Inducing malolactic fermentation in Chardonnay musts and wines using different strains of Oenococcus oeni

Malolactic fermentations (MLF) were induced in a commercially prepared Washington State Chardonnay must to evaluate the influence of timing of inoculation and pre-culture conditions of Oenococcus oeni strains MCW, EQ-54, and WS-8. The must (pH 3.62, 21.5°Brix) was divided into lots and inoculated with Saccharomyces cerevisiae strain CY3079. Strains of O. oeni were pre-cultured by growing in diluted juice or by re-hydration of freeze-dried strains. Bacteria were inoculated into the musts before (Day 0) or after completion of the alcoholic fermentation (Day 22). Yeast populations exceeded 10⁷cfu/mL in all fermentations that proceeded to dryness. However, the viability of most strains of O. oeni quickly declined after inoculation regardless of the timing of inoculation or the strain used. MLF was induced in the wines inoculated with strains EQ-54 and WS-8 but not with MCW, and the rate depended on the time of inoculation. The method used to prepare bacterial starter cultures had no apparent influence on the completion of MLF. Values for volatile acidity were slightly higher (P< 0.05) in wines inoculated with O. oeni before alcoholic fermentation compared with those inoculated after alcoholic fermentation.

Synthesis and characterization of electrochemically active graphite–silicon–tin composite anodes for Li-ion applications

Electrochemically active Si_0.66Sn_0.34 (SiSn) composite alloys dispersed in a carbon (graphite) matrix were synthesized using both wet and dry high-energy mechanical milling (HEMM). The resultant composites are comprised of amorphous carbon (in the case of dry HEMM) or crystalline carbon (in the case of wet HEMM), and crystalline silicon and tin (for both cases) as verified by X-ray diffraction (XRD). The XRD results also indicate the presence of iron–tin intermetallic (FeSn₂) arising as a contaminant during dry HEMM. The composite composition of 85C–15[Si_0.66Sn_0.34] (mol%) resulted in reversible discharge capacities as high as 800 mAh g⁻¹ with a reasonable capacity retention (1.36% loss/cycle). Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) analyses were further conducted to examine the surface of the electrode and to determine the presence/absence of organic species resulting from reactions between the electrode, lithium ions and electrolyte, respectively.     

A Ligand-Directed Nitrophenol Carbonate for Transient in situ Bioconjugation and Drug Delivery

Here we report the first use of ligand-directed proximity accelerated bioconjugation chemistry in the tandem delivery and release of a therapeutic payload. To do this, we designed a nitrophenol carbonate for ligand-directed in situ bioconjugation of a prodrug payload to a protein. The transient nature of our conjugation chemistry renders the protein a depot for time-dependent release of active drug following hydrolysis and self-immolation. In our model system, using an immunostimulant prodrug, biotin ligand, and avidin protein, we observe release of bioavailable immunostimulant both spectroscopically and with an immune cell line over 48 h. Avidin co-crystalized with the nitrophenolate directing group verified the binding pose of the ligand and offered insight into the mechanism of in situ bioconjugation. Overall, this scaffold warrants further investigation for the time-dependent delivery of therapeutics and use in protein ligand pairs beyond biotin and avidin used for this work.
