In this paper we consider the optimization of general 3D truss structures. The design variables are the cross-sections of the truss bars together with the joint coordinates, and are considered to be continuous variables. Using these design variables we simultaneously carry out size optimization (areas) and shape optimization (joint positions). Topology optimization (removal and introduction of bars) is only considered in the sense that bars of minimum cross-sectional area will have a negligible influence on the performance of the structure. The structures are subjected to multiple load cases and the objective of the optimizations is minimum mass with constraints on (possibly multiple) eigenfrequencies, displacements, and stresses. For the case of stress constraints, we deal differently with tensile and compressive stresses, for which we control buckling on the element level. The stress constraints are imposed in correlation with industrial standards, to make the optimized designs valuable from a practical point of view. The optimization problem is solved using SLP (Sequential Linear Programming).  相似文献

Design, technological features, and performance of polymeric diffractive optical coupling elements (DOCEs), developed for surface plasmon resonance (SPR) sensors are reported. The concept is based on input and output coupling of collimated and perpendicularly incident light beams to exchangeable SPR sensor chips. In the SPR sensor chips, one DOCE couples the input light beam to a (bio-)chemical sensor surface and another DOCE images the output light beam onto a detector array. The manufacturing technique is based on an injection moulding process similarly to the manufacturing process of compact discs allowing precision manufacturing and reproduction of the grating topography of the DOCEs. The DOCE-based SPR sensor chip is specifically suitable for miniaturisation and large scale production, while maintaining high optical quality and performance.  相似文献

The authors modeled depressive and anxiety symptom data from 1,391 participants in a longitudinal study of middle-aged and older Swedish twins (M age?=?60.9 years, SD?=?13.3). Although anxiety and depression were highly correlated, a model with distinct Anxiety and Depression factors fit the data better than models with Positive and Negative Affect factors or a single Mental Health factor. Lack of well-being was associated with anxiety rather than depression. Over two 3-year intervals, anxiety symptoms led to depressive symptoms, but the relationship was not reciprocal. Anxiety symptoms were more stable than depression. These findings provide additional support for the idea that anxiety symptoms may reflect a personality trait such as neuroticism more than do depressive symptoms and suggest that low positive affect may not be as specific to depression among older adults as in younger people. The actuator disk is a well‐known and widely used theoretical tool in wind engineering. This work proposes a new theory based on an actuator surface, capable of treating time‐varying vectorial load distributions and yaw/pitch misalignment. A simplified representation of vortex motion is used to arrive at a tractable problem. The theory is not restricted to disks; arbitrary coplanar (optionally disjoint) actuator surfaces may be modeled. Some of the theoretical novelties used in the modeling includes use of the fractional Laplacian and extensive use of the Fourier transform on . Promising experimental validation based on pitch‐step experiments at the Tjæreborg turbine is furnished. Comparisons are also made to existing methodologies. Analysis and numerical work shows that the model encapsulates Coleman's vortex theory.  相似文献

PDZ domains are ubiquitous small protein domains that are mediators of numerous protein–protein interactions, and play a pivotal role in protein trafficking, synaptic transmission, and the assembly of signaling‐transduction complexes. In recent years, PDZ domains have emerged as novel and exciting drug targets for diseases (in the brain in particular), so understanding the molecular details of PDZ domain interactions is of fundamental importance. PDZ domains bind to a protein partner at either a C‐terminal peptide or internal peptide motifs. Here, we examined the importance of a conserved Lys/Arg residue in the ligand‐binding site of the second PDZ domain of PSD‐95, by employing a semisynthetic approach. We generated six semisynthetic PDZ domains comprising different proteogenic and nonproteogenic amino acids representing subtle changes of the conserved Lys/Arg residue. These were tested with four peptide interaction partners, representing the two different binding modes. The results highlight the role of a positively charged amino acid in the β1–β2 loop of PDZ domains, and show subtle differences for canonical and noncanonical interaction partners, thus providing additional insight into the mechanism of PDZ/ligand interaction.  相似文献

Systemic Intermittent Hypoxic Therapy (IHT) relies on the adaptive response to hypoxic stress. We investigated allogenic bone-graft resorption in the lumbar spine in 48 mice. The mice were exposed to IHT for 1 week before surgery or 1 week after surgery and compared with controls after 1 and 4 weeks. Complete graft resorption was observed in 33–36% of the animals in the control group, but none in the preoperative IHT group. Increased bone-graft volume was demonstrated by micro-computed tomography in the preoperative IHT group after 1 week (p = 0.03) while a non-significant difference was observed after 4 weeks (p = 0.12). There were no significant differences in the postoperative IHT group. Increased concentration of immune cells was localized in the graft area, and more positive tartrate-resistant acid phosphatase (TRAP) staining was found in controls compared with IHT allogenic bone grafts. Systemic IHT resulted in a significant increase of the major osteoclast inhibitor osteoprotegerin as well as osteogenic and angiogenic regulators Tgfbr3, Fst3l, Wisp1, and Vegfd. Inflammatory cytokines and receptor activator of nuclear factor kappa-B ligand (RANKL) stimulators IL-6, IL-17a, IL-17f, and IL-23r increased after 1 and 4 weeks, and serum RANKL expression remained constant while Ccl3 and Ccl5 decreased. We conclude that the adaptive response to IHT activates numerous pathways leading to inhibition of osteoclastic activity and inhibition of allogenic bone-graft resorption.  相似文献

Kinetics of oxytetracycline reaction with a hydrous manganese oxide   总被引:4,自引:0,他引:4  
Tetracycline antibiotics comprise a class of broad spectrum antimicrobial agents finding application in human therapy, animal husbandry, aquaculture, and fruit crop production. To better understand the processes affecting these antibiotics in soils and sediments, the kinetics of oxytetracycline transformation by a hydrous manganese oxide (MnO2) were investigated as a function of reactant concentration, pH, and temperature. Oxytetracycline was rapidly degraded by MnO2. Initial reaction rates exhibited pronounced pH-dependence, increasing as pH decreased. Reaction of oxytetracycline with MnO2 was accompanied by generation of Mn(II) ions, suggesting oxidative transformation of the antibiotic. At pH 5.6, apparent reaction orders for oxytetracycline and MnO2 were 0.7 and 0.8. Reaction order with respect to H+ was 0.6 between pH 4 and 9. Initial reaction rates increased by a factor of approximately 2.4 for 10 degrees C temperature increases; the apparent activation energy (60 kJ x mol(-1)) was consistent with a surface-controlled reaction. Reactivity of tetracycline antibiotics toward MnO2 increased in the following order: rolitetracyline oxytetracycline < or =tetracycline approximately meclocycline < chlortetracycline. The initial rate of chlortetracycline degradation by MnO2 was substantially larger than that of the other tetracycline antibiotics investigated. MnO2 reactivity toward oxytetracycline decreased with time; a retarded rate equation was used to describe oxytetracycline reaction with MnO2 under declining rate conditions. This study indicates that natural manganese oxides in soils and sediments are likely to promote appreciable degradation of tetracycline antibiotics, and that reaction rates are strongly dependent on reaction time scale and solution conditions.  相似文献

Three samples of soybean meal (SBM), 3 samples of expeller SBM (SoyPlus, West Central Cooperative, Ralston, IA), 5 samples of distillers dried grains with solubles (DDGS), and 5 samples of fish meal were used to evaluate the furosine and homoarginine procedures to estimate reactive Lys in the rumen-undegraded protein fraction (RUP-Lys). One sample each of SBM, expeller SBM, and DDGS were subjected to additional heat treatment in the lab to ensure there was a wide range in reactive RUP-Lys content among the samples. Furosine is a secondary product of the initial stages of the Maillard reaction and can be used to calculate blocked Lys. Homoarginine is formed via the reaction of reactive Lys with O-methylisourea and can be used to calculate the concentration of reactive Lys. In previous experiments, each sample was ruminally incubated in situ for 16 h, and standardized RUP-Lys digestibility of the samples was determined in cecectomized roosters. All rumen-undegraded residue (RUR) samples were analyzed for furosine and Lys; however, only 9 of the 16 samples contained furosine, and only the 4 unheated DDGS samples contained appreciable amounts of furosine. Blocked RUP-Lys was calculated from the furosine and Lys concentrations of the RUR. Both the intact feed and RUR samples were evaluated using the homoarginine method. All samples were incubated with an O-methylisourea/BaOH solution for 72 h and analyzed for Lys and homoarginine concentrations. Reactive Lys concentrations of the intact feeds and RUR were calculated. Results of the experiment indicate that blocked RUP-Lys determined via the furosine method was negatively correlated with standardized RUP-Lys digestibility, and reactive RUP-Lys determined via the guanidination method was positively correlated with standardized RUP-Lys digestibility. Reactive Lys concentrations of the intact samples were also highly correlated with RUP-Lys digestibility. In conclusion, the furosine assay is useful in predicting RUP-Lys digestibility of DDGS samples, and the guanidination procedure can be used to predict RUP-Lys digestibility of SBM, expeller SBM, DDGS, and fish meal samples.  相似文献

This work concerns the on-going development of efficient and environmentally friendly antifouling paints for biofouling control on large ocean-going ships. It is illustrated how a detailed mathematical model for a self-polishing antifouling paint exposed to seawater can be used as a product engineering tool to obtain a quick estimate of the paint behaviour that a given seawater-soluble pigment will provide. In the present context, “pigment” refers to relevant particulate solids of organic-, inorganic-, or biological nature. Simulations performed at 15 and 30 °C suggest that pigment solubility and seawater diffusivity of dissolved pigment species have a significant influence on the polishing and leaching behaviour of a typical self-polishing paint system. The pigment size distribution, on the other hand, only has a minor influence on the paint–seawater interaction. Simulations also indicate that only compounds which are effective against biofouling at very low seawater concentrations are useful as active antifouling paint ingredients. The need for model verification and exploration of practical issues, subsequent a given pigment has been found of interest, is discussed. The model approach is of relevance in the search for novel antifouling paints and for the development of accelerated test methods.  相似文献
