The multiple cellular and molecular processes induced by injury to the central nervous system (CNS) are still poorly understood. In the present study, we investigated the response of the vasculature and the expression of mRNA for the angiogenic vascular endothelial growth factor (VEGF) following X-irradiation of the spinal cord in the newborn and following traumatic spinal cord injury in the adult rat. Both lesion models induced changes in the density and the distribution pattern of blood vessels: while X-irradiation led to a permanent local increase in vascular density in the fibre tracts of the exposed segments, a transient local sprouting of vessels was induced upon traumatic spinal cord injury. In situ hybridization showed that an increase of VEGF mRNA anticipated and overlapped with the vascular responses in both lesion models. In addition to the temporal correlation of VEGF expression and vascular sprouting, there was a clear correlation in the spatial distribution patterns. Following X-irradiation, the expression of VEGF mRNA was restricted to the fibre tracts, precisely the areas where the changes in the vasculature were observed later on. Upon transection in the adult animal, VEGF was mainly detectable at the border of the lesion area, where the transient increase in vascular density could be observed. Interestingly, according to the type of lesion applied, astrocytes (X-irradiation) or inflammatory cells (presumably microglial cells or macrophages; traumatic lesion) are the cellular sources of VEGF mRNA. Our results strongly indicate that VEGF is crucially involved in mediating vascular changes following different types of injury in the CNS.  相似文献

The most commonly reported failure mode of sliding hip screws in published literature is cut-out of the lag screw. This study investigates the resistance to failure of the femoral head, with lag screws used in two types of sliding hip screws, the gamma locking nail (Howmedica) and the dynamic hip screw (DHS) (Synthes). The investigation consisted of biomechanical tests under static loading conditions on 12 pairs of cadaveric femoral heads, to establish the failure loads due to screw cut-out for the two implant lag screws. The gamma nail appeared to reduce the tendency to cut-out in the osteoporotic bone (soft) associated with elderly patients in whom these devices are commonly used (p < 0.05). In high density bone (hard) the gamma lag screw also appeared to be stronger, because the DHS showed a tendency to bend. The larger diameter of the gamma nail lag screw resists bending and appears to reduce the risk of cut-out compared with the DHS.  相似文献

Binding of the stable melanocortin(4-9) analogue, Org2766 [Met(O2)-Glu-His-Phe-D-Lys-Phe] to cultured rat sciatic nerve Schwann cells was demonstrated using a biotinylated derivative in semiquantitative histochemical and CELISA assays. Org2766 bound to Schwann cells, but not to fibroblasts, and was displaced maximally by unlabeled Org2766, alpha-MSH and ACTH(1-24). Displacement of Org2766 from the binding sites was considerably reduced by N- and C-truncation of the peptide. Specific binding of Org2766 was also demonstrated in the immortal rat Schwann cell line SCL4.1/F7 and was more pronounced in cells displaying a differentiated morphology. Org2766 and alpha-MSH increased cyclic AMP content of Schwann cells but neither stimulated DNA synthesis when applied alone. However, in the presence of a priming (subthreshold) concentration of the mitogen, cholera toxin, Org2766 and alpha-MSH caused a delayed increase in DNA synthesis. Org2766 did not modulate the expression of several differentiation-related Schwann cell markers. However, Org2766 increased immunoreactivity for p75 low-affinity NGF receptor on Schwann cells and evoked the release of neurotrophic factor(s) that synergized with NGF in stimulating neurite outgrowth in rat DRG neurons. The results indicate that Schwann cells are a primary target for the action of Org2766 and provide evidence for an indirect mechanism by which melanocortins might stimulate neurite sprouting in regenerating peripheral nerve axons.  相似文献

A complex history of Cenozoic fluvial activity in the presently hyperarid Eastern Sahara is inferred from SIR data and postflight field investigations in southwest Egypt and northwest Sudan. SIR images were coregistered with Landsat and existing maps as a guide to exploration of the buried paleodrainages (radar rivers) first discovered by SIR-A. Field observations explain the radar responses of three types of radar rivers, RR-1, RR-2, and RR-3. RR-1 (broad, aggraded valleys or basins filled with alluvium) generally give a dark response relative to the surrounding radar-bright tone of the wind-roughened, discontinuously sand-covered bedrock of the remnant interfluves. The alluvial fill in some of the RR-1 valleys gives a locally intermediate to bright response where cemented by diagenetic CaCO3 (caliche). The RR-2 type (braided channels inset in the RR-1 valleys) are filled with noncalichified, loose sediment, and are visible on SIR because of the "caliche effect"-the dark response of unconsolidated alluvium in the inset channels contrasts with the brighter response of calichified alluvium in the broad valleys. The third type, RR-3 (narrow, long, bedrock-incised channels), which are partly visible on the ground and on Landsat in areas where the sand sheet is patchy, are conspicuous on SIR because the dark response of their unconsolidated channel fill contrasts with the brighter response of the surrounding bedrock. Disconformably overlying and concealing the RR-1 valleys, the RR- 2 channels, and parts of the RR-3 channels are eolian sand-sheetsheet de- deposits, which in turn are locally overridden by trains of barchan dunes.  相似文献

Silicon-based ceramics are prone to severe oxidation and volatilization problems in the presence of water vapor. To address these issues, mullite has been investigated in recent years as a possible protective coating material. To analyze the stability of such coatings, a multi-species diffusion model for ionic species is developed. Onsager reciprocity is assumed for the mobility coefficients and the resulting moving boundary problem is solved numerically. The formulation also accounts for substantial Si volatilization away from the outer surface of the coating. This model is used to analyze an initial transient leading to the formation of an internal silica layer, a second transient period that follows this silica formation, and a steady-state regime that can only be attained if Si diffusion is sufficiently fast.  相似文献

Giant silk moths (Lepidoptera: Saturniidae) typically are not well represented as larvae or adults in community level inventories of Lepidoptera, and as a result, little is known about their population dynamics. Furthermore, in recent years, many species of silk moths appear to have experienced population declines. Volatile sex pheromones are powerful sampling tools that can be used in operational conservation and monitoring programs for insects. Here, we describe the identification of the sex attractant pheromone of a giant silk moth, the luna moth Actias luna. Coupled gas chromatography-electroantennographic detection and gas chromatography-mass spectrometric analyses of extracts from pheromone glands of female luna moths supported the identification of (6E,11Z)-6,11-octadecadienal (E6,Z11–18:Ald), (6E)-6-octadecenal (E6–18:Ald), and (11Z)-11-octadecenal (Z11–18:Ald) as the compounds in extracts that elicited responses from antennae of male moths. These identifications were confirmed by synthesis, followed by testing of blends of the synthetic compounds in field trials in Ontario, Canada, and Kentucky, USA. Male moths were attracted to synthetic E6,Z11–18:Ald as a single component. Attraction appeared to be enhanced by addition of E6–18:Ald but not Z11–18:Ald, suggesting that the luna moth pheromone consists of a blend of E6,Z11–18:Ald and E6–18:Ald.  相似文献
