Human herpesvirus 8 (HHV-8) DNA sequences were examined in peripheral blood mononuclear cell (PBMC) DNA samples of 56 children, 15 healthy adults, and 10 renal transplant patients by the polymerase chain reaction (PCR). The PCR amplification was carried out using the published KS330(233) primer pairs to amplify HHV-8 DNA sequences. The PCR-amplified products were confirmed by Southern blot hybridization with radiolabeled 233 bp HHV-8 DNA fragment, which was cloned and sequenced from the PCR-amplified product of Kaposi's sarcoma tissue. Six PCR-amplified product of four children and two renal transplant patients were cloned and sequenced. HHV-8 DNA sequences were detected in 36 of 56 (64%) normal children, in 12 of 15 (80%) healthy adults, and in all 10 renal transplant patients by Southern blot hybridization of PCR-amplified products. Six PCR-amplified products were confirmed by sequencing. These results suggest that HHV-8 infection is prevalent in the Japanese population with infection occurring in early childhood.  相似文献

One of the challenges in understanding ciliary and flagellar motility is determining the mechanisms that locally regulate dynein-driven microtubule sliding. Our recent studies demonstrated that microtubule sliding, in Chlamydomonas flagella, is regulated by phosphorylation. However, the regulatory proteins remain unknown. Here we identify the 138-kD intermediate chain of inner arm dynein I1 as the critical phosphoprotein required for regulation of motility. This conclusion is founded on the results of three different experimental approaches. First, genetic analysis and functional assays revealed that regulation of microtubule sliding, by phosphorylation, requires inner arm dynein I1. Second, in vitro phosphorylation indicated the 138-kD intermediate chain of I1 is the only phosphorylated subunit. Third, in vitro reconstitution demonstrated that phosphorylation and dephosphorylation of the 138-kD intermediate chain inhibits and restores wild-type microtubule sliding, respectively. We conclude that change in phosphorylation of the 138-kD intermediate chain of I1 regulates dynein-driven microtubule sliding. Moreover, based on these and other data, we predict that regulation of I1 activity is involved in modulation of flagellar waveform.  相似文献

PURPOSE: Anesthetics can alter the biodistribution profile of drugs and, consequently, the regional pharmacokinetics of antineoplastic drugs at the tumor site. The effect of coadministered anesthetics on the biodistribution profile of carboplatin was studied in rats. METHODS: Female Wistar rats were used to compare the effects of ketamine/xylazine, thiopental and pentobarbital on the biodistribution of 30 mg/kg radiolabelled 195mPt-carboplatin administered intravenously, with conscious rats as the control group. Blood and urine samples were collected between 5 and 120 min. RESULTS: The percentage values of the injected dose of platinum per ml (%ID/ml) in plasma at the final time-point were respectively, 0.557%, 0.156%, 0.115% and 0.086%, in pentobarbital-, ketamine/xylazine- and thiopental-injected rats, and in conscious animals. Following the same sequence of groups, the %ID/ml values of platinum in the cumulative urine were 0.001%, 0.619%, 0.184% and 0.118%, respectively. Urine output varied from very little in the pentobarbital group, to several milliliters in the other groups. CONCLUSIONS: There was an increase of almost 100-fold in total platinum uptake in the kidneys, cerebrum and cerebellum of rats receiving pentobarbital over the uptake in the control rats, whereas the biodistribution profile of the thiopental group had the least variance. These results demonstrate the importance of anesthetic selection in animal pharmacokinetic studies, as it influences the biodistribution and pharmacokinetic profile of the drug being studied.  相似文献

The prevalence and severity of gingival overgrowth in organ-transplant patients medicated with cyclosporin are greater in patients concomitantly medicated with nifedipine; however, no relationship between the gingival overgrowth and any of the nifedipine pharmacological variables has been demonstrated. The study examined the effect of five nifedipine pharmacological variables (nifedipine dosage, plasma concentration and gingival crevicular fluid concentration, M1 metabolite plasma concentration and the nifedipine: M1 ratio). The effect of the nifedipine variables on the gingival overgrowth score were examined using univariate and multivariate regression analysis. Adjustment for the effect of other risk factors was made by adding the distribution of each of the nifedipine variables in turn to a stepwise regression model containing previously identified risk factors for this condition. Despite the high levels of nifedipine sequestered in the GCF, only the plasma concentration of nifedipine was identified as a risk factor for the severity of gingival overgrowth in these patients (P = 0.01) once adjusted for other known risk factors (R2 for the model = 55%).  相似文献

In the hemopoietic system, interactions between stem cells and components of the bone marrow microenvironment play a pivotal role in blood cell proliferation and differentiation. Among the adhesion molecules, the integrins of the beta 1-subfamily are known to direct cell-cell and cell-matrix interactions and evidence has been provided that CD34-positive stem cells bind either to the bone marrow stroma or to the extracellular matrix proteins through the beta 1-integrins. It seems that changes in their expression pattern or signalling function are likely to reflect disturbances at the hemopoietic bone marrow microenvironmental level. Any alteration of their biological functions makes them attractive candidates for playing decisive roles in the leukemic processes. In this view, beta 1-integrins have been recognized to mediate those cellular interactions and migrations that are important in the biology of leukemia. In this paper we review some aspects of the role played by beta 1-integrins, especially VLA-4 and VLA-5, in adult acute lymphoblastic leukemia in relation with the expression rate of the stem cell antigen CD34.  相似文献

Exposure to fungal aerosols is of concern in indoor environments. However, sampling limitations have previously made it difficult to assess exposures accurately, especially long-term exposures. A prototype personal aerosol sampler, based on cyclone principles and using a 1.5 ml microcentrifuge tube as a particle collection receptacle has been designed and fabricated. Collection efficiency for aerosol particles in the size range of fungal spores has been evaluated for different types of microcentrifuge tubes, together with the effect of a polyethylene glycol coating on the inside of the tube and the effect of adding water to the tube. Monodisperse, fluorescently tagged polymer microspheres with median diameters of 0.5, 1, 2, 3, 6, 11, and 16 μm were used to evaluate sampler performance with particle diameter. The microcentrifuge-tube sampler was tested at flow rates of 2 and 4 liters per minute (l/min). Experimental results indicate that the microcentrifuge-tube sampler has an aspiration efficiency of 100% in calm air for particles up to 16 μm. At 4 l/min, the microcentrifuge-tube sampler is able to collect nearly 100% of particles greater than 3 μm and > 90% of particles between 2.5 and 3 μm. The 50% cutoff size is 1.5 μm. The performance of the sampler did not vary with the different brands of tubes tested or with the presence or absence of a coating on the tube surface. Furthermore, the addition of water to the tube resulted in a slight increase in collection efficiency. A sampling time of 5 h was feasible at 45–50% relative humidity before evaporation led to significant water loss.

The cutoff size of 1.5 μm is comparable to many commercially available bioaerosol samplers. Besides being easy to use, simple to fabricate, and inexpensive, this novel sampler has several advantages over conventional samplers: long-term samples are possible (the limitation of impaction methods); there is no sample transfer loss since the transfer step has been eliminated (the limitation of filter cassettes); laboratory analyses are not dependent solely upon a single analysis method (the limitation of impaction methods), and there is no sampler adherence loss (the limitation of trying to wash microorganisms from filters). In addition, use of the sampler would be applicable in a variety of occupational settings from low bioaerosol concentrations (i.e., indoor environments) to high bioaerosol concentrations (i.e., agricultural setting) by varying sampling time periods and using sensitive analytical methods.  相似文献

The purpose of this study was to numerically simulate the performance of an aerosol sampler with a curved, blunt, multi-orificed inlet in order to understand the sampling characteristics of the first prototype of the button personal inhalable aerosol sampler ("button sampler"). Because the button sampler inlet design is too complicated to apply a three-dimensional model, an axisymmetric two-dimensional model was created to be similar in geometry and to simulate the major features of the airflow through the sampler when facing the wind. Particle trajectories were calculated in a variety of wind velocities and were categorized into 5 groups based on their interactions with the curved surface of the sampling plane. Empirical sampling efficiencies of the button sampler for 3 particle sizes were used to adjust the calculated sampling efficiencies in an attempt to improve the accuracy of the two-dimensional axisymmetric model in accounting for interactions between particles and the surface of the inlet of the button sampler. Sampling efficiencies for other particle sizes were then predicted. The results showed that sampling efficiency decreased with increasing particle size up to approximately 40 w m and then remained virtually unchanged at about 35% up to 100 w m. Although the efficiencies were lower than the American Conference of Governmental Industrial Hygienists' (ACGIH) inhalability curve for larger particles, the pattern of the predicted sampling efficiency was quite similar to the ACGIH inhalability curve. Sampling efficiencies for liquid aerosol particles larger than 15 w m were predicted to be noticeably lower than those for solid particles. The results also showed that the multi-orificed curved surface played an important role in establishing a pressure drop with desired flow alignment inside the sampler, thus greatly reducing the wind effect and significantly improving the uniformity of particle deposition on the filter. The less uniform deposition found at high wind velocity can be improved by increasing the sampling flow rate.  相似文献
