This paper gives a brief overview of microstructure and rheology that have been observed for a range of carbon nanotube (CNT) suspensions. In general, untreated CNT suspensions show a much higher level of observable optical microstructure reflecting their preference to aggregate; they also show higher levels of viscoelasticity over treated CNT suspensions. An unexpected Helical Band texture for untreated CNTs is reported together with a series of parallel plate optical observations showing a broad spectrum of behaviour for different shear conditions. Both steady shear and linear viscoelastic data are presented for treated and untreated systems.  相似文献

The authors introduce the construct of I-sharing--the belief that one shares an identical subjective experience with another person--and the role it plays in liking. In Studies 1-3, participants indicated their liking for an objectively similar and an objectively dissimilar person, one of whom I-shared with them and the other of whom did not. Participants preferred the objectively similar person but only when that person I-shared with them. Studies 4 and 5 highlight the role that feelings of existential isolation and the need for closeness play in people's attraction to I-sharers. In Study 4, people with high needs for interpersonal closeness responded to I-sharers and non-I-sharers with great intensity. In Study 5, priming participants with feelings of existential isolation increased their liking for I-sharers over objectively similar others. The results highlight the importance of shared subjective experience and have implications for interpersonal and intergroup processes. In this work, we introduce microscale isoelectric fractionation (μIF) for isolation and enrichment of molecular species at any desired location in a microfluidic chip. Narrow pH-specific polyacrylamide membranes are photopatterned in situ for customizable device fabrication; multiple membranes of precise pH are easily incorporated throughout existing channel layouts. Samples are electrophoretically driven across the membranes such that charged species, for example, proteins and peptides, are rapidly discretized into fractions based on their isoelectric points (pI) without the use of carrier ampholytes. This format makes fractions easy to compartmentalize and access for integrated preparative or analytical operations on-chip. We present and discuss the key design considerations and trade-offs associated with proper system operation and optimal run conditions. Efficient and reproducible fractionation of model fluorescent pI markers and proteins is achieved using single membrane fractionators at pH 6.5 and 5.3 from both buffer and Escherichia coli cell lysate sample conditions. Effective fractionation is also shown using a serial 3-membrane fractionator tailored for isolating analytes-of-interest from high abundance components of serum. We further demonstrate that proteins focused in pH specific bins can be rapidly and efficiently transferred to another location in the same chip without unwanted dilution or dispersive effects. μIF provides a rapid and versatile option for integrated sample prep or multidimensional analysis, and addresses the glaring proteomic need to isolate trace analytes from high-abundance species in minute volumes of complex samples.  相似文献

With increasing global health threats has come an urgent need to rapidly develop and deploy safe and effective therapies. A common practice to fast track clinical adoption of compounds for new indications is to repurpose already approved therapeutics; however, many compounds considered safe to a specific application or population may elicit undesirable side effects when the dosage, usage directives, and/or clinical context are changed. For example, progenitor and developing cells may have different susceptibilities than mature dormant cells, which may yet be different than mature active cells. Thus, in vitro test systems should reflect the cellular context of the native cell: developing, nascent, or functionally active. To that end, we have developed high-throughput, two- and three-dimensional human induced pluripotent stem cell (hiPSC)-derived neural screening platforms that reflect different neurodevelopmental stages. As a proof of concept, we implemented this in vitro human system to swiftly identify the potential neurotoxicity profiles of 29 therapeutic compounds that could be repurposed as anti-virals. Interestingly, many compounds displayed high toxicity on early-stage neural tissues but not on later stages. Compounds with the safest overall viability profiles were further evaluated for functional assessment in a high-throughput calcium flux assay. Of the 29 drugs tested, only four did not modulate or have other potentially toxic effects on the developing or mature neurospheroids across all the tested dosages. These results highlight the importance of employing human neural cultures at different stages of development to fully understand the neurotoxicity profile of potential therapeutics across normal ontogeny.  相似文献

The potential of integration of functions in microfluidic chips is demonstrated by implementing on-chip preconcentration of proteins prior to on-chip protein sizing by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Two polymeric elements-a thin (approximately 50 microm) size exclusion membrane for preconcentration and a longer (approximately cm) porous monolith for protein sizing-were fabricated in situ using photopolymerization. Contiguous placement of the two polymeric elements in the channels of a microchip enabled simple and zero dead volume integration of the preconcentration with SDS-PAGE. The size exclusion membrane was polymerized in the injection channel using a shaped laser beam, and the sizing monolith was cast by photolithography using a mask and UV lamp. Proteins injected electrophoretically were trapped on the upstream side of the size exclusion membrane (MW cutoff approximately 10 kDa) and eluted off the membrane by reversing the electric field. Subsequently, the concentrated proteins were separated in a cross-linked polyacrylamide monolith that was patterned contiguous to the size exclusion membrane. The extent of protein preconcentration is easily tuned by varying the voltage during injection or by controlling the sample volume loaded. Electric fields applied across the nanoporous membrane resulted in a concentration polarization effect evidenced by decreasing current over time and irreproducible migration of proteins during sizing. To minimize the concentration polarization effect, sieving gels were polymerized only on the separation side of the membrane, and an alternate electrical current path was employed, bypassing the membrane, for most of the elution and separation steps. Electrophoretically sweeping a fixed sample volume against the membrane yields preconcentration factors that are independent of protein mobility. The volume sweeping method also avoids biased protein loading from concentration polarization and sample matrix variations. Mobilities of the concentrated proteins were log-linear with respect to molecular weight, demonstrating the suitability of this approach for protein sizing. Proteins were concentrated rapidly (<5 min) over 1000-fold followed by high-resolution separation in the sieving monolith. Proteins with concentrations as low as 50 fM were detectable with 30 min of preconcentration time. The integrated preconcentration-sizing approach facilitates analysis of low-abundant proteins that cannot be otherwise detected. Moreover, the integrated preconcentration-analysis approach employing in situ formation of photopatterned polymeric elements provides a generic, inexpensive, and versatile method to integrate functions at chip level and can be extended to lowering of detection limits for other applications such as DNA analysis and clinical diagnostics.  相似文献

We related total PCBs (t-PCBs) in white perch (Morone americana), an abundant estuarine resident that supports a valuable recreational and commercial fishery in the mid-Atlantic region, to the amount and spatial arrangement of developed land in watersheds that discharge into 14 subestuaries of Chesapeake Bay. We considered the intensity of development in watersheds using four developed land-use measures (% impervious surface, % total developed land, % high-intensity residential + commercial [%high-res/comm], and % commercial) to represent potential source areas of PCBs to the subestuaries. We further evaluated the importance of source proximity by calculating three inverse-distance weighted (IDW) metrics of development, an approach that weighted developed land near the shoreline more heavily than developed land farther away. Unweighted percentages of each of the four measures of developed land explained 51-69% of the variance in t-PCBs. However, IDWs markedly improved the relationships between % developed land measures and t-PCBs. Percent commercial land, weighted by its simple inverse distance, explained 99% of the variance in t-PCBs, whereas the other three measures explained as much as 93-97%. PCBs historically produced or used in commercial and residential areas are apparently persisting in the environment atthe scale of the watersheds and subestuaries examined in this study, and developed land close to the subestuary has the greatest unit effect on t-PCBs in fish. These findings provide compelling evidence for a strikingly strong linkage between watershed land use and t-PCBs in white perch, and this relationship may prove useful for identifying unsampled subestuaries with a high risk of PCB contamination.  相似文献

Passive optical networks are a prominent broadband access solution to tackle the "last mile" bottleneck in telecommunications infrastructure. Data transmission over standardized PONs is divided into time slots. Toward the end of PON performance improvement, a critical issue relies on resource management in the upstream transmission from multiple optical network units (ONUs) to the optical line terminal (OLT). This includes resource negotiation between the OLT and the associated ONUs, transmission scheduling, and bandwidth allocation. This article provides an overview of the resource management issues along with the state-of-the-art schemes over time-division multiplexed PONs (TDM-PONs). We categorize the schemes in the literature based on their features, and compare their pros and cons. Moreover, we introduce a unified state space model under which all TDM-PON resource management schemes can be evaluated and analyzed for their system level characteristics. Research directions are also highlighted for future studies.  相似文献

Although tin oxide has been the most widely investigated metal oxide material for gas detection, it suffers from the large resistance and high operating temperature. This could be overcome by hybridization with nanostructured carbon. In this work, tin oxide nanoparticles with ultrasmall sizes of 1-3 nm have been uniformly coated onto bundles of single-walled carbon nanotubes by a surfactant assisted solid state synthesis approach for the first time. Gas sensor properties of the as-synthesized nanocomposite material toward NO2 (from 5 to 60 ppm) are measured at 150 degrees C. Compared to the pure carbon tubes gas sensors, the nanocomposite gas sensor responds to NO2 in low concentrations with good linearity, high sensitivity, and fast recovery, while working at a relatively low temperature.  相似文献

The surface tensions of pure molten aluminum, A356 alloy (Al-7 pct Si-0.3 pct Mg), and strontium-modified A356 alloy have been measured under vacuum and hydrogen atmospheres using the sessile drop technique. The values obtained for pure aluminum at 680 °C and for A356 alloy and modified A356 alloy at 630 °C are 1.007, 0.889, and 0.844 N/m, respectively, when measured under vacuum. The addition of hydrogen gas to the atmosphere of the liquid droplet has no significant effect on the surface tension of the unmodified A356 alloy, while it lowers the surface tension of the modified alloy to 0.801 N/m. This effect is possibly due to the formation of SrH2.  相似文献
