Reduced growth factor requirements and accelerated cell-cycle kinetics in adult human melanocytes transformed with SV40 large T antigen

Melanomas develop with high frequency in transgenic mice in which oncogenic sequences of the SV40 DNA tumor virus have been specifically targeted to melanocytes. To investigate the role of SV40 in melanomagenesis, cultured human melanocytes were transformed with a retroviral shuttle vector encoding the SV40 large T antigen and examined for changes in cell-cycle kinetics and growth-factor dependence. Colonies expressing the viral oncogene were morphologically indistinguishable from their non-T-antigen-transformed counterparts. Also like normal melanocytes, the infected cells remained anchorage dependent and non-tumorigenic in nude mice. However, T-antigen-positive cultures exhibited significantly accelerated population doubling times, increased saturation densities with highly confluent monolayers and a three- to fourfold extended life span. Most interestingly, cell-cycle analysis revealed a measurable shift from quiescent to cycling cells in T-antigen-expressing cultures and an acquired ability to progress more rapidly through G1. Moreover, T-antigen-positive melanocytes proliferated in the absence of PMA and required markedly reduced levels of exogenous bFGF. These studies indicate that the viral oncogen of simian virus 40 provides melanocytes with distinct growth advantages that may render these cells unusually susceptible to additional environmental challenges necessary for full expression of the malignant phenotype.     

ENTEROBACTER AGGLOMERANS—A NEW BACTERIAL CONTAMINANT ISOLATED FROM LAGER BEER BREWERIES

From a large number of bacterial strains isolated from all stages of the brewing process in four different beer breweries in South Africa, fifty-five bacterial strains were identified as Enterobacteriaceae by phenotypic analysis. All enterobacterial species previously reported in brewery samples, as well as eighteen strains of Enterobacter agglomerans were found. E. agglomerans strains were isolated from pitching yeast and fermenting wort samples. The occurrence of E. agglomerans in pitching yeast is significant and indicates that these bacteria survive the brewing process. E. agglomerans is regarded as a potential beer spoilage contaminant in lager beer breweries.     

Balancing the Fluency-Consistency Tradeoff in Collaborative Information Search with a Recommender Approach

Creative group work can be supported by collaborative search and annotation of Web resources. In this setting, it is important to help individuals both stay fluent in generating ideas of what to search next (i.e., maintain ideational fluency) and stay consistent in annotating resources (i.e., maintain organization). Based on a model of human memory, we hypothesize that sharing search results with other users, such as through bookmarks and social tags, prompts search processes in memory, which increase ideational fluency, but decrease the consistency of annotations, e.g., the reuse of tags for topically similar resources. To balance this tradeoff, we suggest the tag recommender SoMe, which is designed to simulate search of memory from user-specific tag-topic associations. An experimental field study (N = 18) in a workplace context finds evidence of the expected tradeoff and an advantage of SoMe over a conventional recommender in the collaborative setting. We conclude that sharing search results supports group creativity by increasing the ideational fluency, and that SoMe helps balancing the evidenced fluency-consistency tradeoff.
