Herbal remedies are increasing in popularity as treatments for metabolic conditions such as obesity and Type 2 Diabetes. One potential therapeutic option is fenugreek seeds (Trigonella foenum-graecum), which have been used for treating high cholesterol and Type 2 diabetes. A proposed mechanism for these benefits is through alterations in the microbiome, which impact mammalian host metabolic function. This study used untargeted metabolomics to investigate the fenugreek-induced alterations in the intestinal, liver, and serum profiles of mice fed either a 60% high-fat or low-fat control diet each with or without fenugreek supplementation (2% w/w) for 14 weeks. Metagenomic analyses of intestinal contents found significant alterations in the relative composition of the gut microbiome resulting from fenugreek supplementation. Specifically, Verrucomicrobia, a phylum containing beneficial bacteria which are correlated with health benefits, increased in relative abundance with fenugreek. Metabolomics partial least squares discriminant analysis revealed substantial fenugreek-induced changes in the large intestines. However, it was observed that while the magnitude of changes was less, significant modifications were present in the liver tissues resulting from fenugreek supplementation. Further analyses revealed metabolic processes affected by fenugreek and showed broad ranging impacts in multiple pathways, including carnitine biosynthesis, cholesterol and bile acid metabolism, and arginine biosynthesis. Wood chips were evaluated for their ability to attenuate heavy metals in roadway runoff. Column experiments with controlled synthetic runoff composition and flow rate were used to assess effects of flow rate (intercepted sheetflow from a 3-m wide roadway section), runoff salt concentration, wood exposure to alternating wetting and drying cycles, wood aging, competition among dissolved heavy metals, and removal of particle-associated heavy metals. Overall, wood chips damped the “pulse” of copper in the synthetic runoff such that the effluent was characterized by lower concentrations (3–25% of input) over longer periods of time, but with little retention of the total copper mass. The most effective treatment was wood chips aged up to 9 months. Increased aging and chip water content reduced effluent concentrations, relative to no treatment. Flow rate had no effect on effluent concentrations. The presence of salt (>2?mS/cm) or dissolved lead (500?μg/L) in the runoff caused greater copper effluent concentrations than the no treatment case. Removal of suspended particles (and associated contaminants) was greater than 85% with an estimated capacity of 0.16?g/gwood. Field evaluation with concentrated flow to a gutter containing a wood chip treatment showed little effect on total or dissolved copper and zinc runoff concentrations and indicated that wood chips may be a source of contaminants in subsequent storm events. Applications of wood chips to treat roadway runoff would not provide a significant decrease in total maximum daily load contributions (e.g., kg/d); however, there may be some scenarios for which wood chip treatments to decrease peak storm water concentrations of dissolved heavy metals in sheetflow runoff is desirable. 相似文献

Cutaneous squamous cell carcinoma (cSCC) accounts for 25% of cutaneous malignancies diagnosed in Caucasian populations. Surgical removal in combination with radiation and chemotherapy are effective treatments for cSCC. Nevertheless, the aggressive metastatic forms of cSCC still have a relatively poor patient outcome. Studies have linked actin cytoskeletal dynamics and the Wnt/β-catenin signaling pathway as important modulators of cSCC pathogenesis. Previous studies have also shown that the actin-remodeling protein Flightless (Flii) is a negative regulator of cSCC. The aim of this study was to investigate if the functional effects of Flii on cSCC involve the Wnt/β-catenin signaling pathway. Flii knockdown was performed using siRNA in a human late stage aggressive metastatic cSCC cell line (MET-1) alongside analysis of Flii genetic murine models of 3-methylcholanthrene induced cSCC. Flii was increased in a MET-1 cSCC cell line and reducing Flii expression led to fewer PCNA positive cells and a concomitant reduction in cellular proliferation and symmetrical division. Knockdown of Flii led to decreased β-catenin and a decrease in the expression of the downstream effector of β-catenin signaling protein SOX9. 3-Methylcholanthrene (MCA)-induced cSCC in Flii overexpressing mice showed increased markers of cancer metastasis including talin and keratin-14 and a significant increase in SOX9 alongside a reduction in Flii associated protein (Flap-1). Taken together, this study demonstrates a role for Flii in regulating proteins involved in cSCC proliferation and tumor progression and suggests a potential role for Flii in aggressive metastatic cSCC. 相似文献

Little work has been done to study the fundamental material behaviors and failure mechanisms of cement-based materials including ordinary Portland cement concrete and ultra-high performance concretes (UHPCs) under high strain impact and penetration loads at lower length scales. These high strain rate loadings have many possible effects on UHPCs at the microscale and nanoscale, including alterations in the hydration state and bonding present in phases such as calcium silicate hydrate, in addition to fracture and debonding. In this work, the possible chemical and physical changes in UHPCs subjected to high strain rate impact and penetration loads were investigated using a novel technique wherein nanoindentation measurements were spatially correlated with images using scanning electron microscopy and chemical composition using energy dispersive x-ray microanalysis. Results indicate that impact degrades both the elastic modulus and indentation hardness of UHPCs, and in particular hydrated phases, with damage likely occurring due to microfracturing and debonding. 相似文献

Summary A spirobislactone, 3,3-spirobiphthalide (SBP), has been copolymerized with the diglycidyl ether of bisphenol A (DGEBA), and the properties of the products have been evaluated and related to the proportion (1–15%) of SBP monomer used. Polymers were characterized by infrared spectral analysis,13C NMR, thermal analysis, and mechanical testing. Polymers containing SBP have lower glass transition temperatures and higher decomposition temperatures than DGEBA polymers cured with phthalic anhydride (PA). Materials with higher SBP content have higher Talpha values. Mechanical properties of the SBP copolymers are similar to those of DGEBA-PA polymers. 相似文献

In this study the effects of high temperature and moisture on the impact damage resistance and mechanical strength of Nextel 610/alumina silicate ceramic matrix composites were experimentally evaluated. Composite laminates were exposed to either a 1050°C isothermal furnace-based environment for 30 consecutive days at 6 h a day, or 95% relative humidity environment for 13 consecutive days at 67°C. Low velocity impact, tensile and short beam strength tests were performed on both ambient and environmentally conditioned laminates and damage was characterized using a combination of non-destructive and destructive techniques. High temperature and humidity environmental exposure adversely affected the impact resistance of the composite laminates. For all the environments, planar internal damage area was greater than the back side dent area, which in turn was greater than the impactor side dent area. Evidence of environmental embrittlement through a stiffer tensile response was noted for the high temperature exposed laminates while the short beam strength tests showed greater propensity for interlaminar shear failure in the moisture exposed laminates. Destructive evaluations exposed larger, more pronounced delaminations in the environmentally conditioned laminates in comparison to the ambient ones. External damage metrics of the impactor side dent depth and area directly influenced the post-impact tensile strength of the laminates while no such trend between internal damage area and residual strength could be ascertained. 相似文献

Aging is inevitable and it is one of the major contributors to cognitive decline. However, the mechanisms underlying age-related cognitive decline are still the object of extensive research. At the biological level, it is unknown how the aging brain is subjected to progressive oxidative stress and neuroinflammation which determine, among others, mitochondrial dysfunction. The link between mitochondrial dysfunction and cognitive impairment is becoming ever more clear by the presence of significant neurological disturbances in human mitochondrial diseases. Possibly, the most important lifestyle factor determining mitochondrial functioning is nutrition. One of the promising avenues for biomass processing is the use of water as a reaction medium for wet or aquatic biomass. This review focuses on the hydrothermal catalytic production of fuels and chemicals from aquatic biomass. Two different regimes for conversion of aquatic biomass in hydrothermal conditions are discussed in detail. The first is hydrothermal liquefaction, and the second is hydrothermal gasification. The goals of these processes are to produce liquid‐fuel‐range hydrocarbons and methane or hydrogen, respectively. The catalytic upgrading of biocrude resulting from noncatalytic liquefaction and the stability and degradation of catalysts in high temperature water are also discussed. The review concludes with a brief discussion of the outlook for and opportunities within the field of hydrothermal catalytic valorization of biomass. Poly(vinyl toluene) (PVT) overdoped with 2,5-diphenyloxazole and using 1,4-bis(5-phenyloxazol-2-yl)benzene as a fluorescent secondary dopant can be used to detect and differentiate neutron and gamma radiation via scintillation. The low cost of PVT makes these plastic scintillators attractive for both portable and larger sized first line detection of special nuclear materials. Current fabrication methods rely on thermally initiated radical polymerization that generally requires an approximately 5-day heating process in order to produce high quality scintillators. In this work, we report a proof-of-concept photopolymerization process to prepare plastic scintillators up to 20 g in size in 1 day. These plastic scintillators were comparable to standard thermally polymerized samples in terms of their physical properties and response to various radiation sources. © 2018 Wiley Periodicals, Inc.
