Differential, functional loading of the mandibular condyles has been suggested by several human morphologic studies and by animal strain experiments. To describe articular loading and the simultaneous forces on the dental arch, static bites on a three-dimensional finite element model of the human mandible were simulated. Five clenching tasks were modeled: in the intercuspal position; during left lateral group effort; during left lateral group effort with balancing contact; during incisal clenching; and during right molar clenching. The model's predictions confirmed that the human mandibular condyles are load-bearing, with greater force magnitudes being transmitted bilaterally during intercuspal and incisal clenching, as well as through the balancing-side articulation during unilateral biting. Differential condylar loading depended on the clenching task. Whereas higher forces were found on the lateral and lateroposterior regions of the condyles during intercuspal clenching, the model predicted higher loads on the medial condylar regions during incisal clenching. The inclusion of a balancing-side occlusal contact seemed to decrease the forces on the balancing-side condyle. Whereas the predicted occlusal reaction forces confirmed the lever action of the mandible, the simulated force gradients along the tooth row suggest a complex bending behavior of the jaw.  相似文献

The mapping of allelic loss on the short arm of chromosome 1 has been performed in non-small-cell lung cancer. We used a set of 11 microsatellite loci spanning 1p to examine the frequency of allelic imbalance in a panel of 58 tumours. Fifty-one of 58 (87.9%) cases have shown somatic allelic loss at one or more loci tested. The two shortest regions of the overlap (SRO) of the deletions have been identified: SRO 1 at 1p13.1 and SRO 2 at 1p32-pter. Allelic losses at these regions have been compared among adenocarcinoma and squamous cell carcinoma and no difference has been found. In contrast to SRO 1, deletions at SRO 2 significantly correlated with advanced stage of the disease as well as post-operative metastasizing and relapse. These data may suggest that SRO 1 and SRO 2 can harbour tumour-supressor genes (TSGs) involved in different stages of NSCLC development. SRO 2 is still quite large and its refined mapping should help attempts to clone and identify the putative TSG(s). Microsatellite instability (replication errors) affecting only 6 (10.3%) of 58 tumour samples is an infrequent genetic alteration at the loci tested.  相似文献

However, there have been a significant number of controlled studies, allowing definitive conclusions to be drawn. BACKGROUND: The transition of a fatty streak into an atherosclerotic plaque is characterized by the appearance of focal and diffuse regions of cell death. We have investigated the distribution of apoptotic cell death and apoptosis-related proteins in early and advanced atherosclerotic lesions. METHODS AND RESULTS: Human atherosclerotic plaques were studied by whole-mount carotid endarterectomy specimens (n=18). This approach allowed comparison of adaptive intimal thickenings, fatty streaks, and advanced atherosclerotic plaques of the same patient. The fatty streaks differed from adaptive intimal thickenings by the presence of BAX (P<0.01), a proapoptotic protein of the BCL-2 family. Both regions were composed mainly of smooth muscle cells (SMCs), and macrophage infiltration was low and not different. Apoptosis, as detected by DNA in situ end labeling (terminal deoxynucleotidyl transferase end labeling [TUNEL] and in situ nick translation) was not present in these regions. Apoptosis of SMCs and macrophages, however, was present in advanced atherosclerotic plaques that were present mainly in the carotid sinus. A dense infiltration of macrophages (5.8+/-3% surface area) was present in these advanced atherosclerotic plaques. Cytoplasmic remnants of apoptotic SMCs, enclosed by a cage of thickened basal lamina, were TUNEL negative and remained present in the plaques as matrix vesicles. CONCLUSIONS: We conclude that SMCs within human fatty streaks express BAX, which increases the susceptibility of these cells to undergo apoptosis. The localization of these susceptible SMCs in the deep layer of the fatty streaks could be important in our understanding of the transition of fatty streaks into atherosclerotic plaques, which are characterized by regions of cell death. Matrix vesicles are BAX-immunoreactive cytoplasmic remnants of fragmented SMCs that can calcify and may be considered the graves of SMCs that have died in the plaques.  相似文献

Hydrophobins are small (length, about 100 +/- 25 amino acids), cysteine-rich, hydrophobic proteins that are present in large amounts in fungal cell walls, where they form part of the outermost layer (rodlet layer); sometimes, they can also be secreted into the medium. Different hydrophobins are associated with different developmental stages of a fungus, and their biological functions include protection of the hyphae against desiccation and attack by either bacterial or fungal parasites, hyphal adherence, and the lowering of surface tension of the culture medium to permit aerial growth of the hyphae. We identified and isolated a hydrophobin (fruit body hydrophobin 1 [Fbh1]) present in fruit bodies but absent in both monokaryotic and dikaryotic mycelia of the edible mushroom Pleurotus ostreatus. In order to study the temporal and spatial expression of the fbh1 gene, we determined the N-terminal amino acid sequence of Fbh1. We also synthesized and cloned the double-stranded cDNA corresponding to the full-length mRNA of Fbh1 to use it as a probe in both Northern blot and in situ hybridization experiments. Fbh1 mRNA is detectable in specific parts of the fruit body, and it is absent in other developmental stages.  相似文献

Testicular germ cell neoplasms affect young men in the prime of life. Although the overwhelming majority are malignant, they are curable. In addition to the stage of the disease and the presence of serum markers, there are important pathological changes that have clinical significance. These include (1) the cell type, (2) the amount of the component, and (3) the presence or absence of vascular invasion. Pure embryonal carcinoma or embryonal carcinoma in excess of 80% in a mixed tumor and vascular/lymphatic invasion are high-risk factors as they are predictors of relapse. These factors should be recognized by the pathologist and should be taken into account by the oncologist when selecting the management of a patient with a germ cell tumor of the testis.  相似文献
