Samples of fine particulate matter were collected in a roadway tunnel near Houston, TX over a period of 4 days during two separate sampling periods: one sampling period from 1200 to 1400 local time and another sampling period from 1600 to 1800 local time. During the two sampling periods, the tunnel traffic contained roughly equivalent numbers of heavy-duty diesel trucks. However, during the late afternoon sampling period, the tunnel contained twice as many light-duty gasoline-powered vehicles. The effect of this shift in the vehicle fleet affects the overall emission index (grams pollutant emitted per kilogram carbon in fuel) for fine particles and fine particulate elemental carbon. Additionally, this shift in the fraction of diesel vehicles in the tunnel is used to determine if the chemical mass balancing techniques used to track emissions from gasoline-powered and diesel-powered emissions accurately separates these two emission categories. The results show that the chemical mass balancing calculations apportion roughly equal amounts of the particulate matter measured to diesel vehicles between the two periods and attribute almost twice as much particulate matter in the late afternoon sampling period to gasoline vehicles. Both of these results are consistent with the traffic volume of gasoline and diesel vehicles in the tunnel in the two separate periods and validate the ability for chemical mass balancing techniques to separate these two primary sources of fine particles.  相似文献

A laboratory intercomparison of organic carbon (OC) and elemental carbon (EC) measurements of atmospheric particulate matter samples collected on quartz filters was conducted among eight participants of the ACE-Asia field experiment The intercomparison took place in two stages: the first round of the intercomparison was conducted when filter samples collected during the ACE-Asia experiment were being analyzed for OC and EC, and the second round was conducted after the ACE-Asia experiment and included selected samples from the ACE-Asia experiment Each participant operated ECOC analyzers from the same manufacturer and utilized the same analysis protocol for their measurements. The precision of OC measurements of quartz fiber filters was a function of the filter's carbon loading but was found to be in the range of 4-13% for OC loadings of 1.0-25 microg of C cm(-2). For measurements of EC, the precision was found to be in the range of 6-21% for EC loadings in the range of 0.7-8.4 microg of C cm(-2). It was demonstrated for three ambient samples, four source samples, and three complex mixtures of organic compounds that the relative amount of total evolved carbon allocated as OC and EC (i.e., the ECOC split) is sensitive to the temperature program used for analysis, and the magnitude of the sensitivity is dependent on the types of aerosol particles collected. The fraction of elemental carbon measured in wood smoke and an extract of organic compounds from a wood smoke sample were sensitive to the temperature program used for the ECOC analysis. The ECOC split for the three ambient samples and a coal fly ash sample showed moderate sensitivity to temperature program, while a carbon black sample and a sample of secondary organic aerosol were measured to have the same split of OC and EC with all temperature programs that were examined.  相似文献

Transglutaminase (TGase) was separated from the culture broth of an isolated strain of Streptoverticillium mobaraense. The crude enzyme was prepared by centrifugation, ultrafiltration, precipitation by alcohol, centrifugation and freeze‐drying. The yield after these processes was 65–70%. Then the enzyme was purified to homogeneity by chromatography on CM‐cellulose and Sephadex G‐75 on which the yields were about 70% and 80%, respectively; the purified folds reached 2.5–4.7 and 1.08–2.06, respectively. The molecular weight of this TGase was 39,500–40,100 Da by gel filtration chromatography. Optimum enzyme activity was observed in the pH range of 5.0–7.0, and it was maintained stable at 20–40C. The optimal temperature and pH was 52C and 6.0, respectively. At 1 mM and 5 mM metal ion or inhibitors concentration, TGase activity was strongly inhibited by Zn2+ and NEM, and not affected obviously by Ba2+, Ca2+, Co2+, Cu2+, Fe2+, Fe3+, Mg2+, Mn2+, Na+ as well as PMSF and EDTA. The effects of these additions on this TGase were compared with those of other microbial TGases.  相似文献

Sliced cooked ham was vacuum packaged and held in the dark at −15C. 10C, and 30C, and at 10C in the light, for up to 96 h. At 24 h intervals, samples were analyzed for pigment concentration by spectrophotometry. A decrease in pigment concentration was observed over time under all conditions. Pigment degradation increased at higher temperatures and with light exposure. Kinetic parameters were determined, indicating that pigment degradation follows first order kinetics and the temperature dependence of these reactions indicated an Arrhenius relationship. The activation energy was 3.3 kcal/mol. The visible spectra for the extracted pigments in cooked ham showed the typical pattern of nitrosylmyoglobin. Pigments extracted from ham displayed in the light had spectra with similar absorption maxima to those from those stored in the dark.  相似文献

This study determined quality changes of whole ungutted golden gray mullet (Liza aurata) while stored in ice or in a refrigerator (without ice). Changes in microbiological quality (total viable and psychrophilic counts, lactic acid bacteria and Enterobacteriaceae), chemical quality (pH, total volatile basic nitrogen, trimethylamine, peroxide value, thiobarbituric acid, and free fatty acids), and raw fish sensory attributes were evaluated during 16 days of storage. The sensory attributes of golden gray mullet correlated well with the microbiological analyses (r = 0.92). Based on the overall raw acceptability sensory scores and the microbiological tests, the shelf life of the raw golden gray mullet was 10 days in ice and about 14 days in a refrigerator.  相似文献

Rapidly quenching embrittlement (RQE) sometimes appears in Fe73.5Cu1Nb3Si13.5B9 nanocrystalline soft magnetic alloy (FINEMET) during production process. As a re-sult of RQE the ductility of the as-quenched ribbon drops seriously. The mechanism of RQE has been elucidated in the recent works of current authors. It was believed that RQE is due to the structural relaxation but not related to a-Fe(Si) crystallization. In this paper, the high resolution TEM (HRTEM) method and image digital processing were applied to analyze the HRTEM images of two FINEMET rapidly quenched ribbons with different thicknesses in detail. In the thinner ductile sample, the ordering domains with the size of about 3nm are observed. In the thicker RQE sample, the metastable nanocrystalline domains with the size of 18nm are observed along with the structural relaxation. These domains seem to have Fe3B-like metastable phase struc-ture on nanometer scale. The result indicates that the local atomic ordering regions extend when RQE induced  相似文献

There is a paucity of data in respect of the nutritional quality of complementary foods for infants and young children aged between 6 and 12 months. The primary objective of this study was to examine nutritive values of such complementary infant food on the UK market in order to ascertain their suitability relative to dietary guidelines for the 6–9 months age group. Quantitative analyses were conducted on eight different products representing four popular brands (meat and vegetable based) currently on sale in the UK. Eight major mineral and trace elements, namely: calcium, copper, magnesium, iron, zinc, potassium, sodium and selenium were measured by ICP–OES and ICP–MS. The results of these studies were referenced to the Recommended Nutrient Intake (RNI) values for 6–9 months old children, and a menu of entire daily intake of minerals and trace elements was composed taking into consideration the nutrient and energy intake from milk consumption. Based on these comparisons, all the food samples studied in this work contained less essential minerals than expected from the RNI values except for potassium in meat and vegetable based recipes. These results suggest that commercial complementary infant foods on the UK market may not contain the minimum levels of minerals required for the labelling declaration of micronutrient content (Commission Directive 2006/125/EC). This provides opportunities and scope for product optimisation to improve their nutritive value.  相似文献

The structure, solubility and emulsifying properties of the deamidated barley glutelin were investigated. The SDS–PAGE, size exclusion chromatography combined with a laser photometer (SEC-LP) and Fourier transform infrared spectroscopy (FTIR) results revealed that glutelin underwent a rapid hydrolysis and unfolding even at the initial stage of deamidation, leading to remarkably improved glutelin solubility at both acidic and neutral pHs. The deamidated glutelin demonstrated a strong tendency to form soluble aggregates of very large molecular weight (106–107 g/mol) probably due to disulphide-crosslinking and hydrophobic interactions. These aggregates played a major role in stabilising the emulsions at a broad range of deamidation levels (2.2–5.6% to 43%). In this way, undesirable property changes which result from excessive deamidation can be avoided, leading to wider applications of deamidated barley glutelin as an emulsifying ingredient in food and non-food applications.  相似文献

This study was conducted to obtain annatto extracts with both high antioxidant capacity and colour potential using solvents of different polarities (water, ethanol/water, ethanol, ethanol/ethyl acetate and ethyl acetate). The highest levels of total phenolic compounds were found in the water, ethanol/water and ethanol extracts (0.5 mg GAE mL?1), and the highest level of bixin was found in the ethanol/ethyl acetate extract (5.2 mg mL?1), which was characterised as the reddest and the most vivid one (a* = 40.5, = 46.1, C* = 58.4). The ethanol/ethyl acetate extract also showed the highest antioxidant activity (4.7 μm TEAC mL?1) and the highest percentage of tryptophan protection against singlet oxygen (63.6%). On the other hand, ethyl acetate and ethanol/water were the least effective solvents for the extraction of phenolic compounds and bixin, respectively. According to the multivariate statistical analysis, ethanol/ethyl acetate and ethyl acetate were the most promising solvents to obtain annatto extracts with both antioxidant and colour properties.  相似文献

The antibacterial activity of a chitooligosaccharide mixture prepared by digestion of shrimp chitosan with cellulase at 50 degrees C for 14 h was evaluated. Sugars with 1 to 8 degrees of polymer (DP) were found in this chitooligosaccharide mixture, and the weight percentage of sugars with DP > or = 6 was 44.3%. Minimal lethal concentrations of this mixture against Aeromonas hydrophila, Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella Typhimurium, Shigella dysenteriae, Staphylococcus aureus, Vibrio cholerae, and Vibrio parahaemolyticus in nutrient broth were 5 to 29 ppm, which were much lower than those of the chitosan reactant (50 to 1,000 ppm). The antibacterial activity of this mixture in the sterilized milk against E. coli O157, L. monocytogenes, Salmonella Typhimurium, and S. aureus was much stronger at 4 degrees C than at 37 degrees C. When raw milk was supplemented with either 0.24% or 0.48% (wt/vol) of this oligosaccharide mixture and stored at 4 degrees C for 12 days, its mesophilic and psychrotrophic counts were reduced by at least 3 log cycles, and there was very little change in pH. In addition, this mixture retarded the growth of Salmonella species and caused quicker reduction of Staphylococcus species in raw milk. Accordingly, the shelf life of raw milk at 4 degrees C was extended by at least 4 days.  相似文献
