The tubby strain of mice exhibits maturity-onset obesity and sensory deficits in vision and hearing. The mutated gene, tub , responsible for this phenotype was identified recently, but the function of the TUB protein has not been deduced from its amino acid sequence. This prompted us to undertake expression mapping studies with the hope that they might help to elucidate the biological role of the TUB protein. We report the tub gene expression pattern in embryonic, fetal and adult mice tissues as determined by northern blots and in situ hybridization, using antisense oligonucleotidic probes. In mouse embryos, tub is expressed selectively in differentiating neurons of the ensemble of central and peripheral nervous systems, starting at 9.5 days after conception. In adult mice, tub is transcribed in several major brain areas, including cerebral cortex, hippocampus, several nuclei of the hypothalamus controlling feeding behavior, in the spiral ganglion of the inner ear and in the photoreceptor cells of the retina. These structures contain potential cellular targets of the tubby mutation-induced pathogenesis. The neuronal-specific tub gene distribution allows the establishment of a genotype-phenotype correlation in the tubby mice. This correlation is reminiscent of that observed in fat/fat mice, whose phenotype, also characterized by obesity, is caused by a null mutation in the carboxypeptidase E (CPE) gene. Our observations highlight similarities between CPE, prohormone convertases, several neuropeptides and tub gene expression patterns during embryogenesis, and may narrow down the avenues to explore in order to determine ultimately the function of the TUB protein.  相似文献

This paper deals with two corroded reinforcement concrete beams, which have been stored under sustained load in a chloride environment for 14 and 23 years respectively. The evolution of corrosion pattern of reinforcement and its influence on serviceability are studied. In chloride-induced corrosion process, corrosion cracking affects significantly the corrosion pattern. During the corrosion cracking initiation period, only local pitting corrosion occurs. At early stage of cracking propagation, localized pitting corrosion is still predominant as cracks widths are very small and cracks are not interconnected, but a general corrosion slowly develops as the cracks widen. At late cracking stage, interconnected cracking with wide width develops along large parts of the beam leading to a general corrosion pattern. Macrocells and microcells concepts are used for the interpretation of the results.Mechanical experiments and corrosion simulation tests are performed to clarify the influence of this corrosion pattern evolution on the serviceability of the beams (deflection increase). Experimental results show that, when the corrosion is localized (early cracking stage), the steel–concrete bond loss is the main factor affecting the beams serviceability. The local cross-section loss resulting from pitting attack does not significantly influence the deflection of the beam. When corrosion is generalized (late cracking stage), as the steel–concrete bond is already lost, the generalized steel cross-section reduction becomes the main factor affecting the beams serviceability. But, at this stage, the deflection increase is slower due to the low general corrosion rate.  相似文献

Materials traditionally used in integrated circuits have limitations as those circuits reach submicron design rules. Refractory metals have found their place in very-large-scale-integration device technologies, and an experimental program involving the use of tungsten as a gate material has been undertaken, with special emphasis on purity requirements. Bias temperature stress capacitance-voltage measurements show that very low levels of mobile ion contamination can be obtained in metal oxide semiconductor capacitors with tungsten gates sputtered from a special high-purity target.  相似文献

The ability to efficiently direct visual attention to salient features in the environment is a critical function of the visual system. The finding that people are slower to detect a target that appears at a previously cued location is thought to reflect a mechanism known as inhibition of return (IOR). Past research has shown that difficult target discriminations result in a greater amount of time needed to inhibit previously attended locations (i.e., a delayed onset of inhibition), suggesting that task difficulty plays a critical role in the allocation of attention. In this study, IOR was measured at a wide range of SOAs while participants detected either a perceptually degraded target or a standard, high luminance target. When responses were made to a perceptually degraded target, the time course of IOR was delayed by approximately 250 ms (relative to the control group), suggesting that the difficulty in detecting targets also influences the allocation of attention. The results are consistent with the notion that IOR is not simply a reflexive subcortical mechanism but rather involves top-down attentional control settings. This paper will focus on the study of reinforced concrete beams stored in a chloride environment for a period of 14–23 years under service loading. According to the experimental results, a Serviceability Limit State (SLS) criteria is proposed based on an excessive steel–concrete bond reduction. Corrosion of reinforcement in chloride environment leads to a specific local steel cross-section loss as well as a steel–concrete bond loss. Experimental results have shown that, in the first stage of corrosion propagation period, the deflection is more sensitive to chloride-induced corrosion than the ultimate capacity due to the effect of the tension steel–concrete bond loss even if both are correlated. Given this high sensibility of the bending stiffness to corrosion pitting attacks, it appears that a Serviceability Limit State (SLS) criteria based on excessive deflection of structural members is an adequate factor for SLS assessment. Later in corrosion propagation period, when the bond is already significantly reduced, only the ultimate capacity is affected by the steel cross-section loss. This does not affect the serviceability, because pitting attacks are very localised with an insignificant influence on the global deflection. Then, once the steel–concrete bond is lost in critical parts of the beams (high bending moment areas), pitting corrosion propagation does not affect anymore serviceability (stiffness reduction, bending or corrosion cracks patterns) but still leads to an ultimate capacity reduction, which is not acceptable. As a result, excessive steel–concrete debonding can be considered as the SLS criteria.  相似文献
