Aluminium Alkyls with Heteroatoms. II. Preparation of Tris[(3-trimethylsilyl)-propyl]aluminium Tris[(3-trimethylsilyl)-propyl]aluminium is prepared by disproportionation and by an olefine displacement reaction in a circulation process. Four strains of Aujeszky's Disease virus (ADV) were included in this study; three Mexican field isolates (215,145 and C-8) in conjunction with the Shope reference strain of ADV, which has known pathogenic characteristics. All four strains were included in each treatment, which consisted of heat treatment, trypsin treatment and passed ten times on chicken embryo fibroblasts. Both virus titer and plaque size were determined on the first and tenth passage and on treated and untreated strains. On each of the treatments, the plaque size had significant differences (p = 0.001) which had relation to the two factors studied, namely strain and passage level. There was no significant variation related to the type of treatment between strains. With the strains under study, the authors also made rabbit pathogenicity tests, and it was found that on passage one, the strains caused clear nervous symptoms and death, while on the tenth passage level, the Mexican strains produced slight pruritus, few nervous symptoms and allowed the rabbits to survive. The mouse test revealed an increased median death time after the treatments, as well as a large increase in standard deviations. These data are interpreted as an increased heterogeneity of the strains in all of the treatments to the strains of viruses.  相似文献

Electron Impact Fragmentation of Trimethylsilylalkanones It has been shown that the fragmentation of trimethylsilyl alkanones on electron impact proceeds with a uniform mechanism which differs from that of silicon-free ketones by the fact, that no α cleavage is observed. It consists of a McLafferty rearrangement at the silicon-free alkyl group, followed by a McLafferty-type cleavage involving migration of the silyl group to yield the radical ion of the respective trimethylsilyl enolether.  相似文献

Major phenotypic changes occur in red cell membranes during the perinatal period, but the underlying molecular explanations remain poorly defined. Aquaporin CHIP, the major erythroid and renal water channel, was studied in perinatal rats using affinity-purified anti-CHIP IgG for immunoblotting, flow cytometry, and immunofluorescence microscopy. CHIP was not detected in prenatal red cells but was first identified in circulating red cells on the third postnatal day. Most circulating red cells were positive for CHIP by the seventh postnatal day, and this proportion rose to nearly 100% by the 14th day. The ontogeny of red cell CHIP correlated directly with acquisition of osmotic water permeability and inversely with Arrhenius activation energy. Only minor alterations in the composition of red cell membrane lipids occurred at this time. Immunohistochemical analysis of perinatal kidneys demonstrated a major induction of CHIP in renal proximal tubules and descending thin limbs at birth, coincident with the development of renal concentration mechanisms. Therefore, water channels are unnecessary for oxygen delivery or survival in the prenatal circulation, however CHIP may confer red cells with the ability to rehydrate rapidly after traversing the renal medulla, which becomes hypertonic after birth.  相似文献

After some preliminary experiments the influence of the choice of double-bond isomers on skeletal reactions on γ-alumina was investigated. Double-bond isomerization is not always fast compared with skeletal isomerization. Skeletal isomerization of hexenes was investigated using one double-bond isomer of each skeletal isomer as a feed. A complete reaction scheme was developed and discussed with respect to different reaction mechanisms. Together with results from 2,2,3-trimethylbutene isomerization it could be concluded that a nonclassical cyclopentyl mechanism alone may explain all the reactions.  相似文献

Material mixtures offer new possibilities for synthesizing coating materials with tailored optical and mechanical properties. We present experimental results on mixtures of HfO2, ZrO2, and Al2O3, pursuing applications in UV coating technology, while the mixtures are prepared by magnetron sputtering, ion beam sputtering, plasma ion-assisted deposition (PIAD), and electron beam evaporation without assistance. The properties investigated include the refractive index, optical gap, thermal shift, and mechanical stress. The first high reflectors for UV applications have been deposited by PIAD.  相似文献

Glycosylation has been shown to be important for proper routing and membrane insertion of a number of proteins. In the collecting duct, aquaporin-2 (AQP2) is inserted into the apical membrane after stimulation of vasopressin type-2 receptors and retrieved into an endosomal compartment after withdrawal of vasopressin. The extent of glycosylation of AQP2 in human kidney and urine and the effects of deglycoylation on routing of AQP2 in an AQP2-transfected Madin-Darby canine kidney cell line (clone WT10) were investigated. Semiquantitative immunoblotting of human kidney membranes and urine showed an AQP2 glycosylation of 35 to 45% for medulla, papilla, and urine, with low variation among individuals. The 1-desamino-8-D-arginine vasopressin-induced transcellular osmotic water permeability (Pf) of WT10 cells by a factor of 2.6 +/- 0.2 was reduced to 1.5 +/- 0.1 after pretreatment with the glycosylation inhibitor tunicamycin. However, when WT10 cells were incubated with 8-br-cAMP, the Pf increased by a factor 2.8 +/- 0.2 and by 2.9 +/- 0.2 after prior incubation with tunicamycin. Immunoblot analyses revealed that in WT10 cells, 34% of AQP2 is glycosylated, which was reduced to 2% after tunicamycin treatment. Surface biotinylation and subsequent semiquantitative immunoblotting revealed that stimulation by cAMP increased the level of AQP2 in the apical membrane of WT10 cells 1.5-fold. independent of the presence of tunicamycin. However, in tunicamycin-treated WT10 cells, all AQP2 in the apical membrane was unglycosylated, whereas in untreated cells 30% of AQP2 in the apical membrane was glycosylated. These results prove that glycosylation has no function in the routing of AQP2 in Madin-Darby canine kidney cells.  相似文献
