM2D) cultures, in human malignant breast cells (MDA-MB-231), non-tumor breast epithelial cells (MCF10A) and differentiated AC16 cardiomyocytes, co-treated with Dox. A comprehensive proteomic analysis of CM3D/CM2D was also performed to unravel the underlying mechanism. CM3D/CM2D co-incubation with Dox revealed no significant differences in MDA-MB-231 viability when compared to Dox alone, whereas MCF10A and AC16 viability was consistently improved in Dox+CM3D-treated cells. Moreover, neither CM2D nor CM3D affected Dox anti-migratory and anti-invasive effects in MDA-MB-231. Notably, Ge-LC-MS/MS proteomic analysis revealed that CM3D displayed protective features that might be linked to the regulation of cell proliferation (CAPN1, CST1, LAMC2, RANBP3), migration (CCN3, MMP8, PDCD5), invasion (TIMP1/2), oxidative stress (COX6B1, AIFM1, CD9, GSR) and inflammation (CCN3, ANXA5, CDH13, GDF15). Overall, CM3D decreased Dox-induced cytotoxicity in non-tumor cells, without compromising Dox chemotherapeutic profile in malignant cells, suggesting its potential use as a chemotherapy adjuvant to reduce off-target side effects.  相似文献

Fibers in polymer composites can be designed in various orientations for their usage in service life. Various fiber orientated polymer composites, which are used in aeroplane and aerospace applications, are frequently subjected to thermal cycles because of the changes in body temperatures at a range of −60 to 150°C during flights. It is an important subject to investigate the visco‐elastic properties of the thermal cycled polymer composite materials which have various fiber orientations during service life. Continuous fiber reinforced composites with a various fiber orientations are subjected to 1,000 thermal cycles between the temperatures of 0 and 100°C. Dynamic mechanic thermal analysis (DMTA) experiments are carried out by TA Q800 type equipment. The changes in glass transition temperature (Tg), storage modulus (E′), loss modulus (E′′) and loss factor (tan δ) are inspected as a function of thermal cycles for different fiber orientations. It was observed that thermal and dynamic mechanical properties of the polymer composites were remarkably changed by thermal cycles. It was also determined that the composites with [45°/−45°]s fiber orientation presented the lowest dynamic mechanical properties. POLYM. COMPOS., 2010. © 2009 Society of Plastics Engineers  相似文献

The development of a novel surface-enhanced resonance Raman scattering (SERRS) platform that allows fast and sensitive detection of dopamine (DA) has been reported. The iron-nitrilotriacetic acid attached silver nanoparticle (Ag-Fe(NTA)) substrate provides remarkable sensitivity and reliable repeatability. The advantages of both the surface functionalization for specific analytes and the SERRS are integrated into a single functional unit. While the silver core gives the necessary enhancing properties, the Fe-NTA receptors can trap DA adjacent the silver core and the NTA-Fe-DA complex formed provides resonance enhancement with a 632.8 nm laser. DA could be detected in pM level without any pretreatment with a reliable discrimination against AA, by utilizing low laser power (10 mW) and short data acquisition time (10 s). The high sensitivity along with the improved selectivity of this sensing approach is a significant step toward molecular diagnosis of Parkinson's disease.  相似文献

To fully understand the properties of functional nanostructures such as catalytic nanoclusters, it is necessary to know the positions of all the atoms in the nanostructure. The catalytic properties of metal nanoclusters can often be improved by the addition of a second metal, but little is known about the role of the different metals in these bimetallic catalysts, or about their interactions with each other and the support material. Here we show that aberration-corrected scanning transmission electron microscopy of supported rhodium-iridium clusters, combined with dynamic multislice image simulations, can identify individual atoms, map the full structure, and determine changes in the positions of metal atoms in sequential images. This approach could help in the development of new and improved catalysts and other functional nanostructures.  相似文献

The primary purpose of the study is to investigate the temperature dependence of heat capacity and thermal conductivity of composites having different fiber/matrix combinations by means of heat‐flux differential scanning calorimetry (DSC). The materials used as samples in this study were epoxy‐ and polyester‐based composites. Noncrimp stitched glass, carbon, and aramid fabric were used as reinforcements for making unidirectional composites. For the heat capacity measurements the composite sample and a standard material are separately subjected to same linear temperature program. By recording the heat flow rate into the composite sample as a function of temperature, and comparing it with the heat flow rate into a standard material under the same conditions, the temperature dependence of heat capacity of the composite sample is determined. Measurements were carried out over a wide range of temperatures from about 20 to 250°C. The differential scanning calorimeter was adapted to perform the thermal conductivity measurements in the direction perpendicular to the fiber axis over the temperature range of 45–235°C. The method used in this study utilizes the measurement of rate of heat flow into a sensor material during its first‐order phase transition to obtain the thermal resistance of a composite material placed between the sensor material and the heater in the DSC. Polyester‐ and epoxy‐based composites containing glass and carbon fibers were manufactured using a vacuum‐assisted resin transfer molding (VARTM) process. Fourier transform infrared (FTIR) spectroscopy analyses were conducted to determine the interaction between fibers and matrix material. The results indicate that strong interaction was observed between carbon fiber and epoxy resin. However, weak interactions between remaining fiber‐matrix occur. Scanning electron microscopy (SEM) analysis was also performed to take some information about strength of interaction between fibers and matrix material. From SEM micrographs, it is concluded that the findings in SEM analysis support to that obtained in FTIR analysis. Another aim of the present work was to investigate the influence of matrix on composite properties. Hence, the strengths of composites having same reinforcement but different matrix systems in axial tension and transverse tension were compared. Conducting polymers containing polyether pseudocages (P I , P II , P III ) have been synthesized via chemical oxidation of 1,5‐bis(1,1‐pyrrole)‐3‐oxabutane (M I ), 1,8‐bis(1,1‐pyrrole)‐3,6‐dioxahexane (M II ), and 1,11‐bis(1,1‐pyrrole)‐3,6,9‐trioxaundecane (M III ) using anhydrous FeCl3 in CHCl3. Because as obtained polymer resins did not give any response toward any cations, they were reduced (undoped) using chemical reducing agents. Tetrabutylammonium hydroxide was found to be more effective in undoping to obtain more reproducible and reusable polymer resins. The undoped polymer resins were tried in the extraction of rare earth metal ions from the aqueous medium. Among them, only P III resin removes La(III), Eu(III) and Yb(III) and can be employed for the preconcentration of these metal ions. For batch extraction of La(III), Eu(III) and Yb(III) at neutral pH values, percent recoveries of 98.0 ± 1.0, 90.7 ± 1.4, 87.3 ± 4.0, respectively, has been obtained. The sorption capacity is found as 1.3 mg of La(III) per gram of P III resin. The formation of an integral asymmetric membrane composed of a cylinder‐forming polystyrene‐block‐poly(2‐vinylpyridine) on a nonwoven by using solvent casting followed by solvent/nonsolvent exchange (phase inversion) is reported for the first time. The influence of parameters such as solvent composition, evaporation time of the solution‐cast block copolymer film before phase inversion, and immersion bath temperature is demonstrated. The optimized membranes are characterized in terms of stimuli‐responsive water flux properties. The morphologies of the membranes as well as of the bulk of the block copolymer are imaged by scanning force microscopy, scanning electron microscopy, and transmission electron microscopy.

Tungsten trioxide hydrate (3WO(3)·H(2)O) films with different morphologies were directly grown on fluorine doped tin oxide (FTO) substrate via a facile crystal-seed-assisted hydrothermal method. Scanning electron microscopy (SEM) analysis showed that 3WO(3)·H(2)O thin films composed of platelike, wedgelike, and sheetlike nanostructures could be selectively synthesized by adding Na(2)SO(4), (NH(4))(2)SO(4), and CH(3)COONH(4) as capping agents, respectively. X-ray diffraction (XRD) studies indicated that these films were of orthorhombic structure. The as-prepared thin films after dehydration showed obvious photocatalytic activities. The best film grown using CH(3)COONH(4) as a capping agent generated anodic photocurrents of 1.16 mA/cm(2) for oxidization of methanol and 0.5 mA/cm(2) for water splitting with the highest photoconversion efficiency of about 0.3% under simulated solar illumination.  相似文献
