The standard assay for onchocerciasis diagnosis is microscopic detection of parasites in skin snips. Skin snipping is painful and may potentially transmit bloodborne infections. Thus, an alternative method for the diagnosis of onchocerciasis that does not require skin snipping is needed. A polymerase chain reaction (PCR)-based assay was shown to detect the presence of parasite DNA in superficial skin scrapings. Detection of parasite DNA in both skin snips and skin scratches was found to be more sensitive for detecting low-density infections than was microscopic examination of skin snips. The skin scratch PCR assay is minimally invasive and painless and does not present the risk of transmitting bloodborne infections. These properties make the skin scratch an attractive alternative to the skin snip for detecting O. volvulus infection. 相似文献

An introduction outlining the scope and limitations of this paper is followed by a brief explanation of the essential structural features of metal-complex dyes. The earliest metal-complex dyes were produced directly within the fibre material by reacting a metallisable dye with a chromium compound in situ. The first chromium complex dyes prepared in substance, were the chromium complexes containing sulpho-groups, synthesised by R Bohn of BASF, in 1912, the application of which was first made possible after the discovery by Ciba of a satisfactory dyeing process based on the use of a dyebath made strongly acid with sulphuric acid. These two classes of dye dominated the dyeing industry into the 1950s. At that time 1:2 metal-complex dyes without sulphonic acid groups but containing nonionic substituents as solubilising groups were introduced. These complexes were applicable from neutral to weakly acid dyebaths, which minimised the damage to the fibres. These dyeings were characterised by high fastness to light and wet treatments. Later, for commercial reasons, sulphonic acid groups were again introduced into the dye molecule to increase their hydrophilic properties. These dyes also have become well established as a result of the introduction of new dyeing methods and the development of dyeing auxiliaries designed to promote level dyeing. In addition to these types of metal-complex dyes, further metal-complex dyes selected from the extensive patent literature of the last 50 years because of their interesting structure are described. 相似文献

Death of oligodendrocyte (OL) precursors can be triggered in vitro by cystine deprivation, a form of oxidative stress that involves depletion of intracellular glutathione. We report here that OLs demonstrate maturation-dependent differences in survival when subjected to free radical-mediated injury induced by glutathione depletion. Using immunopanning to isolate rat preoligodendrocytes (preOLs), we generated highly enriched populations of preOLs and mature OLs under chemically defined conditions. Cystine deprivation caused a similar decrease in glutathione levels in OLs at both stages. However, preOLs were completely killed by cystine deprivation, whereas mature OLs remained viable. Although the glutathione-depleting agents buthionine sulfoximine and diethylmaleate were more potent in depleting glutathione in mature OLs, both agents were significantly more toxic to preOLs. Glutathione depletion markedly increased intracellular free radical generation in preOLs, but not in mature OLs, as indicated by oxidation of the redox-sensitive probe dihydrorhodamine 123. The antioxidants alpha-tocopherol, idebenone, and glutathione monoethylester prevented the oxidation of dihydrorhodamine in cystine-depleted preOLs and markedly protected against cell death. When the intracellular glutathione level was not manipulated, preOLs were also more vulnerable than mature OLs to exogenous free radical toxicity generated by a xanthine-xanthine oxidase system. Ultrastructural features of free radical-mediated injury in glutathione-depleted preOLs included nuclear condensation, margination of chromatin, and mitochondrial swelling. These observations indicate that preOLs are significantly more sensitive to the toxic effects of glutathione depletion and that oligodendroglial maturation is associated with decreased susceptibility to oxidative stress. 相似文献

Nondeterminism is introduced into an ordinary iterative programming language by treating procedure calls as nondeterministic assignment statements. The effect of such assignment statements is assumed to be determined solely by the entry-exit specifications of the corresponding procedures. The nondeterminism which this approach yields is not necessarily bounded. The paper discusses the problem of defining a denotational semantic for programming languages with this kind of, possibly unbounded, nondeterminism. As an additional constraint, the semantics is required to be continuous, in the sense that the underlying semantic algebra is continuous. Methylxanthines produce intracellular accumulation of cyclic 3'5'-AMP (cAMP) by inhibition of phosphodiesterase and mucosal cAMP accumulation. Cyclic AMP is thought to mediate small intestinal secretion caused by some enterotoxins, hormones, and methylxanthines. These studies were designed to evaluate the effect of caffeine on small intestinal net fluid movement and transit times. The administration of caffeine in amounts ordinarily contained in many beverages and medications (75 to 300 mg) resulted in striking net secretion in the jejunum which lasted at least 15 minutes. This occurred in six of seven studies. Baseline net absorption of 0.5 ml per cm per hr was reversed to net secretion of 6.0 +/- 2.2 ml per cm per hr after oral caffeine ingestion (P less than 0.01). Net secretion also occurred in the ileum in seven of eight studies, but the onset of secretion appeared 35 min later than in the jejunum. These patterns of secretion correlated best with the passage of the intestinal bolus of caffeine rather than plasma caffeine levels. In contrast to other net secretory conditions, which increase the speed of transit, small intestinal transit times, as determined by dye dilution methods, were unchanged by caffeine.
