Reductionist studies of adipose tissue biology require reliable in vitro adipocyte culturing models. Current protocols for adipogenesis induction in stromal vascular fraction-derived preadipocytes require extended culturing periods and have low adipogenic rates. We compared the adipogenic efficiency of a 7-d co-culture model of visceral (VIS) and subcutaneous (SC) stromal vascular fraction-derived preadipocytes with mature adipocytes with a 14-d standard adipocyte differentiation protocol. We obtained preadipocytes and mature adipocytes from SC and VIS adipose tissue of nonlactating, nongestating Holstein cows (n = 6). Adipogenesis induction was performed using a standard protocol for 7 (SD7; control) or 14 d (SD14), and a co-culture model for 7 d (CC7). Culture conditions, including medium composition, were the same for all treatments. For CC7, 900 primary adipocytes/cm2 were placed in 0.4-μm transwell inserts and co-cultured with preadipocytes for adipogenesis induction. Both CC7 and SD14 similarly stimulated gene expression of adipogenic genes such as ADIPOQ, CEBPA, and CEBPB in VIS and SC. The CC7 increased triacylglycerol accumulation compared with SD14 and SD7. CC7 augmented triacylglycerol accumulation by 40- and 16-fold in SC and VIS compared with 22- and 4-fold increment in SD14, respectively. Lipolytic responses to 2-h β-adrenergic stimulation with 1 µM isoproterenol were higher in CC7 and SD14 than SD7 in SC; CC7 increased glycerol release compared with SD7 in VIS but SD7 and SD14 had similar responses. Overall, CC7 was more efficient in inducing adipogenesis in preadipocytes from VIS and SC than SD14. Furthermore, CC7 stimulated similar lipolysis and lipogenic responses than SD14 but in a shorter time. The adipogenic approach of co-culturing preadipocytes with mature adipocytes will improve the use of reductionist models to study adipocyte physiology in dairy cows and the assessment of pharmacological or nutritional interventions for enhancing dairy cow health and production. 相似文献

Aluminum oxide films can provide excellent surface passivation on both p‐type and n‐type surfaces of silicon wafers and solar cells. Even though radio frequency magnetron sputtering is capable of depositing aluminum oxide with concentrations of negative charges comparable to some of the other deposition methods, the surface passivation has not been as good. In this paper, we compare the composition and bonding of aluminum oxide deposited by thermal atomic layer deposition and sputtering, and find that the interfacial silicon oxide layer and hydrogen concentration can explain the differences in the surface passivation. The cross-linked poly-2-isopropyl-2-oxazolines (PiPrOx) differing in cross-linker content have been synthesized and characterized by light scattering, chromatography, and rheometry. The dramatic influence of the cross-linker content on solubility and uni- or bimodality was observed. For the first time, the interpolymer complex of cross-linked PiPrOx with polyethylene oxide is manufactured in fibrous form by the electrospinning method. The role of intermolecular hydrogen bonds in the structure and properties of electrospun fibers (EF) was studied by infrared spectroscopy, differential scanning calorimetry, and scanning electron microscopy. It was established that these interactions allow to obtain a uniform fibrous membrane. At the same time, hydrogen bonding is responsible for the self-organization of PiPrOx in water. 相似文献

Fiber drawing enables scalable fabrication of multifunctional flexible fibers that integrate electrical, optical, and microfluidic modalities to record and modulate neural activity. Constraints on thermomechanical properties of materials, however, have prevented integrated drawing of metal electrodes with low-loss polymer waveguides for concurrent electrical recording and optical neuromodulation. Here, two fabrication approaches are introduced: 1) an iterative thermal drawing with a soft, low melting temperature (Tm) metal indium, and 2) a metal convergence drawing with traditionally non-drawable high Tm metal tungsten. Both approaches deliver multifunctional flexible neural interfaces with low-impedance metallic electrodes and low-loss waveguides, capable of recording optically-evoked and spontaneous neural activity in mice over several weeks. These fibers are coupled with a light-weight mechanical microdrive (1 g) that enables depth-specific interrogation of neural circuits in mice following chronic implantation. Finally, the compatibility of these fibers with magnetic resonance imaging is demonstrated and they are applied to visualize the delivery of chemical payloads through the integrated channels in real time. ABSTRACT People around the world spend hours of their daily lives using smartphones; therefore, it is important to conduct cross-cultural research on the effects of smartphone use on health and well-being as culture influences values, motivations and communication patterns. The purpose of this study was to explore 5 popular uses of the smartphone – messaging, browsing the Internet, posting social content, reading social content, and playing games – how they relate to anxiety and depression scores, and how they vary depending on the country of the participants: Spain, the United States, and Colombia. In all three countries the ranking of most popular uses was the same: (1) Messaging, (2) Reading social content, and (3) Browsing the Internet. In the USA, game playing contributed to anxiety scores whereas reading social content was a protective factor; regarding depression scores, text messaging was a contributing factor. In Spain, browsing the Internet contributed to anxiety scores; regarding depression scores, messaging was a contributing factor and posting social content was a protective factor. In Colombia, no specific use influenced anxiety scores; regarding depression scores, only game playing was a protective factor. The effects of a circumferential feeding groove on the dynamic force response of a squeeze film damper are determined experimentally. Open end and sealed damper configurations are tested for different groove depths, Journal orbit radii, whirl frequency and fluid viscosity conditions. Significant levels of dynamic pressures are measured at the groove in dampers with groove depth-to-clearance ratios equal to 5 and 10. These pressures affect considerably the dynamic forces of the dampers tested. Large tangential (damping) forces are produced at the circumferential groove which contribute significantly to the damping characteristics of the test articles. For uncavitated lubricant conditions, radial forces of substantial magnitude are determined at the groove and at the thin film land where the squeeze film Reynolds numbers are typically less than 1. 相似文献

Recognizing common reporting patterns of aquatic invasive Zebra mussels (Dreissena polymorpha, ZM) and Eurasian watermilfoil (Myriophyllum spicatum, EWM) helps to better understand invasions. We hypothesize that confirmed invasions may be confounded by human population density, leading to overrepresentation of invasions in highly populated areas and underrepresentation in less populated areas. Here we recognize dispersal patterns of confirmed ZM and EWM invasions in Minnesota, USA, using spatial clustering and directionality tests, while adjusting for human density. By 2015, 125 (0.68%) and 304 (1.65%) of 18,411 Minnesota waterbodies were reported to have ZM and EWM, respectively. A multivariate multinomial model of the scan test was used to identify clustering of invasions. The resulting 23 clusters included 13 with either or both ZM and EWM, and most clusters (11/13) occurred in areas with >10 people per square kilometer. Whereas, among the 10 clusters without invasion, nine were from less populated areas. The standard deviation ellipse and the spatiotemporal directionality tests indicated a northwestern trend of invasions, which is in the same direction as the I-94 interstate highway connecting urban centers. Results suggested that confirmed ZM and EWM invasions are potentially confounded by human densities, which is explained by varying human impact on either or both dispersal and reporting of invasions. Considering this impact of human density, we suggest that a combination of passive and targeted surveillance, where the magnitude of efforts are stratified by the human densities, may provide insight into the true invasion status and its progression in the Great Lakes region. 相似文献
