Effect of cattle manure on selenate and selenite retention in soil   总被引:2,自引:0,他引:2  
The aim of the present investigation was to study the effect of manure on retention of selenium (Se) in soil. Addition of cattle manure in combination with selenite and selenate reduced the adsorption of both anions to a loam soil in a batch experiment. The results were explained by the content of low-molecular-weight organic acids in the manure which compete with Se for the sorption sites. In a pot experiment with loam and peat soils and with two pH levels within each soil, cattle slurry added together with selenate was found to increase the Se concentration in grain at the highest pH level (6.1 and 6.8 for the loam and peat, respectively). At a lower pH (5.4 and 6.0 for the loam and peat, respectively) there was no significant effect of slurry on Se concentration in grain. Application of slurry also increased the residual effect of Se applied to the loam soil in the preceding growing season. In the peat soil, no residual effect of Se was found either with or without the addition of slurry.  相似文献

Interleukin-6 (IL-6) is one of the pathogenetic elements in inflammatory and age-related diseases such as rheumatoid arthritis, osteoporosis, atherosclerosis, and late-onset B cell neoplasia. In these diseases or during aging, the decrease in production of sex hormones such as dehydroepiandrosterone (DHEA) is thought to play an important role in IL-6-mediated pathogenetic effects in mice. In humans, we investigated the correlation of serum levels of DHEA, DHEA sulfate (DHEAS), or androstenedione (ASD) and IL-6, tumor necrosis factor-alpha, or IL-2 with age in 120 female and male healthy subjects (15-75 yr of age). Serum DHEA, DHEAS, and ASD levels significantly decreased with age (all P < 0.001), whereas serum IL-6 levels significantly increased with age (P < 0.001). DHEA/DHEAS and IL-6 (but not tumor necrosis factor-alpha or IL-2) were inversely correlated (all patients: r = -0.242/-0.312; P = 0.010/0.001). In female and male subjects, DHEA and ASD concentration dependently inhibited IL-6 production from peripheral blood mononuclear cells (P = 0.001). The concentration-response curve for DHEA was U shaped (maximal effective concentration, 1-5 x 10(-8) mol/L), which may be the optimal range for immunomodulation. In summary, the data indicate a functional link between DHEA or ASD and IL-6. It is concluded that the increase in IL-6 production during the process of aging might be due to diminished DHEA and ASD secretion. Immunosenescence may be directly related to endocrinosenescence, which, in turn, may be a significant cofactor for the manifestation of inflammatory and age-related diseases.  相似文献

In previous research, rats exposed to daily, 3 h sessions of schedule-induced polydipsia (SIP) self-administered high doses of cocaine orally. However, a strong and durable preference for cocaine solution to water requires training in addition to mere oral self-administration exposure. If cocaine is dissolved in a preferred vehicle solution, and the vehicle is subsequently faded to water, then a strong preference for cocaine remains. A similar preference can be instituted for lidocaine solution. Such preferences may develop because the gustatory property of a drug becomes associated with the preferred vehicle and remains to function as a durable conditioned reinforcer after vehicle fading. To determine if drug preference is solely a function of this posited conditioning mechanism, or whether it also depends upon the SIP condition, rats were exposed to daily, 3 h sessions of single-ration feeding, rather than the SIP condition. A preferred vehicle (glucose/saccharin solution) was slowly faded from a 0.19 mg/ml lidocaine solution, which was presented concurrently with a choice for water. Although a preference for lidocaine solution to water could be generated, it occurred for only 5 out of 9 rats, and the preference was relatively unstable. By contrast, in two previous studies using SIP, 26 out of 27 rats maintained a preference for lidocaine solution. Thus, SIP not only exaggerates the amount of drug solution ingested but also contributes to the fixation of the associative drug solution choice.  相似文献

We are developing a system to control G protein signaling in vivo to regulate a broad range of physiologic responses. Our system utilizes G protein-coupled peptide receptors engineered to respond exclusively to synthetic small molecule ligands and not to their natural ligand(s). These engineered receptors are designated RASSLs (receptor activated solely by a synthetic ligand). We have made two prototype RASSLs that are based on the human kappa opioid receptor. Small molecule drugs that activate the kappa receptor are nonaddictive and safe to administer in vivo. Binding and signaling assays reveal 200-2000-fold reductions in the ability of our RASSLs to bind or be activated by dynorphin, an endogenous peptide ligand of the kappa opioid receptor. In a high-throughput signaling assay, these prototype RASSLs expressed in Chinese hamster ovary K1 cells showed little or no response to a panel of 21 opioid peptides but still signaled normally in response to small molecule drugs such as spiradoline. Activation of a RASSL by spiradoline also caused proliferation of rat-1a tissue culture cells. These data provide evidence that G protein-coupled receptors can be made into RASSLs. The potential in vivo applications for RASSLs include the positive enrichment of transfected cells and the development of new animal models of disease.  相似文献

The Engrailed-1 gene, En1, a murine homologue of the Drosophila homeobox gene engrailed (en), is required for midbrain and cerebellum development and dorsal/ventral patterning of the limbs. In Drosophila, en is involved in regulating a number of key patterning processes including segmentation of the epidermis. An important question is whether, during evolution, the biochemical properties of En proteins have been conserved, revealing a common underlying molecular mechanism to their diverse developmental activities. To address this question, we have replaced the coding sequences of En1 with Drosophila en. Mice expressing Drosophila en in place of En1 have a near complete rescue of the lethal En1 mutant brain defect and most skeletal abnormalities. In contrast, expression of Drosophila en in the embryonic limbs of En1 mutants does not lead to repression of Wnt7a in the embryonic ventral ectoderm or full rescue of the embryonic dorsal/ventral patterning defects. Furthermore, neither En2 nor en rescue the postnatal limb abnormalities that develop in rare En1 null mutants that survive. These studies demonstrate that the biochemical activity utilized in mouse to mediate brain development has been retained by Engrailed proteins across the phyla, and indicate that during evolution vertebrate En proteins have acquired two unique functions during embryonic and postnatal limb development and that only En1 can function postnatally.  相似文献

We arrange various types of probabilistic transition systems studied in the literature in an expressiveness hierarchy. The expressiveness criterion is the existence of an embedding of systems of the one class into those of the other. An embedding here is a system transformation which preserves and reflects bisimilarity. To facilitate the task, we define the classes of systems and the corresponding notion of bisimilarity coalgebraically and use the new technical result that an embedding arises from a natural transformation with injective components between the two coalgebra functors under consideration. Moreover, we argue that coalgebraic bisimilarity, on which we base our results, coincides with the concrete notions proposed in the literature for the different system classes, exemplified by a detailed proof for the case of general Segala-type systems.  相似文献

Examines the literature on drug dependence from a behavioral viewpoint. Dependence on drugs and alcohol results from an interaction between the individual and the substance within precipitating environmental conditions. Animals and humans self-administer the same drugs, suggesting the biological normality of chemical dependency and the value of laboratory research on the generation and alleviation of such dependencies. Functional or behavioral toxicity can be a serious social problem, and research is needed on why many individuals do not become addicted to drugs and alcohol to which they are exposed, and how social processes can be used to increase an individual's resistance to substance abuse. Zusammenfassung Eine Methode zur quantitativen Bestimmung von Kaffeesäuremethylester und Kaffeesäureäthylester wurde ausgearbeitet: Nach Extraktion und säulenchromatographischer Reinigung an Polyamid wurden die Ester mittels Hochdruckflüssigkeitschromatographie an Nucleosil 10 C8 isokratisch getrennt. Zur Identifizierung und zur quantitativen Bestimmung dieser Verbindungen wurde ein elektrochemischer Detektor verwendet. Die Nachweisgrenzeliegt für Kaffeesäuremethylester bei 0,02 ppm, für Kaffeesäureäthylester bei 0,025 ppm. Die Identifizierungerfolgte zusätzlich über ein Quadrupol GC-MS-System (SIM-Verfahren).
Determination of methyl caffeate und ethyl caffeate in vegetables by high pressure liquid chromatography with electrochemical detection
Summary A method for quantitative determination of methyl caffeate and ethyl caffeate was developed: After extraction and preliminary purification on polyamide columns the extracts were analysed by high pressure liquid chromatography, employing isocratic elution from Nucleosil 10 C8. For identification and quantitative determination an electrochemical detector was used. The detection limit of methyl caffeate is 0.02 ppm and for ethyl caffeate it is 0.025 ppm. Selected ion monitoring mass spectrometry was applied for confirmatory identification of these compounds.
