Regulators of G protein signaling (RGS) proteins act as GTPase-activating proteins (GAPs) toward the alpha subunits of heterotrimeric, signal-transducing G proteins. RGS11 contains a G protein gamma subunit-like (GGL) domain between its Dishevelled/Egl-10/Pleckstrin and RGS domains. GGL domains are also found in RGS6, RGS7, RGS9, and the Caenorhabditis elegans protein EGL-10. Coexpression of RGS11 with different Gbeta subunits reveals specific interaction between RGS11 and Gbeta5. The expression of mRNA for RGS11 and Gbeta5 in human tissues overlaps. The Gbeta5/RGS11 heterodimer acts as a GAP on Galphao, apparently selectively. RGS proteins that contain GGL domains appear to act as GAPs for Galpha proteins and form complexes with specific Gbeta subunits, adding to the combinatorial complexity of G protein-mediated signaling pathways.  相似文献

Endoscopic treatment of chronic pancreatitis has drawn benefits from endoscopic procedures previously described for the main bile duct. Endotherapy is developing throughout the world. Cyst drainage procedures certainly represent the largest step forward in that non-surgical approach, whatever it is either direct (through the stomach or the duodenum) or indirect through the papilla in the duodenum. This procedure gives similar results to surgery with a lower morbidity. Pancreatic duct drainage associated with stone clearance is feasible and provides good results which have not yet been compared with those obtained thanks to surgery. Nonetheless, when the duct is not widely dilated it has been proven to be a satisfactory alternative to surgery. By contrast, chronic cholestasis does not appear to be a good indication of endotherapy.  相似文献

The solution secondary structure of the highly active Y55F/Y88F "Tyr-14-only" mutant of delta 5-3-ketosteroid isomerase complexed with 19-nortestosterone hemisuccinate has been shown to consist of three helices, a six-stranded mixed beta-sheet, and five turns. The steroid binds near the general acid, Tyr-14, on helix 1, near the general base, Asp-38, on the first strand of the beta-sheet, and on the hydrophobic face of the beta-sheet [Zhao, Q., Abeygunawardana, C., & Mildvan, A. S. (1997) Biochemistry 36, 3458-3472]. On this hydrophobic face, Asp-99 is the only polar residue. Free isomerase shows a deshielded exchangeable proton resonance at 13.1 ppm assigned to the N epsilon H of neutral His-100. Its fractionation factor (phi = 0.79) and slow exchange with solvent suggest it to be buried or involved in an H-bond. The binding of dihydroequilenin or estradiol to isomerase induces the appearance of two additional deshielded proton resonances, one at 18.2 ppm assigned to the gamma-carboxyl proton of Asp-99, and the other, at 11.6 ppm, assigned to the zeta-OH proton of Tyr-14. While mutation of Asp-99 to Ala results in the disappearance of only the resonance near 18 ppm [Wu, R. W., Ebrahemian, S., Zwrotny, M. E., Thornberg, L. D., Perez-Alverado, G. C., Brothers, P., Pollack, R. M., & Summers, M. F. (1997) Science 276, 415-418], both of these resonances disappear in mutants lacking Tyr-14, suggesting an H-bonded catalytic diad, Asp-99-COOH--Tyr14-OH--O-steroid enolate. The catalytic diad is further supported by NOEs from the beta 1 and beta 2 protons of Asp-99 to the epsilon protons of Tyr-14, and from the zeta-OH proton of Tyr-14 to the gamma-carboxyl proton of Asp-99, indicating close proximity of these two residues, and by other data from the literature. A strong, low-barrier H-bond between Asp-99 and Tyr-14 is indicated by the 6.2 ppm deshielding, low fractionation factor (phi = 0.34) and slow exchange of the resonance at 18.2 ppm. A normal H-bond between Tyr-14 and the steroid is indicated by the 1.8 ppm deshielding, fractionation factor of 0.97 and the slow exchange of the resonance at 11.6 ppm. It is suggested that the 10(4.7)-fold contribution of Tyr-14 to catalysis is made possible by strong H-bonding from Asp-99 in the catalytic diad which strengthens general acid catalysis by Tyr-14. It is also noted that highly deshielded proton resonance on enzymes between 15 and 20 ppm, assigned to low-barrier H-bonds, generally involve carboxyl groups.  相似文献

The preparation of the 11.7Li2O–39.7WO3–10.9CaO–37.7P2O5 glass (pgLWCP) was based on a one-step heat treatment of the 6Li2O–18WO3–43CaO–33P2O5 (gLWCP) glass followed by leaching of the β-Ca2P2O7 phase formed during the crystallization process. The porous structure was formed in the region formerly occupied by the β-Ca2P2O7 phase. The gLWCP undergoes devitrification through surface crystallization. This process occurs after a thermal treatment in lower temperature and in a shorter period of time than that required for the complete crystallization. After acid leaching treatment, we obtained a core-/shell-like structure with a very well-defined dense glass (gLWCP)/porous glass (pgLWCP) interface. The pgLWCP exhibits reversible coloration–decoloration reactions.  相似文献

This paper describes the highlights of presentations and discussions during the Third International BCI Meeting in a workshop that evaluated potential brain-computer interface (BCI) signals and currently available recording methods. It defined the main potential user populations and their needs, addressed the relative advantages and disadvantages of noninvasive and implanted (i.e., invasive) methodologies, considered ethical issues, and focused on the challenges involved in translating BCI systems from the laboratory to widespread clinical use. The workshop stressed the critical importance of developing useful applications that establish the practical value of BCI technology.  相似文献

Bipolar junction transistors (BJTs) have been at the core of linear electronics from its beginnings. Here, we suggest an Early‐based inspired geometric model of BJT devices and its application to derive models of related electronic circuits (more specifically a common‐emitter configuration). The approach involves using a beam of isolines converging at the Early voltage as the model of a BJT. The angles of the isolines are experimentally verified often to vary almost linearly with the base current. A numerical methodology is suggested for the Early voltage estimation and applied to 12 real‐world small signal NPN BJTs. Interesting results are obtained, including the identification of diverse Early voltage values for different BJT types and estimation of transfer functions.  相似文献

This study evaluates the quality of sausage obtained from the meat of Nellore cattle fed diets containing different levels (0, 5, 10, and 15 g per kg total DM) of lauric acid (C12:0) from palm kernel cake. A linear reduction (p ≤ 0.05) in lipid oxidation, as reflected by linear decreases in the lightness (L*), yellowness (b*), and saturation (C*) color parameters, is observed on days 7, 14, and 21 of maturation of sausage, and these decreases are accompanied by linear increases in the redness (a*) color and the linoleic (C18:2n–6) and linolenic (C18:3n–3) fatty acid contents. The inclusion of lauric acid in the diet induces linear reductions in the shear force and cooking loss and does not significantly affect various indices, including the centesimal composition, water activity, water holding capacity, composition of most fatty acids (FA), hypocholesterolemic‐to‐hypercholesterolemic FA ratio, atherogenicity, thrombogenicity, and desirable fatty acids. The use of dietary lauric acid from palm kernel cake is recommended at doses up to 15 g per kg because its presence reduces lipid oxidation and improves color parameters, softness, and linoleic and linolenic FA without affecting the FA composition of sausage from Nellore bull's meat. Practical applications : Sausages are products manufactured from chopped or ground meats packaged into animal casings. This product appeared in Brazil through the adaptation of traditional recipes brought by German and Italian immigrant families to weather conditions and the national palate. However, due to its production characteristics, sausage can become a product with lipid characteristics that are undesirable for human consumption. The inclusion of lauric acid (C12:0) in the animal diet might change the biohydrogenation process in the rumen, improve the quality of the deposited FA and reduce lipid oxidation in sausage manufactured from meat. The FA composition and physicochemical properties of sausage predict its acceptance by the consumer market. An optimal advantage would be achieved if these products can be used as not only preservatives but also functional ingredients with antioxidant properties and products for the treatment of metabolic syndrome and cardiovascular diseases through atherogenic action.  相似文献

The Munsell Color System and the Natural Color System are widely used but they have some limitations due to the manufacturing process and sampling choices. To estimate quantitatively these limitations we compared the colors of natural scenes with the colors represented by these systems under a wide range of illuminants. Spectral data from the two systems and from natural scenes were used in the analysis. It was found that a considerable portion of natural colors are not accounted by these systems, mainly colors with low lightness levels. Under D65 the Munsell Color System color volume corresponds to 72% of the Natural Color System color volume which in turn represents only 53% of the natural scenes color volume. If individual colors are considered, less than half are contained within these systems. To obtain a complete match to the natural colors contained by the color systems thresholds of 7 and 5 CIELAB units would be required for Munsell Color System and Natural Color System, respectively. Variations with the illuminant are generally modest showing that both system work similarly across different illuminations. Although these Color Systems have limitations in describing low lightness colors they perform quite well for medium to high levels of lightness.  相似文献

Fluorescent markers are critical for tracking the position and movement of molecules both in vivo and in vitro. Conventionally, synthetic dyes are non‐covalently added to polymers for fluorescent tracking, but often diffuse away. Here we demonstrate, for the first time, a facile method for the synthesis of fluorescent poly(lactic acid) nano‐/microfibres for biomedical applications using solution spin blowing. Pyrene‐end‐capped poly(l ‐lactide) (PLLA) derivatives were synthesised using the ring‐opening polymerisation of l ‐lactide and they were characterised using spectroscopic and thermal analyses. Submicrometre‐sized fluorescent fibres were produced from these PLLA derivatives using solution blow spinning techniques generating polymer blends and core–shell fibres. Such system could be further exploited to incorporate electrically conductive carbon allotropes via the pyrene aromaticity, producing fluorescent and electrically active fibres for in vitro monitoring and electrical stimulation. © 2018 Society of Chemical Industry  相似文献
