Comparison of changes in temperature, moisture content, infestation and germination were made in six aerated 20 t bins of malting barley. Two were at about 13.5% moisture content, three at about 15.5% moisture content, and one at about 16.5% moisture content. The grain started at between 20–25°C and, during aeration, fell at a rate dependent on the moisture content, damper grain being cooler, presumably due to evaporative cooling. The ‘high’ moisture content grain was often over 5°C cooler at 1m and 2m than the ‘low’ moisture content bins. Moisture uptake at the surface was related to bulk moisture content and trends in mite population changes were related to moisture content. Mite population achieved highest numbers at the grain surface but usually before the moisture content absorption was at its peak. They were commonest in the ‘high’ moisture content bin and least numerous in the ‘low’ moisture content bins. There were no apparent differences between bins in the numbers of insects trapped, for instance they were not less numerous in the coolest ‘high’ moisture content bin. However, the trends of numbers trapped followed a similar pattern in all bins; normally O. surinamensis and S. granarius only began to decline after nine weeks storage in December when temperatures fell below 10°C. Germination loss at the surface of the dampest bin was sufficient to cause rejection of the entire bulk for malting. A promoter that augments gene expression in response to stimulation of ionizing radiation would be a desired tool for radiogenetic therapy, a combination of radiotherapy and gene therapy. Although various promoters occurring naturally or artificially have been used for researches, one showing higher reactivity to ionizing radiation is desirable. In the present study, we attempted to improve a radiation-responsive promoter of the p21 through a technique called DNA shuffling. A library of DNA fragments was constructed by re-ligation of randomly digested promoter fragments and improved promoters were chosen out of the library. We repeated this process twice to obtain a promoter showing 2.6-fold better reactivity to ionizing radiation compared with its parent, p21 promoter after 10 Gy γ-ray irradiation. Nucleotide sequence analyses revealed that the obtained promoter was densely packed with some of the cis-acting elements including binding sites for p53, NF-κB, NRF-2, AP-1 and NF-Y more than p21 promoter. In addition, it was shown that its induction by ionizing radiation was dependent upon p53 status of a cell line, suggesting that the promoter retained properties of the p21 promoter. ABSTRACT: The smoothness and perceived quality of an ice cream depends in large part on the small size of ice crystals in the product. Understanding the mechanisms responsible for producing the disc‐shaped crystals found in ice cream will greatly aid manufacturers in predicting how processing and formulation changes will affect their product. Because ice cream mix is opaque, it has not yet been possible to observe ice crystallization in ice cream in situ. Studies to date, therefore, have used analogues or have related observed effects to a hypothesized mechanism. Still, some elements of the crystallization mechanism are well accepted. Because of the large supercooling at the freezer wall, ice nucleates there before being swept into the bulk of the freezer. In the bulk, heat and mass transfer cause some crystals to melt and others to grow. By the time the ice cream reaches the freezer exit, the ice crystals have become small, rounded discs. 相似文献

Commercially available colostrum replacers (CR) are commonly used when maternal colostrum (MC) is unavailable, for managerial convenience, to ensure quality consistency at first feeding, or in disease control and eradication programs. The objective of this study was to determine the efficacy of feeding First Day Formula (Accelerated Genetics, Baraboo, WI) CR versus pooled MC on immunological status, growth, and health of preweaned dairy calves. A total of 1,220 Jersey and Jersey × Holstein calves born on a California Central Valley dairy farm were assigned after birth to receive either CR or MC following a systematic allocation procedure. Calves assigned to MC were tube fed 2.8 L of MC, and calves assigned to CR were tube fed a total of 500 g of CR (150 g of immunoglobulin G; IgG) mixed into 1.9 L of water at 1 h ± 5 min after the calf was born. A subset of calves was selected for passive transfer (n = 592) and growth (n = 268) analyses. Although both coliform count and total bacteria count were low for MC and CR fed to calves during the study, the predicted probability of calves receiving contaminated liquid feed (coliform count >10,000 cfu/mL) at first feeding was reduced for calves fed CR (1.5%) compared with calves fed MC (6.1%). The mean blood concentration of IgG was lower for calves fed CR than for calves fed MC (19.6 vs. 23.4 mg/mL). However, the apparent efficiency of absorption of IgG did not differ between treatments (34.4 and 35.9% for CR and MC, respectively). Total proteins were lower in calves fed CR compared with MC at 24 h (5.16 vs. 5.84 g/dL, respectively). Calves fed CR were 1.5 kg lighter at weaning and gained 0.03 kg less per day (0.30 vs. 0.33 kg/d, respectively) than calves fed MC before weaning. Height at weaning did not differ between the 2 treatment groups. Calves fed CR tended to have a higher predicted probability of not being treated for diarrhea than calves fed MC (0.142 vs. 0.110, respectively). However, when the disease was present, CR had a higher number of treatment days compared with MC (11.6 vs. 10.8 d, respectively). The hazard ratio of dying did not differ between MC and CR; however, CR calves had a numerically higher risk (hazard ratio = 1.347) of dying compared with calves that received MC. In conclusion, IgG absorption and serum concentration of calves were adequate when calves were fed either CR or MC. The CR-fed calves had a lower probability of receiving contaminated liquid feed and performed similar in terms of health compared with calves receiving high-quality MC, although they were slightly lighter at weaning. The efficacy of Silico-sec® and Diasecticide™ applied as dry dust and slurry applications to wooden surfaces was assessed in the laboratory against adult Sitophilus granarius, and Lepidoglyphus destructor and against larvae of Ephestia kuehniella. Wood was used as the test substrate as it was considered to present a realistic challenge to efficacy due to its 3-dimensional structure. Against the insects, doses of 10 g/m2 Silico-sec® and 20 g/m2 Diasecticide™ were evaluated. Against the mite 1 and 3 g/m2 of the respective Silico-sec® and Diasecticide™ dry dusts and 20 g/m2 of the slurries were assessed. The effects on the insects and mites were assessed one day, 4 weeks and 12 weeks after treatment of the wood. Mortalities were evaluated after exposure times of 24 h and 7 days for the mite and insects, respectively, at conditions of 15 °C and 80% r.h., chosen to represent typical UK conditions after harvest. Of the four treatments, the Silico-sec® dry dust was the most effective against all the pest species, producing 80-95, 100 and 93-100% mean mortalities of S. granarius, E. kuehniella and L. destructor, respectively, over the 12-week experimental period. The Diasecticide™ dry dust produced <11, >99 and 68-93% mortalities of the respective species. The slurry treatments were less effective than the dry dusts with the Silico-sec® slurry producing 29-46, 83-97 and <8% mortalities of the respective species and the Diasecticide™ slurry being ineffective against S. granarius and L. destructor and producing 34-61% mortality of E. kuehniella. This study built on information gained from preliminary work and assessed DEs against tolerant pests on a surface where control was expected to be difficult, in order to provide dose recommendations which would encompass a range of infestation situations in the field. 相似文献

Abstract: The production of grape seed flour (GSF) from a waste product generated during winemaking, is of interest in product development applications due to its potential health benefits. However, before GSF can used in baking as a source of additional antioxidants, research on its heat stability is required. The overall objective of this study was to assess changes in phenolic content and antioxidant activity of GSF during heating. Merlot GSF was heated at 5 temperatures (120 to 240 °C) for 0 to 90 min. At each time/temperature combination, total phenolic content (TPC), total flavanoid content (TFC), Trolox equivalent antioxidant capacity (TEAC), 2,2 diphenyl‐1‐picrylhydrazyl (DPPH) radical scavenging, and ferric ion reducing antioxidant power (FRAP) of the extracts were determined. Specific polyphenolic compounds, including catechin, gallocatechin, epicatechin, and gallic acid were also measured. Results showed that when Merlot GSF was heated to ≥180 °C, significant decreases in the TPC and antioxidant activity, measured using FRAP, DPPH, and TEAC, were observed. Longer heating times also caused a reduction in antioxidant capability. Catechin and epicatechin content decreased with increasing heating temperature while gallocatechin and gallic acid content increased. Both catechin and epicatechin content had strong positive correlations (r > 0.91) with TPC and TFC, as well with FRAP and TEAC, suggesting that the GSF antioxidant activity is related to the presence of these particular compounds. Overall, while a decrease in antioxidant content was observed during heating, this occurred at higher baking temperatures. Thus GSF may be suitable for use as an ingredient in baked goods to bolster antioxidant content. Practical Application: In order for Merlot grape seed flour (GSF) to be used in baking as a source of additional antioxidant compounds, the impact of heating on the polyphenolic compounds in the GSF needed to be examined. Thermal treatment of Merlot GSF caused significant decreases in the TPC, antioxidant power, and specific polyphenolic compounds when heated ≥180 °C. Thus while antioxidant content decreased with higher heating temperatures, this occurred at higher baking temperatures. Grape seed flour (GSF) from grape pomace, a waste product generated during winemaking, was explored for use in bread production due to its potential health benefits. This study evaluated the consumer acceptance and physical properties of bread, including total phenolic content (TPC), made with varying levels of GSF. Dough and breads were prepared using different levels of replacement of hard red spring wheat flour (HRS) with GSF (0 to 10 g GSF/100 g HRS) and stored for 0, 2, or 6 wk at -20 °C. Replacement of 10 g GSF/100 g HRS increased the bread TPC from 0.064 mg tannic acid/g dry weight to 4.25 mg tannic acid/g dry weight. Consumer acceptance and instrumental analyses were used to investigate changes in sensory and texture properties due to GSF replacement. Replacement above 5 g GSF/100 g HRS decreased the loaf brightness and volume, with an increase in the bread hardness and porosity. Generally, breads containing ≥ 7.5 g GSF/100 g HRS were characterized by lower consumer acceptance. A reduction in overall and bitterness acceptance was observed in bread at 10 g GSF/100 g HRS, with decreased acceptance of astringency and sweetness at 7.5 and 10 g GSF/100 g HRS. Based on these results, the replacement of 5 g GSF/100 g HRS is recommended for the production of fortified breads with acceptable physical and sensory properties and high TPC activity compared to refined bread. PRACTICAL APPLICATIONS: This study shows that grape seed flour (GSF) can be used to replace hard red spring wheat flour (HRS) in bread production, with moderate impact on the physical and sensory properties of the bread. Replacement of up to 10 g GSF/100 g HRS significantly decreased overall consumer acceptance of the bread, with lower consumer acceptance of sweetness and astringency at 7.5 and 10 g GSF/100 g HRS. Thus, a replacement value of 5 g GSF/100 g HRS is recommended for the production of fortified breads. 相似文献

Chemical concentration quotients measured between water and total organic carbon (TOC) in sediment (II(SOC)) or suspended particulates (pi(poc)) in southern Lake Michigan reveal up to 2 orders of magnitude differences for polychlorinated biphenyl (PCB), dibenzo-p-dioxin (PCDD), dibenzofuran (PCDF), and polycyclic aromatic hydrocarbon (PAH) compounds with similar octanol-water partition coefficients (K(ow)S). Apparent disequilibria for PAHs, PCDDs, and PCDFs, determined as measured II(SOC)S or pi(poc)S divided by their organic carbon equilibrium partitioning values, are significantly greater than disequilibria of PCBs with similar K(ow)S. Apparent disequilibria, when adjusted for black carbon content by using published black carbon nonlinear partition coefficients (K(f,bc)S) and a Freundlich exponent (n(f)) value = 0.7, still exceed equilibrium predictions for the PAHs, PCBs, and PCDDs but with the PCDF disequilibria uniquely below equilibrium. While Monte Carlo analysis of all the variables associated with the black carbon adjusted disequilibria provides wide confidence intervals for individual chemicals, the large class disequilibria differences between PAHs and PCDFs with respect to the PCBs and PCDDs are highly significant. Use of the PCDD K(f,bc)S for calculating both the PCDF and PCDD disequilibria eliminates their extreme divergence. On the basis of the complexity of carbonaceous geosorbent effects and the apparent variable degrees of chemical sequestration in particles, the disequilibria can be adjusted by chemical class to meet expected near equilibrium conditions between suspended particles and water in the hypolimnion. Although these adjustments to the disequilibria calculations produce consistent and plausible values, the complexities of variable carbonaceous geosorbent affinities for these chemicals in Lake Michigan presently favor use of measured, rather than a priori modeled, steady-state total organic carbon-water concentration quotients indexed to TOC as biogenic organic carbon. 相似文献

Groundwater transport models that accurately describe spreading of nonreactive solutes in an aquifer can poorly predict concentrations of reactive solutes. The dispersive term in the advection-dispersion equation can overpredict pore-scale mixing, and thereby overpredict homogeneous chemical reaction. We quantified this experimentally by imaging instantaneous colorimetric reactions between solutions of aqueous CuSO4 and EDTA4- within a 30-cm long translucent chamber packed with cryolite sand that closely matched the optical index of refraction of water. A charge-coupled device camera was used to quantify concentrations of blue CuEDTA2- within the chamber as it was produced by mixing of the two reactants at different flow rates. We compared these experimental results with a new analytic solution for instantaneous bimolecular reaction coupled with advection and dispersion of the product and reactants. For all flow rates, the concentrations of CuEDTA2- recorded in the experiments were about 20% less than predicted by the analytic solution, thereby demonstrating that models assuming complete mixing at the pore scale can overpredict reaction during transport. 相似文献
