Antimicrobial resistance represents a major threat to human health and knowledge of the underlying mechanisms is therefore vital. Here, we report the discovery and characterization of oxidoreductases that inactivate the broad-spectrum antibiotic chloramphenicol via dual oxidation of the C3-hydroxyl group. Accordingly, chloramphenicol oxidation either depends on standalone glucose-methanol-choline (GMC)-type flavoenzymes, or on additional aldehyde dehydrogenases that boost overall turnover. These enzymes also enable the inactivation of the chloramphenicol analogues thiamphenicol and azidamfenicol, but not of the C3-fluorinated florfenicol. Notably, distinct isofunctional enzymes can be found in Gram-positive (e. g., Streptomyces sp.) and Gram-negative (e. g., Sphingobium sp.) bacteria, which presumably evolved their selectivity for chloramphenicol independently based on phylogenetic analyses. Mechanistic and structural studies provide further insights into the catalytic mechanisms of these biotechnologically interesting enzymes, which, in sum, are both a curse and a blessing by contributing to the spread of antibiotic resistance as well as to the bioremediation of chloramphenicol.  相似文献

Thioredoxin reductase (TrxR) is overexpressed in cancer cells and is therefore a putative cancer target. Inhibition of this enzyme is considered an important strategy for the development of new chemotherapeutic agents with a specific mechanism of action. Organotin compounds have been described as experimental antitumor agents, yet their mechanism of action remains largely unknown. Based on the outcome of a virtual screening study, various di‐ and tri‐n‐butyltin(IV) carboxylates were synthesized, and their biological properties were evaluated. All synthesized compounds were able to inhibit TrxR selectively within the micromolar range and showed potent antitumor activity against HT‐29 and MCF‐7 cancer cell lines. Moreover, tin(IV) organometallics were found to strongly induce apoptosis in the BJAB lymphoma cell line. Mass spectrometry and atomic absorption spectroscopy experiments revealed metal binding to proteins, and efficient cellular uptake was observed using a di‐n‐butyltin(IV) complex as an example.  相似文献

Crosslinked particles containing butyl acrylate, methyl methacrylate, and allyl methacrylate are prepared by free‐radical emulsion polymerization. The glass transition temperatures of the polymers are varied by the crosslinking densities in the latex particles. Aqueous acrylate dispersions with polymers of different glass transition temperatures are electrospun with PVA as the matrix polymer. The effects of crosslinking density and Tg on the structure and mechanical properties of the fibers are studied. Crosslinking of unreacted allyl groups is induced by UV irradiation to stabilize the fibers by interparticle crosslinking. Both the ability to undergo interparticle crosslinking and the E modulus depend on the merging of the particles during the electrospinning process.

A simulation study of heterogeneously catalyzed reactive distillation experiments carried out with the D + R tray, a novel type of laboratory equipment, is presented. One advantage of the D + R tray is that reaction and distillation are alternating stage‐wise, in a well‐defined way that can be modeled straightforwardly. An equilibrium stage model is used to describe the distillation and a plug flow reactor model to describe the catalyst bed reactors. The model parameters are derived from a systematic experimental characterization of the D + R tray both as a reactor and as a distillation unit. A validated physicochemical fluid property model is used. The primary experimental data are reconciled. Results from the predictive simulations are in good agreement with the experimental results. With the aim of establishing a versatile and easy synthesis of branched saccharides for biological applications, we used molecular-dynamics simulations to model Lewis(y) to two classes of di- or triantennary saccharide mimetics. One set of mimetics was based on 1,3,5-tris(hydroxymethyl)cyclohexane (TMC) as the core, the other on furan, and both were derivatised with galactose and/or fucose. The TMC-based saccharides were biotinylated, while the furan disaccharides were treated with maleimide-activated biotin in a Diels-Alder fashion to yield oxazatricyclodecanes (OTDs). These were then assayed as cell-surface labels in human colon (SW480 and CaCo-2), liver (PLC), Glia (U333 CG 343) and ovary (SKOV-3) tumour cell lines. Discrete staining patterns were observed in all cells, usually at one or two poles of the cells, particularly with the asymmetric 3-beta-L-fucopyranosyloxymethyl-4-beta-D-galactopyranosyloxymethyl-OTD. Normal SV40-transformed fibroblasts (SV80) showed no staining. Adhesion of the highly metastatic mouse melanoma line B16 F10 to fibronectin was inhibited by 80 % by the TMC-digalactoside and by 30 % by 3,4-bis-(beta-D-galactopyranosyloxymethyl)furan. None of the saccharide mimetics inhibited the adhesion of the less metastatic B16 F1 line. Migration of B16 F10 cells through Matrigel was greatly inhibited by the TMC-digalactoside and weakly inhibited by the TMC-trigalactoside. The saccharide mimetics that had shown the best structural agreements with the terminal saccharides of Lewis(y) in the molecular dynamics simulation were also the most biologically potent compounds; this underlines the predictive nature of molecular dynamics simulations. The use of the non-saccharide cores enabled us to adapt spacer lengths and terminal saccharides to optimise the structures to bind more avidly to cell-surface lectins.  相似文献

Grain growth kinetics of mullite in laboratory-produced and commercial (3M-Nextel 720) alumino silicate fibers was analyzed in the temperature range between 1500° and 1700°C. The lab fibers consist of mullite plus traces of α-alumina, while the phase content of the commercial Nextel 720 fibers is about 60 wt% mullite plus 40 wt%α-alumina. The temperature-induced grain coarsening of mullite follows the empirical law D 1/ n − D 01/ n = kt . Two different temperature regimes can be distinguished with respect to the grain growth exponents: above 1600°C the growth exponent is ≈1/3, while below 1600°C the growth exponent of ≈1/12 is exceptionally low. Laboratory-produced and commercial fibers show almost identical mullite grain growth kinetics.  相似文献

The selectivity of partial hydrogenation reactions of unsaturated substrates was studied in a membrane reactor operating at 323 K and 40 bar hydrogen pressure. The reactor system was constructed as a loop of a saturation vessel and a membrane module in which the reaction mixture was resaturated with hydrogen up to 100 times. In a porous membrane made from cross-linked polyacrylic acid palladium nanoparticles were incorporated as catalysts. A well-defined residence time within the membrane was achieved due to a defined pore structure of the membrane and a convective mass flow of the reaction mixture through the membrane. The selectivity for the partially hydrogenated products was investigated as a function of the pore size of the PAA membrane and was compared to commercially available catalysts. Compared to experiments with supported catalysts (Pd/C and Pd/Al2O3) in a slurry and a fixed bed reactor the selectivity for the desired products could be increased by 3% (1-octyne) up to 40% (geraniol).  相似文献

Silicon-substituted hydroxyapatite (Si-HA) with up to 1.8 wt% Si content was prepared successfully by a hydrothermal method, using Ca(NO3)2, (NH4)3PO4 or (NH4)2HPO4 and Si(OCH2CH3)4 (TEOS) as starting materials. Silicon has been incorporated in hydroxyapatite (HA) lattice by partially replacing phosphate (PO43−) groups with silicate (SiO44−) groups resulting in Si-HA described as Ca10(PO4)6−x(SiO4)x(OH)2−x. X-ray diffraction (XRD), Fourier transform IR spectroscopy (FTIR), inductively coupled plasma AES (ICP-AES) and scanning electron microscopy (SEM) techniques reveal that the substitution of phosphate groups by silicate groups causes some OH loss to maintain the charge balance and changes the lattice parameters of HA. The crystal shape of Si-HA has not altered compared to silicon-free reference hydroxyapatite but Si-incorporation reduces the size of Si-HA crystallites. Based on in vitro tests, soaking the specimens in simulated body fluid (SBF), and MTT assays by human osteoblast-like cells, Si-substituted hydroxyapatite is more bioactive than pure hydroxyapatite.  相似文献
