There is increased consumer awareness that foods contain microcomponents that may have beneficial effects on health maintenance and disease prevention. In milk fat these functional food components include EPA, DHA, and CLA. The opportunity to enhance the content of these FA in milk has improved as a result of recent advances that have better defined the interrelationships between rumen fermentation, lipid metabolism, and milk fat synthesis. Dietary lipids undergo extensive hydrolysis and biohydrogenation in the rumen. Milk fat is predominantly TG, and de novo FA synthesis and the uptake of circulating FA contribute nearly equal amounts (molar basis) to the FA in milk fat. Transfer of dietary EPA and DHA to milk fat is very low (<4%); this is, to a large extent, related to their extensive biohydrogenation in the rumen, and also partly due to the fact that they are not transported in the plasma lipid fractions that serve as major mammary sources of FA uptake (TG and nonesterified FA). Milk contains over 20 isomers of CLA but the predominant one is cis-9,trans-11 (75–90% of total CLA). Biomedical studies with animal models have shown that this isomer has anticarcinogenic and anti-atherogenic activities. cis-9,trans-11-CLA is produced as an intermediate in the rumen biohydrogenation of linoleic acid but not of linolenic acid. However, it is only a transient intermediate, and the major source of milk fat CLA is from endogenous synthesis. Vaccenic acid, produced as a rumen biohydrogenation intermediate from both linoleic acid and linolenic acid, is the substrate, and Δ9-desaturase in the mammary gland and other tissues catalyzes the reaction. Diet can markedly affect milk fat CLA content, and there are also substantial differences among individual cows. Thus, strategies to enhance milk fat CLA involve increasing rumen outflow of vaccenic acid and increasing Δ9-desaturase activity, and through these, several-fold increases in the content of CLA in milk fat can be routinely achieved. Overall, concentrations of CLA, and to a lesser extent EPA and DHA, can be significantly enhanced through the use of diet formulation and nutritional management of dairy cows.  相似文献

Graphene oxide (GO) was controllably reduced using electron beam generated plasmas produced in methane/argon mixtures. Operating parameters including, total pressure, duty factor (the ratio of the pulse duration to the pulse period), treatment time, and methane flow rate were varied to determine their impact on the reduction process. The resulting surface chemistry, topography and crystallinity of the reduced graphene oxide (r-GO) was determined with X-ray photoelectron spectroscopy (XPS), atomic force microscopy (AFM) and Raman spectroscopy. Oxygen concentration could be tuned to values between 43 at.% and 5 at.%, and under certain operating conditions the sp2 cluster size grows. System diagnostics identified the critical operating parameters for the reduction of oxygen in GO.  相似文献

In previous studies, high plasma insulin was associated with earlier resumption of postpartum estrous cycles in dairy cows. The objective of this experiment was to quantify hormonal and ovarian responses to dietary starch and fat contents. Thirty cows were fed on a standard diet from calving until 40 d in milk (DIM) and then 6 cows were allocated to each of 5 isoenergetic diets containing 231, 183, 159, 135, and 87 g of starch and 39, 42, 43, 45, and 48 g of fat/kg of dry matter (DM) for diets 1 to 5, respectively, until 70 DIM. Estrus was synchronized at 60 DIM. Between 60 and 70 DIM, energy intake, milk yield, and energy balance were similar among diet groups. Plasma insulin-to-glucagon ratio increased with increasing dietary starch and decreasing dietary fat concentrations, reaching a break point at 159 g of starch, 43 g of fat/kg of DM (diets 1 to 5: mean 3.86, 3.78, 3.59, 2.98, 2.06 ± standard error 0.22). Growth hormone, insulin-like growth factor-I, and leptin did not vary among diets. The greatest dietary starch concentration was associated with elevated plasma urea-N (diets 1 to 5: mean 3.69, 3.01, 2.94, 2.95, 2.75, ± standard error 0.13 mmol/L, respectively) and delayed postovulatory progesterone increase (progesterone at 3 to 5 d postovulation for diets 1 to 5: mean 2.7, 5.9, 4.2, 5.6, 4.3 ± standard error 0.9 ng/mL, respectively). The number of small (<5 mm) ovarian follicles was positively related to starch intake (r = 0.381) and plasma insulin concentration (r = 0.402). It is concluded that to maintain adequate insulin-to-glucagon ratio in cows at the start of the breeding period, dietary starch concentration should be above 160 g/kg of DM and dietary fat below 44 g/kg of DM, and this should have a positive effect on ovarian function.  相似文献

We use the semiclassical limit of electromagnetic wave scattering theory to determine the properties of the exterior caustics of a diagonally incident plane wave scattered by an infinitely long homogeneous dielectric circular cylinder in both the near zone and the far zone. The transmission caustic has an exterior/interior cusp transition as the tilt angle of the incident beam is increased, and each of the rainbow caustics has a farzone rainbow/exterior cusp transition and an exterior/interior cusp transition as the incident beam tilt angle is increased. We experimentally observe and analyze both transitions of the first-order rainbow. We also compare the predictions of the semiclassical approximation with those of ray theory and exact electromagnetic wave scattering theory.  相似文献

The utility of the one-beam cross-correlation dynamic light-scattering system for sizing small particles in suspension was previously limited by its small-intensity signal-to-baseline ratio for strongly turbid suspensions. We describe three improvements in the optical system and sample cell that raise the ratio to a value comparable with that of other cross-correlation dynamic light-scattering systems. These improvements are (i) using a square cross-sectional sample cell to minimize the attenuation of the incident beam and singly scattered light, (ii) placing a 200-microm-wide slit between the sample cell and the detector fibers to mask off the region of weak single scattering and strong multiple scattering from the detectors' field of view, and (iii) aligning the center of the detectors' field of view with the region of strongest single scattering. We analyze a number of suspensions of polystyrene latex spheres with a diameter between 65 and 562 nm in water using this improved one-beam instrument and find that the measured radius is determined in a 2-min data collection time to better than +/-10% for volume fractions of the suspended polystyrene latex spheres up to a few percent.  相似文献

An Escherichia coli O157:H7 subtyping method based on PCR amplification of variable DNA sequences between the repetitive element IS3 was developed. Template DNA was prepared by boiling cells in Chelex. Two separate IS3 PCR amplifications were performed for each isolate: one with a single primer (primer IS3A) and one with two primers (primers IS3A and IS3B). The IS3 PCR subtyping method was applied to 35 epidemiologically related and unrelated E. coli O157:H7 isolates that had been previously characterized by pulsed-field gel electrophoresis (PFGE). PFGE identified 25 different subtypes (difference of one or more bands). PCR with single primer IS3A and primer pair IS3A-IS3B identified 6 and 14 different subtypes, respectively. By combining the results of the two PCR amplifications, 15 different IS3 PCR subtypes were identified. While not as sensitive as PFGE, IS3 PCR subtyping grouped all outbreak-related isolates. IS3 PCR banding patterns were reproducible between amplifications and between subcultures. IS3 PCR could serve as a simple, rapid screening method for the identification of unrelated E. coli O157:H7 isolates.  相似文献

OBJECTIVE: To report on a rare case of primary leiomyosarcoma of the adrenal. To our knowledge, this is the fourth case reported in the literature. METHODS: A patient with primary leiomyosarcoma of the adrenal gland is presented. The clinical features are described, and the diagnostic and therapeutic aspects of this rare primary mesenchymal tumor are discussed. RESULTS/CONCLUSIONS: The aggressive nature and the poor prognosis of this rare tumor type are emphasized.  相似文献
