Acquisition of odor-guided or visually-guided delayed win-shift behavior was evaluated in rats after lidocaine-induced inactivation within the agranular insular area of the prefrontal cortex (PFC) or the prelimbic area of the PFC. Additional sites and tasks were used to control for neuroanatomical and behavioral specificity of lidocaine inactivation of the agranular insular and prelimbic areas. Results showed that acquisition of the odor-guided delayed win-shift task was dependent on the agranular insular area, whereas acquisition of the visually-guided version was dependent on the prelimbic area. This dissociation suggests that the stimulus modality used is critical for revealing working memory functions of different PFC subregions. Data are reported from 3,213 research eyewitnesses confirming that accurate eyewitness identifications from lineups are made faster than are inaccurate identifications. However, consistent with predictions from the recognition and search literatures, the authors did not find support for the "10-12-s rule" in which lineup identifications faster than 10-12 s maximally discriminate between accurate and inaccurate identifications (D. Dunning & S. Perretta, 2002). Instead, the time frame that proved most discriminating was highly variable across experiments, ranging from 5 s to 29 s, and the maximally discriminating time was often unimpressive in its ability to sort accurate from inaccurate identifications. Many glomerulopathies are characterized by progressive mesangial (interstitial) expansion which can be quantitated by morphometric analysis. The purpose of this study was to analyze mesangial and glomerular volumes using a new computer-assisted reconstruction (CAR) method. CAR was compared to two standard planar methods, point-counting and linear integration, for accuracy and time efficiency. In Phase I of the study, a computer-based model of the mesangial space was created by placing spherical and ellipsoidal objects of known volume into an enclosing volume mimicking the glomerulus. The simulated mesangium occupied approximately 10 percent of the glomerular volume. The model glomerulus was sectioned serially into ten sections of equal thickness and the three morphometric methods applied to determine the mesangial/glomerular volume. The complexity of the mesangial model was varied by increasing the number of mesangial regions from one to ten to 100. The CAR method estimated the model mesangial volume more accurately (1–9 percent error) through each level of complexity compared to point-counting (3–17 percent error) and linear integration (3–18 percent error). The point-counting method consistently overestimated (P < 0.05) the fractional mesangial volume for the ten- and 100-region mesangium models. In Phase II of the study, a normal rat glomerulus was sectioned serially (215 sections) and a transmission electron micrograph (TEM) of every fifth section (n = 43) was obtained. Each TEM image (2% of glomerular surface) was digitized for analysis by CAR. Point-counting and linear integration were also performed on the whole glomerular TEMs (n = 10, randomly chosen). The estimated relative mesangial/glomerular volume was 6.6 ± 0.1 percent by CAR (x? ± SD), 9.7 ± 1.5 by linear integration, and 14.9 ± 3.4 by point-counting. The point-counting method was most efficient, requiring 40 ± 8 sec/section, followed by CAR at 85 ± 24 sec/section. Linear integration was least efficient (93 ± 23 sec/section). We conclude that CAR is the most accurate morphometric method of the three compared for estimating mesangial and glomerular methods, although it is more time consuming than the point-counting method and requires more complex instrumentation. CAR is the only method that will analyze the shape and three-dimensional complexity of glomerular structures using TEMs.  相似文献

Confocal scanning laser microscopy (CSLM) represents an exciting new tool for scientific disciplines which focus on mechanistic studies such as experimental pathology. Enhanced resolution in the specimen plane and rejection of out-of-focus fluorescence flare allow analysis of specific nucleic acid sequences, enzymes, structural macromolecules, and cellular homeostasis utilizing fluorescent probes. Four different experimental applications are discussed which utilize CSLM to evaluate pathological processes at the subcellular, cellular, and tissue levels. Programmed cell death, or apoptosis, is a natural process of significance both during development and as a response to toxic stimuli. CSLM-imaging of nuclei of human B lymphoblastoid cells following exposure to a monofunctional alkylating agent suggests that the degradation of chromatin characteristic of apoptosis may occur in asymmetric patterns. Surfactant apoprotein-A is the major non-serum protein component of pulmonary surfactant and is essential for the extracellular function of surfactant. CSLM of alveolar type II cells suggests that apoprotein-A is present in both the cytoplasm, predominantly in lamellar bodies, and in the nucleus. The tumor promoter, phorbol myristate acetate, rapidly stimulated the formation of vacuoles in human neutrophils. CSLM using Lucifer Yellow as a probe suggests that cylindrical vacuoles are formed by fluid-phase pinocytosis. The blood-nerve barrier (BNB) in peripheral nerves may be an important target during toxin-induced neuropathies. Ricin-induced permeability of the BNB in the rat was rapidly visualized by CSLM as leakage of fluorescein isothiocynate (FITC)-dextran into the endoneurial compartment.  相似文献

Generalist herbivores forage on a variety of plant species, allowing them to gain nutrients while limiting ingestion of harmful toxins. As the capacity to mix diets appears important for maximizing intake, the spatial scale in heterogeneity of food resources should influence the foraging behavior of herbivores. Our aim was to identify how the feeding strategy of a generalist mammalian herbivore, the common brushtail possum, responds to foods within a spatially defined environment. We evaluated foraging responses against increasing spatial separation between pairs of artificial diets that differed in flavor and toxin profile, to determine how distance and diet affect intake and behavior. Foraging responses were influenced by the type of diet or the degree of spatial separation between foods but not by their interaction. Diet influenced intake, time spent feeding, and feeding rate, but had no effect on nightly foraging interval, number of feeding bouts, or bout length. The number of switches between paired food resources and foraging efficiency (intake per unit distance, which accounts for the energetic costs of travelling), were influenced only by distance. Titrating foraging against a range of distances demonstrated how quickly foraging efficiency can decline in response to the spatial separation of food resources, highlighting the importance of spatial heterogeneity of plants within the home range of an herbivore.  相似文献

Inspired by the incorporation of metallocene functionalities into a variety of bioactive structures, particularly antimicrobial peptides, we endeavored to broaden the structural variety of quaternary ammonium compounds (QACs) by the incorporation of the ferrocene moiety. Accordingly, 23 ferrocene-containing mono- and bisQACs were prepared in high yields and tested for activity against a variety of bacteria, including Gram-negative strains and a panel of clinically isolated MRSA strains. Ferrocene QACs were shown to be effective antiseptics with some displaying single-digit micromolar activity against all bacteria tested, demonstrating yet another step in the expansion of structural variety of antiseptic QACs.  相似文献

The reversible oxidation of methionine residues in proteins has emerged as a biologically important post-translational modification. However, detection and quantitation of methionine sulfoxide in proteins is difficult. Our aim is to develop a method for specifically derivatizing methionine sulfoxide residues. We report a Pummerer rearrangement of methionine sulfoxide treated sequentially with trimethylsilyl chloride and then 2-mercaptoimidazole or pyridine-2-thiol to produce a dithioacetal product. This derivative is stable to standard mass spectrometry conditions, and its formation identified oxidized methionine residues. The scope and requirements of dithioacetal formation are reported for methionine sulfoxide and model substrates. The reaction intermediates have been investigated by computational techniques and by 13C NMR spectroscopy. These provide evidence for an α-chlorinated intermediate. The derivatization allows for detection and quantitation of methionine sulfoxide in proteins by mass spectrometry and potentially by immunochemical methods.  相似文献

Terror Management Theory (TMT) suggests that mortality salience (MS), or death reminders, should impact environmental behaviour and decision-making by increasing consumption and resource usage, shifting aesthetic preferences toward cultivated landscapes, and affecting adherence to environmental norms. We examined MS effects on residential flood risk perceptions in Toronto, Canada, following the major urban flood in July 2013. Survey responses were analyzed to assess risk perceptions across nine categories; analyses through the Wilcoxon-Mann–Whitney test show statistically insignificant differences between MS and control groups for all subcategories. Coupled with the TMT literature, the results indicate that existential anxiety is more likely to affect flood risk perceptions through worldview defense when the social dimension of flooding is made clear. Toronto residents’ societal beliefs about flooding play a major role in guiding flood management strategies. Our results provide scholarly and practical direction for flood risk communication strategies to promote the adoption of private flood-prevention measures, suggesting that anthropocentric framing of flooding issues will not induce negative environmental behaviour and may increase concern for flooding issues.  相似文献

Particle emissions from multiple fused deposition modeling consumer 3D printers were systematically quantified utilizing an established emission testing protocol (Blue Angel) to allow quantitative exposure assessments for printers operating in different environments. The data are consistent with particle generation from volatilization of the polymer filament as it is heated by the extruder. Typically, as printing begins, a burst of new particle formation leads to the smallest sizes and maximum number concentrations produced throughout the print job. For acrylonitrile butadiene styrene (ABS) filaments, instantaneous concentrations were up to 106 #/cm3 with mean particle sizes of 20 to 40 nm when measured in a well mixed 1 m3 chamber with 1 air change per hour. Particles are continuously formed during printing and the size distribution evolves consistent with vapor condensation and particle coagulation. Particles emitted per mass of filament consumed (particle yield) varied widely due to factors including printer brand, and type and brand of filament. Higher extruder temperatures result in larger emissions. For filament materials tested, average particle number yields ranged from 7.3 × 108 to 5.2 × 1010 g?1 (approximately 0.65 to 24 ppm), with trace additives apparently driving the large variations. Nanoparticles (diameters less than 100 nm) dominate number distributions, whereas diameters in the range of 200 to 500 nm contribute most to estimated mass. Because 3D printers are often used in public spaces and personal residences, the general public and particularly susceptible populations, such as children, can be exposed to high concentrations of non-engineered nanoparticles of potential toxicity.
