OBJECTIVES: Major operative trauma like aorta-coronary bypass operation may lead to postoperative immunodisturbance, putting the patient at an increased risk for infection and sepsis. The monocyte/macrophage system and the endotoxin receptor CD14 are important in the early recognition and elimination of invading bacteria. The aim of this study was to analyze changes in membrane-associated CD14 and soluble CD14 during and after cardiac involving cardiopulmonary bypass. METHODS: We studied numbers of leukocytes, monocytes, and monocyte subpopulations, expression of monocyte membrane-associated CD14 and plasma levels of soluble CD14 in 10 patients (63 +/- 8 years of age), who underwent elective cardiopulmonary bypass. RESULTS: Cardiopulmonary bypass induced marked postoperative monocytosis, which was maximal 20 hours after the operation (485 +/- 242 cells/microl before, 1080 +/- 264 cells/microl 20 hours after surgery). Expression of membrane-associated CD14 on classical CD14++ monocytes decreased significantly by 40%, reaching a nadir 20 hours after surgery (p < 0.05). At the time of maximal membrane-associated CD14 suppression, the levels of soluble CD14 measured by enzyme-linked immunosorbent assay were clearly increased (3.2 +/- 1.0 microg/ml before versus 5.6 +/- 1.0 microg/ml 20 hours after, p < 0.001). No significant change of the percentage of small (alpha) and large (beta) forms of soluble CD14 was found. CONCLUSIONS: Cardiopulmonary bypass leads to reduced membrane-associated CD14 expression on peripheral blood monocytes and increased levels of soluble CD14 through shedding or secretion of membrane-associated CD14 from the cell surface. These findings indicate that bypass is associated with significant monocyte activation.  相似文献

Initiation factor eIF4E binds to the 5'-cap of eukaryotic mRNAs and plays a key role in the mechanism and regulation of translation. It may be regulated through its own phosphorylation and through inhibitory binding proteins (4E-BPs), which modulate its availability for initiation complex assembly. eIF4E phosphorylation is enhanced by phorbol esters. We show, using specific inhibitors, that this involves both the p38 mitogen-activated protein (MAP) kinase and Erk signaling pathways. Cell stresses such as arsenite and anisomycin and the cytokines tumor necrosis factor-alpha and interleukin-1beta also cause increased phosphorylation of eIF4E, which is abolished by the specific p38 MAP kinase inhibitor, SB203580. These changes in eIF4E phosphorylation parallel the activity of the eIF4E kinase, Mnk1. However other stresses such as heat shock, sorbitol, and H2O2, which also stimulate p38 MAP kinase and increase Mnk1 activity, do not increase phosphorylation of eIF4E. The latter stresses increase the binding of eIF4E to 4E-BP1, and we show that this blocks the phosphorylation of eIF4E by Mnk1 in vitro, which may explain the absence of an increase in eIF4E phosphorylation under these conditions.  相似文献

Brucellae are Gram-negative, aerobic, non-motile coccobacilli causing brucellosis in man and animals. The disease is one of the most significant yet neglected global zoonoses. Especially in developing countries, brucellosis is causing public health problems and economic losses to private animal owners and national revenues. Composed of oligonucleotides, aptamers are chemical analogues of antibodies that are promising components for developing aptamer-based rapid, sensitive, and specific tests to identify the Brucella group of bacteria. For this purpose, aptamers were generated and selected by an enhanced protocol of cell systematic evolution of ligands by exponential enrichment (cell-SELEX). This enhanced cell-SELEX procedure involved the combination of both conventional and toggle cell-SELEX to boost the specificity and binding affinity to whole Brucella cells. This procedure, combined with high-throughput sequencing of the resulting aptamer pools, comprehensive bioinformatics analysis, and wet lab validation assays, led to the selection of a highly sensitive and specific aptamer for those Brucella species known to circulate in Egypt. The isolated candidate aptamer showed dissociation constant (KD) values of 43.5 ± 11, 61.5 ± 8, and 56 ± 10.8 nM for B. melitensis, B. abortus, and B. suis, respectively. This is the first development of a Brucella-specific aptamer using an enhanced combination of conventional and toggle cell-SELEX to the authors’ best knowledge.  相似文献

Heat flow was calculated from XRD data and compared with measured heat flow from calorimetric experiments. It was shown that the heat released during the hydration of a commercial Ordinary Portland Cement can be assigned mainly to three mechanisms, the silicate reaction (sum of dissolution of alite and precipitation of C-S-H-phase and portlandite), the dissolution of C3A, and the precipitation of ettringite. The contributions made by anhydrite dissolution and gypsum dissolution to the heat released during hydration turned out to be quite small. It is possible to explain, on the basis of the data produced, the origin of the heat flow curve of the cement used.  相似文献

The hydration process of a commercial Portland cement was followed by means of heat flow calorimetry. The measured heat flow was compared with calculated heat flow curves based on XRD data. Examined in particular was the influence of one selected superplasticizer on the hydration of the Portland cement. It was shown that the superplasticizer in question retards both the aluminate reaction and the silicate reaction. It is certainly conceivable that there are more than only one explanation for the interaction between the superplasticizer and the cement. A complexation of Ca2 + ions from pore solution by the superplasticizer is as thinkable as the adsorption of the polymer on the nuclei or the anhydrous grain surfaces which in turn might lead to the prevention of the growth of the nuclei or the dissolution of the anhydrous grains.  相似文献

Hydration of partially amorphized α‐TCP powders with Sr2+ concentrations ranging from 0 to 10 mol% substitution for Ca2+ was analyzed by isothermal calorimetry and quantitative in‐situ XRD. Hydration of both crystalline α‐TCP and amorphous TCP (ATCP) forming CDHA was retarded to an increasing extent with increasing Sr2+ content. Sr2+ slightly reduced the crystallite size (XRD coherent scattering domains) of the CDHA formed during hydration, while the size of crystals visible under SEM was not noticeably affected. Reaction enthalpies of ΔHR(Sr‐α‐TCP→Sr‐CDHA) = 122 ± 8 J/gTCP and ΔHR(Sr‐ATCP→Sr‐CDHA) = 257 ± 8 J/gTCP were determined for the hydration of crystalline α‐TCP and ATCP containing 5 mol% Sr2+ substitution for Ca2+. This is comparable with the corresponding reaction enthalpies previously obtained for undoped samples, which are 106 ± 7 J/gTCP for α‐TCP and 250 ± 7 J/gTCP for ATCP.  相似文献

Tetra(arylalkynyl)pyridines and tetra(arylalkenyl)pyridines and their benzene analogues were prepared by palladium‐catalyzed cross‐coupling reactions from the commercially available chlorinated substrates in good to excellent yields. The photophysical properties of selected compounds were investigated and compared to each other. Very good fluorescence quantum yields were observed, especially for the pyridine derivatives.  相似文献

The structural information on the influence of ionic additions in biphasic (hydroxyapatite (HAP) and β-tricalciumphosphate (β-TCP)) mixtures ranging from single ionic substitutions to combined ionic substitutions of most of the essential ions embedded in biological apatite was analyzed through the Rietveld refinement technique. The results have proved that the determined quantitative phase composition of HAP and β-TCP in biphasic mixtures was dependent on the initial calcium (Ca) deficiency of the precursor powders precipitated from the different molar concentrations used in the synthesis. The substitution of cations (Na+, Mg2+, and K+) improved the stabilization of the β-TCP structure whereas anions (F and Cl) were found incorporated at the OH site of the HAP phase. Rietveld analysis of X-ray powder diffraction data from the present study proved to be a powerful technique to describe the position and occupancy of certain ions like Mg2+ and Cl in the biphasic mixtures. However, it has also shown limitations in tracking back other ions like Na+, K+, and F, which require the use of other complementary characterization methods.  相似文献
