Fibrous hydrogels are a key component of soft animal tissues. They support cellular functions and facilitate efficient mechanical communication between cells. Due to their nonlinear mechanical properties, fibrous materials display non-trivial force propagation at the microscale, that is enhanced compared to that of linear-elastic materials. In the body, tissues are constantly subjected to external loads that tense or compress them, modifying their micro-mechanical properties into an anisotropic state. However, it is unknown how force propagation is modified by this isotropic-to-anisotropic transition. Here, force propagation in tensed fibrin hydrogels is directly measured. Local perturbations are induced by oscillating microspheres using optical tweezers. 1-point and 2-point microrheology are combined to simultaneously measure the shear modulus and force propagation. A mathematical framework to quantify anisotropic force propagation trends is suggested. Results show that force propagation becomes anisotropic in tensed gels, with, surprisingly, stronger response to perturbations perpendicular to the axis of tension. Importantly, external tension can also increase the range of force transmission. Possible implications and future directions for research are discussed. These results suggest a mechanism for favored directions of mechanical communication between cells in a tissue under external loads.  相似文献

DNA barcoding is a technique for discriminating and identifying species using short, variable, and standardized DNA regions. Here, we tested for the first time the performance of plastid and nuclear regions as DNA barcodes in Passiflora. This genus is a largely variable, with more than 900 species of high ecological, commercial, and ornamental importance. We analyzed 1034 accessions of 222 species representing the four subgenera of Passiflora and evaluated the effectiveness of five plastid regions and three nuclear datasets currently employed as DNA barcodes in plants using barcoding gap, applied similarity-, and tree-based methods. The plastid regions were able to identify less than 45% of species, whereas the nuclear datasets were efficient for more than 50% using “best match” and “best close match” methods of TaxonDNA software. All subgenera presented higher interspecific pairwise distances and did not fully overlap with the intraspecific distance, and similarity-based methods showed better results than tree-based methods. The nuclear ribosomal internal transcribed spacer 1 (ITS1) region presented a higher discrimination power than the other datasets and also showed other desirable characteristics as a DNA barcode for this genus. Therefore, we suggest that this region should be used as a starting point to identify Passiflora species.  相似文献

Solution crystallization analysis by laser light scattering (SCALLS) involves the observation of the scattering of diode mercury laser lamp light after it passes through a polymer solution. An increase in turbidity occurs when the hot polymer solution is cooled and the polymer starts to crystallize out of solution. This causes a decrease in the amount of laser light that can pass through the solution and an increase in the amount of scattered light. The reverse of this process leads to the turbidity decreasing with an increase in temperature. According to this concept, it is possible to follow the solution crystallization of various polypropylenes under controlled cooling. In this study, SCALLS was able to differentiate between different isotactic and syndiotactic polypropylenes with similar chemical structures, but different tacticity and molecular weights. Furthermore, SCALLS provided good crystallization information that is similar to that from crystallization analysis fractionation and temperature rising elution fractionation. In addition, SCALLS can be used as a quantitative tool for the measurement of weight fractions during dissolution. © 2014 Society of Chemical Industry  相似文献

This work aimed to develop an orange juice powder by spray drying with lactic acid bacteria (Lactobacillus plantarum 299v and Pediococcus acidilactici HA‐6111‐2), testing their survival both during drying and storage (room temperature and 4 °C). Initially, the best conditions for spray drying were chosen to allow the best survival of each LAB: (i) inlet air temperature of 120 °C and (ii) 0.5:2 ratio of the orange juice soluble solids and drying agent added (prebiotics: 10 DE maltodextrin or gum Arabic). Survival of LAB was not affected by drying process, and it was higher when cultures were stored at 4 °C. A slightly higher protection was conferred by 10 DE maltodextrin, in the case of L. plantarum and at 4 °C. Pediococcus acidilactici was more resistant during storage at 4 °C, with logarithmic reductions lower than 1 log‐unit. It was demonstrated that it is possible to produce a functional nondairy product, orange juice powder supplemented with prebiotic compounds, containing viable LAB for at least 7 months, when stored at 4 °C.  相似文献

Tropical ecosystems are particularly rich in edible plant species with different bioactive substances. Among the plants with promising benefits for health are species from the genus Dioscorea (Dioscoreaceae), especially those named yam. Recent studies have shown the beneficial effects of different species of Dioscorea, and its main constituent, diosgenine, in the treatment of food allergy. In this study we evaluated the potential of D. trifida, the only yam native from South America, in the treatment of ovalbumin (OVA) induced food allergy in Balb/c mice. HPLC/DAD analysis showed the presence of three very distinctive groups of natural products in extracts and fractions: (I) very polar substances, including allantoin, (II) phenolic substances as flavonoids and phenolic acids and (III) diosgenin and derivatives. Sensitive mice received casein feed with supplementation of crude extract (CE) and fractions. The supplementation with all products from D. trifida reduced IGE, intestinal oedema and mucus production, parameters observed in OVA allergic mice. The results showed the potential of this food to prevent or treat this disease and the necessity to be better explored.  相似文献

Integrated permanent magnet microactuators have been fabricated using micromachined polymer magnets. The hard magnetic material utilized is a polymer composite, consisting of magnetically hard ceramic ferrite powder imbedded in a commercial epoxy resin to a volume loading of 80%. The magnets have the form of thin disks approximately 4 mm in diameter and 90 μm in thickness. These disks have been magnetized in the thickness direction, and even in this geometrically unfavorable direction showed typical permanent magnet behavior with an intrinsic coercivity Hci of 4000 Oe (320 kA/m) and a residual induction Br of 600 Gauss (60 mT). Cantilever beam-type magnetic actuators carrying a screen-printed disk magnet on their free ends have been fabricated on an epoxy board. A planar coil on the opposite side of the substrate is used to drive the beams vertically. The actuators exhibit hard magnetic behavior allowing both attraction and repulsion by reversing the current direction. Static and dynamic testing of the magnetic actuators have been performed. The experimental data are compared with theoretical results obtained from both finite element simulations and analytical models. Good agreement is obtained between simulation and experiment  相似文献
