The comparison of starch granule deformation and extent of starch gelatinization in a few common Indian unleavened flat breads (Chapati, Parontha and Poorie) was made. The starch granule swelling was evaluated by scanning electron as well as polarized light microscopy. The peak viscosity in Haake rotary viscosimeter and hydration capacity were used for comparing the extent of starch gelatinization in crust and crumb of these baked products. Although a few of the starch granules on the outside of crust, maintained a weak outline those in the crumb area gave only a matrix of gelatinized starch. The extent of starch gelatinization was reflected by the related increase in hydration capacity of these products. However, the peak viscosity values were found to be inversely related to the extent of starch gelatinization. As compared to crumb, a higher extent of browning was observed in the crust. Water absorption, type of cereal, mode and efficiency of heat transfer were found to control the extent of starch gelatinization in these baked products.  相似文献

Silver nanoparticles (AgNPs) are the most commercialized nanomaterials and presumed to be biocompatible based on the biological effects of the bulk material. However, their physico-chemical properties differ significantly to the bulk materials and are associated with unique biological properties. The study investigated the antimicrobial and cytotoxicity effects of AgNPs synthesized using gum arabic (GA), sodium borohydride (NaBH4), and their combination as reducing agents. The AgNPs were characterized using ultraviolet-visible spectrophotometry (UV-Vis), dynamic light scattering (DLS), transmission electron microscopy (TEM), and Fourier-transform infrared spectroscopy (FT-IR). The anti-bacterial activity was assessed using agar well diffusion and microdilution assays, and the cytotoxicity effects on Caco-2, HT-29 and KMST-6 cells using MTT assay. The GA-synthesized AgNPs (GA-AgNPs) demonstrated higher bactericidal activity against all bacteria, and non-selective cytotoxicity towards normal and cancer cells. AgNPs reduced by NaBH4 (C-AgNPs) and the combination of GA and NaBH4 (GAC-AgNPs) had insignificant anti-bacterial activity and cytotoxicity at ≥50 µg/mL. The study showed that despite the notion that AgNPs are safe and biocompatible, their toxicity cannot be overruled and that their toxicity can be channeled by using biocompatible polymers, thereby providing a therapeutic window at concentrations that are least harmful to mammalian cells but toxic to bacteria.  相似文献

Mineral fillers of flake configuration such as mica can, in principle, produce large increases in modulus and strength especially if the flake is oriented predominantly in a plane. Additionally, the resulting composites have the advantage of possessing isotropic properties in the plane of orientation, thereby minimizing the warpage normally associated with the injection molding of fiber-reinforced thermoplastics. However, to properly transmit stress from the matrix to the mica and minimize the effect of defects in morphology, a ductile matrix is required with adequate interaction or adhesion between phases. Many of the above conclusions are documented in the literature and are confirmed by our investigations of the system mica-polypropylene. The novelty of this effort rests upon (1) successful attempts to identify specific non-silane additives which by co-blending suitably modify the mica-polypropylene interface, (2) to demonstrate the remarkable effects of time-temperature during melt processing on the behavior of this system; and, (3) to show the relation between composition and time-temperature effects on resultant mechanical, physical, and thermal properties. Most notably, large improvements in tensile and flexural strength and heat distortion temperature can be attributed to the use of small amounts of chlorinated organic compounds blended under carefully selected melt process conditions. Some speculation concerning the mechanism of interaction will be discussed and the resultant potential for increased application will also be outlined.  相似文献

NMR flow devices provide longitudinal real-time quantitative metabolome characterisation of living cells. However, discrimination of intra- and extracellular contributions to the spectra represents a major challenge in metabolomic NMR studies. The present NMR study demonstrates the possibility to quantitatively measure both metabolic intracellular fingerprints and extracellular footprints on human control fibroblasts by using a commercially available flow tube system with a standard 5 mm NMR probe. We performed a comprehensive 3D cell culture system characterisation. Diffusion NMR was employed for intra- and extracellular metabolites separation. In addition, complementary extracellular footprints were determined. The implemented perfused NMR bioreactor system allowed the determination of 35 metabolites and intra- and extracellular separation of 19 metabolites based on diffusion rate differences. We show the reliability and sensitivity of NMR diffusion measurements to detect metabolite concentration changes in both intra- and extracellular compartments during perfusion with different selective culture media, and upon complex I inhibition with rotenone. We also demonstrate the sensitivity of extracellular footprints to determine metabolic variations at different flow rates. The current method is of potential use for the metabolomic characterisation of defect fibroblasts and for improving physiological comprehension.  相似文献

The VLDL receptor has been described as a new member of the LDL receptor supergene family that specifically binds VLDL in vitro via apolipoprotein E and lipoprotein lipase. Both apolipoprotein E and lipoprotein lipase are constituents of chylomicron remnants, another triglyceride-rich lipoprotein which has been proposed as a physiological ligand for the VLDL receptor. We used human chylomicron remnants to study their uptake into LDL, receptor-deficient Chinese hamster ovary cells overexpressing the human VLDL receptor. The uptake into these cells was compared to that into cells transfected with an empty transfection vector. Human chylomicron remnants were produced in vitro by hydrolysis with lipoprotein lipase, and were labeled with 125I. The uptake of these remnants into the cells overexpressing the VLDL receptor was found to be about 3-fold higher than the uptake into the control cells. The addition of a surplus of either apolipoprotein E or inactivated lipoprotein lipase to the remnants led to an increase in particle uptake. The chylomicron remnant uptake was inhibited by addition of the 39 kDa receptor associated protein These in vitro experiments strongly support the idea that the VLDL receptor is a physiological receptor for chylomicron remnants. The increase of receptor-mediated uptake induced by the addition of apoE or lipoprotein lipase underlines the role of these two proteins in this process.  相似文献

The structure of the metalloproteinase and hemorrhagic toxin atrolysin C form d (EC 3.4.24.42), from the venom of the western diamondback rattlesnake Crotalus atrox, has been determined to atomic resolution by x-ray crystallographic methods. This study illuminates the nature of inhibitor binding with natural (< Glu-Asn-Trp, where < Glu is pyroglutamic acid) and synthetic (SCH 47890) ligands. The primary specificity pocket is exceptionally deep; the nature of inhibitor and productive substrate binding is discussed. Insights gained from the study of these complexes facilitate the design of potential drugs to treat diseases where matrix metalloproteinases have been implicated, e.g., arthritis and tumor metastasis.  相似文献

By using the D(d, p)3H nuclear reaction combined with the Rutherford-backscattering method, the amount of D2O in relation to H2O in water from the Dead Sea is measured. The amount of deuterium in the samples is determined from the integrated yield of the D(d, p)3H reaction in relation to the 16O(d, p0)17O or 16O(d, p1)17O reaction. For the low deuteron energies used (E 0.63 MeV), the differential cross section of the 16O(d, p0)17O reaction was determined experimentally. No significant change of deuterium concentration in Dead-Sea water down to a depth of 36 m could be detected.  相似文献
