The first 2 years of the ITER IO has seen substantial progress towards the construction of the magnets, in three main areas. Firstly, the design has been developed under the conflicting constraints to minimise construction costs and to maximise plasma physics performance. Building construction momentum while updating the design to take account of new physics assessments of the coil requirements has been challenging. Secondly, with a stabilising design, it has been possible for the Domestic Agencies to launch the first industrial procurement contracts. And thirdly, critical R&D to confirm the performance of the Nb3Sn cable in conduit design is proceeding successfully.The design consolidation has been accompanied by design reviews involving the international community. The reviews conducted by magnet experts have enabled a consensus to be built on choosing between some of the design options in the original ITER basic design in 2001. The major design decisions were to maintain the circular Nb3Sn conductor embedded in radial plates for the toroidal field (TF) coils and to maintain NbTi-based conductors for the PF coils. Cold testing, at low current, is also being introduced for quality control purposes for all coils.  相似文献

The common practice of increasing dietary energy density during the close-up dry period (last ~3 wk prepartum) has been recently associated with a higher incidence of metabolic disorders after calving. Despite these reports, over-feeding of metabolizable energy (ME) during the far-off, nonlactating period is a common management policy aimed at achieving optimum calving body condition score (BCS) in pasture-based systems, as cows are generally thinner than total mixed ration cows at the end of lactation. Our hypothesis was that both far-off and close-up overfeeding influence the peripartum adipose tissue changes associated with energy balance and inflammatory state. Sixty mid-lactation, grazing dairy cows of mixed age and breed were randomly allocated to 1 of 2 groups that were managed through late lactation to achieve a low and high BCS (approximately 4.25 and 5.0 on a 10-point scale) at dry-off. The low BCS cows were then overfed ME to ensure that they achieved the same BCS as the higher BCS group by calving. Within each rate of BCS gain treatment, cows were offered 65, 90, or 120% of their pre-calving ME requirements for 3 wk pre-calving in a 2 × 3 factorial arrangement of treatments (i.e., 10 cows/treatment). Subcutaneous adipose tissue was collected via biopsy at ?1, 1, and 4 wk relative to parturition. Quantitative PCR was used to measure mRNA and microRNA expression of targets related to adipogenesis and inflammation. Cows overfed in the far-off period had increased expression of miR-143 and miR-378 prepartum (?1 wk) indicating greater adipogenesis, consistent with their rapid gain in BCS following dry-off. Furthermore, the lower postpartum expression of IL6, TNF, TLR4, TLR9, and miR-145, and a higher abundance of miR-99a indicated lower body fat mobilization in early lactation in the same group. In the close-up period, feeding either 65 or 120% of ME requirements caused changes in FASN, IL1B, IL6R, TLR9, and the microRNA miR-143, miR-155, and miR-378. Their respective expression patterns indicate a tentative negative-feedback mechanism in metabolically compromised, feed-restricted cows, and a possible immune-related stimulation of lipolysis in apparently static adipocytes in overfed cows. Data from cows fed 90% of ME requirements indicate the existence of a balance between lipolytic (inflammatory-related) and anti-lipolytic signals, to prime the mobilization machinery in light of imminent lactation. Overall, results indicate that far-off dry cow nutrition influences peripartum adipose tissue metabolism, with neither strategy negatively affecting the physiological adaptation to lactation. Furthermore, to ensure a favorable transition, cows should be subjected to a small feed restriction in the close-up period, irrespective of far-off nutritional management.  相似文献

This work evaluates chronic safety in middle‐aged ovariectomized rats supplemented with different dosages of green tea polyphenols (GTP) in drinking water. The experiment used 6‐mo‐old sham (n = 39) and ovariectomized (OVX, n = 143) female rats. All sham (n = 39) and 39 of the OVX animals received no GTP treatment and their samples were collected for outcome measures at baseline, 3 mo, and 6 mo (n = 13 per group for each). The remaining OVX animals were randomized into 4 groups receiving 0.15%, 0.5%, 1%, and 1.5% (n = 26 for each) of GTP (wt/vol), respectively, in drinking water for 3 and 6 mo. No mortality or abnormal treatment‐related findings in clinical observations or ophthalmologic examinations were noted. No treatment‐related macroscopic or microscopic findings were noted for animals administered 1.5% GTP supplementation. Throughout the study, there was no difference in the body weight among all OVX groups. In all OVX groups, feed intake and water consumption significantly decreased with GTP dose throughout the study period. At 6 mo, GTP intake did not affect hematology, clinical chemistry, and urinalysis, except for phosphorus and blood urea nitrogen (increased), total cholesterol, lactate dehydrogenase, and urine pH (decreased). This study reveals that the no‐observed‐adverse‐effect level (NOAEL) of GTP is 1.5% (wt/vol) in drinking water, the highest dose used in this study.  相似文献

The recent EU Commission proposal for promoting the supply of power from renewable energy sources was originally based on a pan-European, harmonised tradable green certificate (TGC) scheme. We suggest, on the basis of a multi-disciplinary analysis, that a pan-EU TGC system is not the way forward for Europe. It is vital that the Commission (and the majority of Member States) avoids implementation of such policy designs put forward by a coalition of vested interests. They should instead look at, and act upon, the available evidence from those countries that have experimented with TGCs (e.g. Flanders, UK and Sweden) and design policies that stand a better chance of meeting the criteria of effectiveness, efficiency and equity. In particular, the policies must enable EU to meet the immense innovation/industrialisation challenge by inducing the development of a capital goods industry that can, eventually, diffuse a broad range of technologies that use renewable energy sources. Only then we can acquire an ability to implement an industrial revolution in the energy system in a way that broadly meets the criteria of effectiveness and dynamic efficiency.  相似文献

One of the initial steps of modern drug discovery is the identification of small organic molecules able to inhibit a target macromolecule of therapeutic interest. A small proportion of these hits are further developed into lead compounds, which in turn may ultimately lead to a marketed drug. A commonly used screening protocol used for this task is high-throughput screening (HTS). However, the performance of HTS against antibacterial targets has generally been unsatisfactory, with high costs and low rates of hit identification. Here, we present a novel computational methodology that is able to identify a high proportion of structurally diverse inhibitors by searching unusually large molecular databases in a time-, cost- and resource-efficient manner. This virtual screening methodology was tested prospectively on two versions of an antibacterial target (type II dehydroquinase from Mycobacterium tuberculosis and Streptomyces coelicolor), for which HTS has not provided satisfactory results and consequently practically all known inhibitors are derivatives of the same core scaffold. Overall, our protocols identified 100 new inhibitors, with calculated Ki ranging from 4 to 250 μM (confirmed hit rates are 60% and 62% against each version of the target). Most importantly, over 50 new active molecular scaffolds were discovered that underscore the benefits that a wide application of prospectively validated in silico screening tools is likely to bring to antibacterial hit identification.  相似文献

Spiritual struggle appeared consistently to predict poor health outcomes, including mortality. Despite surging interest in the health benefits of religion and spirituality, the health hassle of existential conflicts and proinflammatory cytokines as a potential physiological mechanism has been overlooked. Based on psychological and theological assumptions, we argue for the universal nature of spiritual struggle, a crisis-related existential conflict, and for investigating its physiological influence as essential to understanding human nature. Increased levels of inflammatory cytokines such as interleukin-6 (IL-6) have been linked with adverse health outcomes and negative emotions. This study thus examined spiritual struggle related to plasma IL-6 in 235 adult patients undergoing cardiac surgery, along with positive religious coping, general coping, and optimism, controlling for standardized clinical medical indicators. Multiple regression analysis, following a preplanned sequence, showed that spiritual struggle (p = .011), behavioral coping (p = .013) were positively associated with excess plasma IL-6, controlling for medical correlates (e.g., left ventricular ejection fraction). We conclude that spiritual struggle, indicating the crisis in an existential relation, and behavioral coping strategies are associated with elevated pre-operative plasma IL-6. Membrane biofouling is one of the major obstacles limiting membrane applications in water treatment. In this study, Bdellovibrio bacteriovorus HD 100, a Gram-negative predatory bacterium, was evaluated as a novel way to mitigate membrane biofouling and its subsequent performance decline. Dead-end microfiltration (MF) tests were carried out on Escherichia coli DH5α and B. bacteriovorus HD 100 co-culture feed solutions. Predation of E. coli was performed at either a low or high multiplicity of infection (MOI), which is defined as the predator to prey cell ratio. The MOIs tested were 2 and 200, and the viability of both the E. coli prey and the predator was monitored over 48 h. The higher MOI (high predator, HP) culture showed a nearly 6-log loss in E. coli number after 24 h when compared to both the control and low MOI (low predator, LP) cultures, whereas the E. coli population within both predated cultures (HP and LP) became nearly identical at 48 h and 4-log lower than that of the control. The unpredated cultures led to significant loss in water flux at 12, 24, and 48 h of culture, but the HP and LP membranes showed less loss of flux by comparison. Analysis of the total membrane resistance showed a similar trend as the flux decline pattern; however, irreversible resistance of the membrane was much higher for the 48 h LP culture compared to the unpredated and HP cultures at 48 h. This increase in irreversible resistance was attributed mainly to E. coli debris, which accumulated in the medium after the predator lysed the prey cells. These results show that pretreatment of wastewater using a suitable concentration of predatory bacteria such as B. bacteriovorus can enhance membrane performance.  相似文献

This work presents the dielectrophoretic manipulation of sub-micron particles suspended in water and the investigation of their optical responses using a microfluidic system. The particles are made of silica and have different diameters of 600, 450, and 250 nm. Experiments show a very interesting feature of the curved microelectrodes, in which the particles are pushed toward or away from the microchannel centerline depending on their levitation heights, which is further analyzed by numerical simulations. In doing so, applying an AC signal of 12 Vp–p and 5 MHz across the microelectrodes along with a flow rate of 1 μl/min within the microchannel leads to the formation of a tunable band of particles along the centerline. Experiments show that the 250 nm particles guide the longitudinal light along the microchannel due to their small scattering. This arrangement is employed to study the feasibility of developing an optofluidic system, which can be potentially used for the formation of particles-core/liquid-cladding optical waveguides.  相似文献
