BACKGROUND: Meningeal hemangiopericytoma is an uncommon neoplasm with a high propensity for recurrence. The purpose of this study was to analyze the efficacy of different treatment options in patients with recurrent disease. METHODS: The records of all patients with recurrent meningeal hemangiopericytoma treated at the study institution between 1976 and 1996 were reviewed. RESULTS: Thirty-four consecutive patients were studied. The mainstay of treatment was brain surgery in 21 patients (62%); the median time to recurrence from surgery was 12 months. Ten patients (29%) had 20 recurrent central nervous system (CNS) lesions treated by stereotactic radiosurgery. Of these, 3 previously nonirradiated patients (all with lesion size < 25 mm) achieved a complete response, which persisted at a median of 3 years. In 14 lesions (70%) a partial response (PR) occurred with a median duration of 12 months, whereas 3 lesions (15%) remained stable. Two patients with inoperable CNS lesions received external beam radiation therapy and both had PRs lasting 14 and 24 months, respectively. Nine patients (26%) received radiation therapy for metastatic disease. Of these, seven patients remained stable with good symptomatic relief, and two patients experienced tumor progression. Chemotherapy with doxorubicin-containing regimens was administered in 7 patients (21%); there was only 1 PR that lasted 8 months. The median survival from first recurrence was 4.6 years. CONCLUSIONS: Surgical management is important for the successful treatment of patients with recurrent meningeal hemangiopericytoma. Radiosurgery plays a definite role in the treatment of smaller recurrent CNS lesions. Radiation therapy is helpful in the management of inoperable tumors. More effective chemotherapeutic agents are needed.  相似文献

Quorum sensing control mediated by N-acyl homoserine lactone (AHL) signaling molecules has been established as a key feature of the regulation of exoenzyme production in many gram-negative bacteria. In Chromobacterium violaceum ATCC 31532 a number of phenotypic characteristics, including production of the purple pigment violacein, hydrogen cyanide, antibiotics, and exoproteases are known to be regulated by the endogenous AHL N-hexanoyl-L-homoserine lactone (HHL). In this study we show that C. violaceum produces a set of chitinolytic enzymes whose production is regulated by HHL. The chitinolytic activity was induced in strains grown in the presence of chitin as the sole carbon source and quantitated in the secreted proteins by using p-nitrophenol analogs of disaccharide, trisaccharide, and tetrasaccharide oligomers of N-acetylglucosamine. By using 4-methylumbelliferyl analogs of the same oligomers of N-acetylglucosamine as substrates for proteins separated and renatured by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, at least six enzymes were detected: a chitobiase with high specificity to a dimeric substrate of 87 kDa, two N-acetylglucosaminidases with apparent molecular masses of 162 and 133 kDa, two endochitinases of 108 and 67 kDa, and a chitobiosidase of 56 kDa. In addition, two unidentified bands of >205 kDa were found where a tetrameric chitin derivative was used as a substrate. A pleiotropic mini-Tn5 mutant of C. violaceum (CV026) that is defective in HHL production and other quorum-sensing-regulated factors was also found to be completely deficient in chitinolytic activity. Growth of this mutant on minimal medium with chitin supplemented with culture supernatant from the C. violaceum wild-type strain or 10 microM synthetic HHL restored chitinase production to the level shown by the parental strain. These results constitute the most complete evidence so far for regulation of chitinolytic activity by AHL signaling in a gram-negative bacterium.  相似文献

Two different olivo-cortico-nuclear zones in the cat cerebellum have been compared quantitatively as regards the numbers of cells projecting to them from within several sources of mossy fibres (MFs), namely the basal pontine nuclei (BPN), nucleus reticularis tegmenti pontis (NRTP), and the ipsilateral lateral reticular nucleus (LRN). The zones chosen were the C3 zone in lobule V of the anterior lobe and the C1 zone in pars copularis of the paramedian lobule (PMLpc), localised by recording climbing fibre-mediated potentials evoked on their surface as a result of volleys set up in their spino-olivocerebellar paths. The zones were injected with fluorescent-labelled latex microspheres and cell bodies, retrogradely labelled in the MF source nuclei and in the contralateral inferior olive, were counted and mapped. Evidence was obtained that tracer efficiency was very high in both the MF projections and the olivo-cerebellar projection and that each olivocerebellar axon may provide only one climbing fibre to the upper part of a lobule V folium but an average of nearly two to the same part of a PML folium. When the numbers of labelled cells in each MF source nucleus were expressed as a percentage of the total number of labelled pontine cells, the biggest source for lobule V was the contralateral BPN, followed by LRN, contralateral NRTP, ipsilateral BPN, and ipsilateral NRTP. For PMLpc, the order was similar except that ipsilateral BPNs exceeded contralateral NRTPs, but the dominance of contralateral BPN as a source was much greater. Cell totals were converted into projection densities (i.e., numbers of cells labelled per square millimetre of cortical sheet involved in the injection site); densities for PMLpc were found to be almost three times greater than those for lobule V for contralateral BPN but the two densities were not significantly different for ipsilateral BPN. The three other MF sources projected at higher densities to lobule V than to PML. These findings indicate that two cortical zones, both of which receive climbing fibres from the rostral part of the dorsal accessory olive and project to nucleus interpositus anterior, nevertheless differ markedly in regard to both the relative and the absolute sizes of the projections they receive from several of their most important sources of MFs.  相似文献

The ability of Phanerochaete laevis HHB-1625 to transform polycyclic aromatic hydrocarbons (PAHs) in liquid culture was studied in relation to its complement of extracellular ligninolytic enzymes. In nitrogen-limited liquid medium, P. laevis produced high levels of manganese peroxidase (MnP). MnP activity was strongly regulated by the amount of Mn2+ in the culture medium, as has been previously shown for several other white rot species. Low levels of laccase were also detected. No lignin peroxidase (LiP) was found in the culture medium, either by spectrophotometric assay or by Western blotting (immunoblotting). Despite the apparent reliance of the strain primarily on MnP, liquid cultures of P. laevis were capable of extensive transformation of anthracene, phenanthrene, benz[a]anthracene, and benzo[a]pyrene. Crude extracellular peroxidases from P. laevis transformed all of the above PAHs, either in MnP-Mn2+ reactions or in MnP-based lipid peroxidation systems. In contrast to previously published studies with Phanerochaete chrysosporium, metabolism of each of the four PAHs yielded predominantly polar products, with no significant accumulation of quinones. Further studies with benz[a]anthracene and its 7,12-dione indicated that only small amounts of quinone products were ever present in P. laevis cultures and that quinone intermediates of PAH metabolism were degraded faster and more extensively by P. laevis than by P. chrysosporium.  相似文献

Most parts of the human body are cylindrical in shape. Generalized cylinders, with two cross-sectional openings, are a logical choice to represent these cylindrical shapes. However, a variety of human body regions can be visualized as surfaces with multiple openings or multimouth (MM) surfaces. Some examples of such surfaces are the pelvis, the chest, and the palms of the hands. We investigated the suitability of non-uniform rational B-spline (NURBS) formulation for creating multimouth surfaces. The aim was to compare a recently developed peptide alimentation method and the protein-free and regression methods for determining endogenous ileal nitrogen (N) and amino acid excretion in the rat, and to apply the endogenous loss estimates to determine the true ileal digestibility of a meat and bone meal (MBM). Preliminary investigations determined the effect of the time of sampling of digesta after a meal (slaughter method) for rats given a protein-free or an enzymically hydrolysed casein (EHC) based diet, on ileal digesta and endogenous N excretion. There was a significant (P < 0.05) effect of the time of sampling on the amount of digesta collected and the endogenous N excretion for both the EHC and protein-free fed rats. The amount of digesta collected from the terminal 20 cm of ileum and the endogenous N excretion for both the EHC and protein-free fed rats were greatest and least variable at 3 h post-feeding. In the main study, endogenous ileal amino acid excretions were determined in the growing rat fed an EHC-based diet (n = 6) and with subsequent treatment of the digesta using ultrafiltration or in six rats given a protein-free diet or by extrapolation from data for 30 animals given five diets which contained graded levels of MBM as the sole protein source. For the EHC treatment, the ileal digesta precipitate plus retentate was used to determine the endogenous flows. The ultrafiltration step excludes unabsorbed dietary amino acids from the measure of endogenous loss. Chromic oxide was the reference marker in all diets. The endogenous N flows determined by the protein-free and regression methods were similar but both significantly (P < 0.01) lower than those for rats fed the EHC-based diet. The mean endogenous ileal N flows determined by the peptide alimentation method, the protein-free and regression approaches were 1866, 1103 and 1019 μg g?1 freeze dry matter intake, respectively. The endogenous amino acid flows at the terminal ileum were lower when determined using the traditional protein-free or regression methods. The true ileal digestibility coefficients for MBM were considerably higher based on the EHC versus protein-free endogenous flows.  相似文献

Previous research has suggested that ratings used to make administrative decisions are lenient when compared with ratings obtained for research purposes only. The present study examined the effects of the purpose of rating on multivariate measures of accuracy in observing teacher behavior as well as measures of accuracy in evaluating teaching performance. 45 undergraduates viewed and evaluated videotaped lectures under conditions in which they were informed that their ratings would be used for research only or for making important decisions about those being rated. The purpose of rating did not affect measures of accuracy in rating the frequency with which a number of critical behaviors occurred on each tape. The purpose of rating also did not affect multivariate measures of performance rating accuracy. Purpose did, however, affect the relationship between accuracy in observing teacher behavior and accuracy in evaluating teaching performance. It is suggested that purpose affects the way in which raters process behavioral information without necessarily affecting the general level of rating. Heterologous primers were used to amplify an exon and intron-containing segment of the bovine homologue of the human dopachrome tautomerase gene. After confirmation of homology by sequence analysis (exon sequence similarity greater than 90%), bovine-specific primers were developed for synteny mapping purposes. The dopachrome tautomerase gene was assigned to bovine chromosome 12 (BTA12) with 97% concordance to the coagulation factor 10 locus. Together with previous synteny mapping of bovine chromosome 12 genes, fms-related tyrosine kinase, esterase D and 5-hydroxytryptamine receptor 2, this assignment further indicates conservation between human chromosome 13q and bovine chromosome 12.  相似文献

This longitudinal study of adolescents from a Nova Scotia coastal town examined whether change in self-reported depressive symptoms could be predicted from adolescents' concurrent and prior reports of anxiety symptoms and risk behaviors. Questionnaires were completed by 131 youths (80 females, 51 males; aged 12–17 yrs) in their classrooms 2 years apart. Results show that adolescents' concurrent reports of involvement in risk behaviors were linked to an increase in depressive symptoms only when they also were high on anxiety symptoms. Prior involvement in risk behaviors, but not prior anxiety symptoms, also predicted an increase in depressive symptoms. The importance of evaluating covariation among multiple types of symptoms and targeting the positive and negative consequences of risk behaviors adolescents is discussed.
