Introduction of influenza viruses with gene segments of avian origin into the human population may result in the emergence of new pathogenic human influenza viruses. The recent infection of a 3-year-old boy with an influenza A (H5N1) virus of avian origin can be considered as an example of such an event. However, this virus, influenza A/Hong Kong/156/97 (H5N1) and the 17 additional H5N1 viruses isolated from humans by the end of 1997 lack the ability to spread efficiently amongst humans and therefore have limited pandemic potential. However, the possibility of reassortment of these viruses with currently circulating human viruses illustrates the need for pandemic preparedness.  相似文献

Using a polymerase chain reaction strategy we identified a serine proteinase inhibitor (serpin) in human bone marrow that is related to the cellular serpin proteinase inhibitor 6 (PI-6) and the viral serpin cytokine response modifier A (CrmA). This serpin, proteinase inhibitor 9 (PI-9), has an unusual reactive center P1(Glu)-P1'(Cys), which suggests that it inhibits serine proteinases that cleave after acidic residues. The only known serine proteinase with this specificity is granzyme B, a granule cytotoxin produced by cytotoxic lymphocytes. To test the interaction of PI-9 with granzyme B we prepared recombinant hexa-histidine tagged PI-9 in a yeast expression system. Addition of the recombinant protein to native granzyme B resulted in an SDS-resistant complex typical of serpin-serine proteinase interactions. Further analysis showed that complex formation followed bimolecular kinetics with a second order rate constant of 1.7 +/- 0.3 x 10(6) M-1 s-1, which is in the range for a physiologically significant serpin-proteinase interaction. Recombinant PI-9 also completely abrogated granzyme B and perforin-mediated cytotoxicity in vitro. Examination of PI-9 mRNA distribution demonstrated that it is expressed in immune tissue, primarily in lymphocytes. The highest levels of PI-9 mRNA and protein were observed in natural killer cell leukemia cell lines and in interleukin-2 stimulated peripheral blood mononuclear cells, which also produce granzyme B. Like PI-6, PI-9 was shown to be a cytosolic protein that is not secreted. Fractionation of natural killer cells and stimulated peripheral blood mononuclear cells demonstrated that PI-9 is in a separate subcellular compartment to granzyme B. These results suggest that PI-9 serves to inactivate misdirected granzyme B following cytotoxic cell degranulation. This may explain why cytotoxic cells are not damaged by their own granzyme B during destruction of abnormal cells.  相似文献

The support of broadband multimedia applications over an enterprise network poses a broad range of networking challenges for efficient resource management, intelligent switching, and access control for distributed information. We propose two server-based scheduling algorithms, with low computational complexity, to guarantee multimedia information synchronization at the destination, with minimum presentation delays and buffer requirements. We outline a communication framework, highlighting the issues and challenges faced by today's enterprise networks to support multimedia services. We describe the proposed connection establishment and resource allocation schemes in resource-constrained enterprise networks. The objective is to manage the limited resources of the network for maximum utilization. Towards this end we present a dynamic capacity allocation scheme to support connections for multiple users. Specifically, we show that the channel capacity allocation problem can be formulated as a quadratic programming problem. This allocation scheme is implemented at each intermediate switch to dynamically determine the capacity allocation. The effects of interswitch rate mismatch and network delay offset scheduling have also been incorporated in the management framework. OBJECTIVE: To answer the question whether positive selection of (pre)schizophrenic Surinams in Surinam before emigration to the Netherlands can explain the high admission rates for schizophrenia of Surinams in the Netherlands. DESIGN: Retrospective research. SETTING: 's Lands Psychiatrisch Ziekenhuis (National Psychiatric Hospital) Paramaribo, Surinam (LPZ). METHOD: The Netherlands Central Bureau of Statistics in Voorburg and the Central Registry Office in Paramaribo provided data on emigration to the Netherlands of the Surinam population and on remigration. Files of (pre)schizophrenic patients were selected from the records of LPZ. Subsequently, these patients were traced and their pattern of migration was studied. Using the chi 2 test the significance of observed differences was assessed. RESULTS: Surinam (pre)schizophrenic patients did not emigrate more frequently to the Netherlands (6.3%) compared with the Surinam population in general (9.6%) and they remigrated more frequently to Surinam than the Surinams in general. CONCLUSION: The positive selection hypothesis must be rejected.  相似文献

The static respiration activity index test was designed to assess the biodegradability (or biostability) of compost samples by monitoring their oxygen consumption rate under cultivation conditions. However, as revealed in this study, the measured activity index correlated not only with the biochemical characteristics of the tested sample but also with the operational conditions, including sample quantity, sample moisture content and cultivation temperature. It was speculated that this observation could be attributed to the limited oxygen transport to the sample. Hence the oxygen transport and consumption in the tested sample were modelled using a one‐dimensional, pseudo‐steady state model to estimate the relative significance of mass transfer resistance to the bioreaction rate. A so‐called critical thickness of the sample, above which mass transfer resistance became significant, was identified and assessed under various experimental conditions. Zr-rich PZT thin films were synthesized by metallorganic decomposition and their dielectric and pyroelectric properties were investigated with different ratios of zirconium/titanium and poling condition. All the films became effectively (1 1 1) textured and well crystallized at the annealing temperature of 700 °C. With increasing Zr content, coercive field increased and voltage dependent capacitance curve appeared asymmetrical, indicating the presence of antiferroelectric phase, PbZrO3, in film composition. The pyroelectric coefficient in the practically applicable temperature ranges of 20–60 °C was found to be maximum for the thin film with 0.85 mol of zirconium in PZT. Further increase in zirconium content led to severe deterioration in pyroelectric properties. The values of pyroelectric coefficient and figures of merit were greatly influenced by poling direction and temperature. The result was explained in terms of electric phase and state of polarization in film.  相似文献

In patients undergoing somatostatin receptor scintigraphy, treatment with octreotide (Sandostatin) is usually discontinued 24-48 h before and after injection with the radioligand 111In-pentetreotide ([111In-DTPA(O)]octreotide) (Octreoscan) because octreotide competes with radioligand for the same receptors. However, D?rr et al. and Soresi et al. reported improved visualization of carcinoid and small cell lung cancer lesions, respectively, during continued octreotide treatment. We found that intravenous administration of unlabeled octreotide to rats inhibited the binding of an optimal dose (0.5 microg) of 111In-pentetreotide to somatostatin receptors in pancreas and adrenals in a mass- and time-dependent way. Pretreatment with unlabeled octreotide never increased receptor binding of 111In-pentetreotide. Administration of 100 microg of octreotide decreased receptor-bound radioactivity if given simultaneously with or 10 or 20 min after injection of the radioligand, but had no effect if given 30 min after the radioligand. These findings indicate rapid processing of receptor-bound octreotide and suggest that octreotide treatment of patients undergoing 111In-pentetreotide scintigraphy may be reinitiated as soon as 1 h after radioligand administration.  相似文献
