Polyphasic characterization of a PCP-to-phenol dechlorinating microbial community enriched from paddy soil 总被引:4,自引:0,他引:4

Dechlorination of PCP has been observed previously under anaerobic condition in paddy soil. However, there is poor information about the dechlorination pathway of PCP and the microbial community associated with the PCP dechlorination in paddy soil. In this study, an anaerobic microbial community dechlorinating PCP was enriched by serial transfers from a paddy soil using a medium containing PCP, lactate and the steam-sterilized paddy soil. The enriched microbial community dechlorinated PCP completely to phenol under the anaerobic condition by a dechlorinating pathway as follows; PCP-->2,3,4,5-tetrachlorophenol-->3,4,5-trichlorophenol-->3,5-dichlorophenol-->3-chlorophenol-->phenol. Intermediate products such as 3-chlorophenol were not accumulated, which were immediately dechlorinated to phenol. The enriched microbial community was characterized physiologically by testing the effects of electron donors and electron acceptors on the dechlorinating activity. The dechlorinating activity was promoted with lactate, pyruvate, and hydrogen as electron donors but not with acetate. Electron acceptors, nitrate and sulphate, inhibited the dechlorinating activity competitively but not iron (III). The microbial group associated with the anaerobic dechlorination was characterized by the effect of specific inhibitors on the PCP dechlorination. Effects of specific metabolic inhibitors and antibiotics indicated the involvement of Gram-positive spore-forming bacteria with the PCP dechlorinating activity, which was represented as bacteria of phylum Firmicutes. The structure of the microbial community was characterized by fluorescence in situ hybridization, quinone profiling, and PCR-DGGE (denaturing gel gradient electrophoresis). The combined results indicated the predominance of Clostridium species of phylum Firmicutes in the microbial community. Desulfitobacterium spp. known as anaerobic Gram-positive spore-forming bacteria dechlorinating PCP were not detected by PCR using a specific primer set. We constructed a replication-defective retroviral vector plasmid for the expression of a single-chain antibody fragment (scFv), derived from a chicken anti-human prion protein monoclonal antibody, fused with the Fc region of human IgG1. CHO-K1 and NS-1 cells were transformed with the viral vector pseudotyped with vesicular stomatitis virus G protein (VSV-G), and scFv-Fc producer clones were established. Among the established clones, CHO-2A9 cells produced a large amount of the product with an antibody-like dimerized structure in serum-free culture that facilitated the purification of scFv-Fc. The scFv-Fc specifically recognized the epitope sequence of prion protein in solid-phase enzyme-linked immunosorbent assay (ELISA) and Western blot analysis. The injection test into quails revealed that the scFv became more stable in vivo by fusion with the Fc region. The scFv-Fc will be a useful tool for the detection of mammalian prion proteins. 相似文献

Microbial analyses by fluorescence in situ hybridization of well-settled granular sludge in brewery wastewater treatment plants 总被引:3,自引:0,他引:3
The characteristics of granular sludge from full-scale upflow anaerobic sludge blanket reactors used for the treatment of brewery wastewater were investigated. Fluorescence in situ hybridization (FISH) analyses of settled granules from a reactor that had been treating brewery wastewater stably at COD removal rates of over 90% for more than 6 months showed that a methanogen of the genus Methanosaeta was predominant near the granule surface and that Bacteria were not abundant. The center of the granules was composed of dead or resting cells, or both, which were used as a support for active archaeal and bacterial cells near the surface. Periodic analysis of granules from full-scale plants showed that granules containing methanogens deep within them tended to float. Granules with a Bacteria layer on the surface also tended to float. On the basis of these findings, well-settled granules are considered to have methanogens that develop near the granule surface so that the gases generated during methane fermentation are readily released. 相似文献

The effects of soy protein isolate (SPI) on the gel properties of different grade Alaska pollock and common carp surimi at different setting conditions were evaluated and compared. Breaking force and distance of gels decreased with increasing SPI concentrations in direct cook (85 °C for 30 min) and in cook after setting at 30 °C for 60 min conditions. The effect of SPI on gel strength of common carp surimi was less than in Alaska pollock surimi. The breaking force obtained for addition of 10% SPI to Alaska pollock surimi was higher than for surimi alone when cooked after incubation at 50 °C for 60 min. Addition of SPI decreased the whiteness and increased the yellowness of the gel. The gel structure showed that the addition of SPI modified the microstructure of the fish protein gel, thus resulting in surimi with different gelling properties. This paper investigates the validity of the method used in the Japanese offshore wind map (NeoWins) to seamlessly connecting satellite‐derived wind speed for open oceans to mesoscale model‐simulated wind speed for coastal waters. In the NeoWins, the former was obtained from the satellite‐borne Advanced Scatterometer (ASCAT), and the latter was obtained from numerical simulations using the Weather Research and Forecasting (WRF) model. In this study, the consistency of the ASCAT and WRF 10‐m height wind speeds is examined in their overwrapping areas. The comparison between ASCAT and WRF model reveals that their differences in annual mean wind speed are mostly within ±5% and that the ASCAT annual mean wind speed is, as a whole, slightly higher than the WRF annual mean wind speed. The results indicate that there are no large wind speed gaps between WRF and ASCAT in most parts of the Japanese offshore areas. It is moreover found that the discrepancies between the two wind speeds are due to two factors: one is the existence of winter sea ice in the ASCAT observation in the Sea of Okhotsk in ASCAT observation and the other is that the accuracy of the WRF wind speed depends on atmospheric stability. 相似文献

Low malt beers have high sales volumes in Japan, but improving their mouthfeel, including softness, smoothness and decreasing astringency, is challenging because the compounds responsible remain unclear. In this study, beer was fractionated by preparative size‐exclusion chromatography, with the polypeptide and maltodextrin fractions purified using solid‐phase extraction and ion‐exchange resin. Sensory data from a spike test showed that the mouthfeel (softness, smoothness, and reduced astringency) of low malt beer was improved both by the degree of polymerisation (DP) of maltodextrins (DP of 2‐10; at increased concentration of 40 to 60%; P < 0.01) and by 10 ‐ 20 kilodalton (kDa) high molecular weight (HMW) polypeptide and 2‐3 kDa low molecular weight polypeptide fractions (at a 50% increase in concentration; P < 0.01). Furthermore, highly purified 10 to 20 kDa HMW polypeptides improved the softness and smoothness and decreased the astringency (at a 25% increase in concentration). This report is the first to provide experimental sensory data indicating that HMW polypeptides improve the mouthfeel of beer. Based on these findings, a new low malt beer was developed that showed significantly higher levels of the 10‐20 kDa HMW polypeptides with an overall improved mouthfeel. Mass spectrometric analysis of the 10 to 20 kDa proteins identified several unique foam positive proteins, including barley dimeric alpha‐amylase inhibitor‐1 and non‐specific lipid‐transfer protein 1. The surface of a glassy carbon electrode was modified with a DNA-alginate complex film in which water-soluble DNA was encapsulated into a calcium-alginate gel. The resulting modified electrode (DAFE) was used to detect ethidium bromide (EtBr), after its accumulation on the electrode. The intercalative behavior of EtBr for dsDNA in the film was investigated by measuring the electrode response based on the intercalation of EtBr separated from nonspecific interactions (electrostatic interaction). The accumulation of EtBr in the dsDAF was enhanced by applying a negative potential below -200 mV at the dsDAFE. When a positive potential above +700 mV was applied to the dsDAFE for a constant time with stirring in a Tris buffer solution, the amount accumulated decreased. These results indicate that it is possible to electrochemically control the accumulation and release of EtBr when a dsDAFE is used. In addition, the accumulation and detection of EtBr in spiked river water samples and daunomycin, an antitumor agent, is described. 相似文献

The effects of C/N ratio and total organic carbon (TOC) loading on nitrogen removal through simultaneous nitrate reduction and anaerobic ammonium oxidation in a single reactor were examined. Granular sludge taken from a methane fermentation reactor was placed in an upflow reactor and supplied with synthetic wastewater containing nitrate at a C/N ratio of 1 to grow heterotrophic denitrifying bacteria. When nitrogen removal ratio reached 30%, anammox sludge attached to nonwoven-carrier was added into the same reactor and then ammonia was added to the synthetic wastewater. Nitrogen removal ratio was markedly increased to 80-94%. In this system, nitrogen removal ratio was affected by C/N ratio and TOC loading, not by the amount of granular sludge. A stable isotopic analysis using 15N-labeled nitrate showed that N2 gas was formed by anammox reaction. 相似文献
