A simple device is described to couple a fast-scanning acoustooptic tunable filter-based NIR spectrophotometer to a distillation apparatus for monitoring the condensed vapor in real time. The device consists of a small funnel whose glass neck (2-mm diameter) is bent into an "U" format to produce a flow cell of approximately 150-microL inner volume. A pair of optical fibers is used to deliver the monochromatic light and to collect the fraction passing through the glass tube. The end of the condenser of the distillation head touches the wall of the small funnel. The condensed liquid flows uncoupled from pressure changes in the interior of the distillation head. Absorbance spectra were obtained, during the distillation, as averages of 50 scans (4 s) every 5 s in the spectral range 950-1800 nm with nominal resolution of 2.0 nm. In the first experiments, the distillations were performed at constant power supplied to the sample (25 mL) in a microdistillation apparatus working without any type of reflux column. The usefulness of the real-time monitoring of distillation is demonstrated using some prepared binary mixtures and by comparing the distillation behavior of adulterated and regular gasoline samples. Data analysis and interpretation are facilitated by employing principal component analysis. The system accesses the composition of the condensate, which can separate and concentrate one or more compounds present in the original sample.  相似文献

The biological response to zirconia (ZrO2) is not completely understood, which prompted us to address its effect on pre-osteoblastic cells in both direct and indirect manner. Our results showed that zirconia triggers important intracellular signaling mainly by governing survival signals which leads to cell adhesion and proliferation by modulating signaling cascade responsible for dynamic cytoskeleton rearrangement, as observed by fluorescence microscopy. The phosphorylations of Focal Adhesion Kinase (FAK) and Rac1 decreased in response to ZrO2 enriched medium. This corroborates the result of the crystal violet assay, which indicated a significant decrease of pre-osteoblast adhesion in responding to ZrO2 enriched medium. However, we credit this decrease on pre-osteoblast adhesion to the need to govern intracellular repertory of intracellular pathways involved with cell cycle progression, because we found a significant up-phosphorylation of Mitogen-Activated Protein Kinase (MAPK)-p38 and Cyclin-dependent kinase 2 (CDK2), while p15 (a cell cycle suppressor) decreased. Importantly, Protein phosphatase 2?A (PP2A) activity decreased, guaranteeing the significant up-phosphorylation of MAPK -p38 in response to ZrO2 enriched medium. Complementarily, there was a regulation of Matrix Metalloproteinases (MMPs) in response to Zirconia and this remodeling could affect cell phenotype by interfering on cell anchorage. Altogether, our results show a repertory of signaling molecules, which suggests that ECM remodel as a pre-requisite to pre-osteoblast phenotype by affecting their anchoring in responding to zirconia.  相似文献

A flow injection analyser has been constructed to allow an operator-free determination of up to 40 samples. Besides the usual FIA apparatus, the analyser includes a home-made sample introduction device made with three electromechanical three-way valves and an auto-sampler from Technicon which has been adapted to be commanded by an external digital signal. The analyser is controlled by a single board SDK-8085 microcomputer. The necessary interface to couple the analyser components to the microcomputer is also described. The analyser was evaluated for a Cr(VI)-FIA determination showing a very good performance with a relative standard deviation for 15 signals from the injection of 100 μl of a 1.0 mg.ml-1 standard Cr(VI) solution being equal to 0.5%.  相似文献

The effect of different overrun levels on the sensory acceptance and survival of probiotic bacteria in ice cream was investigated. Vanilla ice creams supplemented with Lactobacillus acidophilus were processed with overruns of 45%, 60%, and 90%. Viable probiotic bacterial counts and sensory acceptance were assessed. All the ice creams presented a minimum count of 6 log CFU/g at the end of 60 d of frozen storage. However, higher overrun levels negatively influenced cell viability, being reported a decrease of 2 log CFU/g for the 90% overrun treatment. In addition, it was not reported an influence about acceptability with respect to appearance, aroma, and taste of the ice creams (P > 0.05). Overall, the results suggest that lower overrun levels should be adopted during the manufacture of ice cream in order to maintain its probiotic status through the shelf life.  相似文献

An automated system to perform liquid-liquid extraction in monosegmented flow analysis is described. The system is controlled by a microcomputer that can track the localization of the aqueous monosegmented sample in the manifold. Optical switches are employed to sense the gas-liquid interface of the air bubbles that define the monosegment. The logical level changes, generated by the switches, are flagged by the computer through a home-made interface that also contains the analogue-to-digital converter for signal acquisition. The sequence of operations, necessary for a single extraction or for concentration of the analyte in the organic phase, is triggered by these logical transitions. The system was evaluated for extraction of Cd(II), Cu(II) and Zn(II) and concentration of Cd(II) from aqueous solutions at pH 9.9 (NH3/NH4Cl buffer) into chloroform containing PAN (1-(2-pyridylazo)-2-naphthol) . The results show a mean repeatability of 3% (rsd) for a 2.0 mg l-1 Cd(II) solution and a linear increase of the concentration factor for a 0.5mg l-1 Cd(II) solution observed for up to nine extraction cycles.  相似文献

A spectrometer has been constructed to detect the radiation emitted by thermally excited samples in the near-infrared spectral region extending from 1500 to 3000 nm. The instrument employs an acousto-optical tunable filter (AOTF) made of TeO2 and attains maximum sensitivity by making effective use of the two diffracted beams produced by the anisotropic AOTF. The full exploitation of the transmitted power of the monochromatic beams is reported for the first time and became possible because the detector does not saturate when employed for the acquisition of the weak emission signal in the NIR region, even when exposed to the total (nondiffracted) beam. Thus, modulation and lock-in-based detection can be employed to find the intensity of the diffracted beams superimposed on the nondiffracted beam. The resolution is slighted degraded in view of the small (approximately 10 nm) difference in the wavelength diffracted in the ordinary and extraordinary beams. The instrument has been evaluated in terms of signal-to-noise ratio, effect of sample thickness, and excitation temperature and for its potential in analytical applications in monitoring high-temperature kinetics, for qualitative identification of inorganic solids, for use with a closed cell to obtain spectra of species that evaporate at the temperatures (> 150 degrees C) necessary for sample excitation, and for quantitative purposes in the determination of soybean oil content in olive oil. The feasibility of near-infrared emission spectroscopy has been demonstrated together with some of its advantages over mid-infrared emission spectroscopy, such as greater tolerance to sample thickness, suitable signal-to-noise, and its use in the investigation of kinetic phenomena and phase transitions at high temperatures.  相似文献

The atomic force microscope (AFM) offers the possibility to obtain high-resolution images of biological samples immersed in a liquid. This ability was exploited to image the erythrocyte plasma membrane Ca2+ ATPase reconstituted in asolectin liposomes. The liposomes prepared in the presence of the Ca2+ ATPase exhibited particles protruding from their flat surface, which were absent in control preparations, (i.e., in liposomes prepared in the absence of the ATPase). Incubation of the liposomes containing the pump with IgM type monoclonal antibodies raised against the Ca2+ ATPase induces a 3–5-fold increase in the size of the protruding particles, demonstrating that the protruding object observed is the reconstituted Ca2+ ATPase.  相似文献

Formestane is a selective inhibitor of oestrogen synthesis by aromatase enzymes and induces disease regression in breast cancer patients. This phase II randomised study was carried out to determine whether there were any differences in the effects of two different doses of formestane on oestradiol (E2) serum levels and to evaluate the corresponding clinical activity in post-menopausal patients with positive or unknown oestrogen receptor status pretreated or not for advanced disease. Furthermore, possible drug interference with adrenal steroidogenesis was assessed by measuring 17-hydroxycorticosteroid (17-OHCS) urinary levels. A total of 143 patients entered the study and were randomly assigned to receive formestane 250 mg (72 patients) or formestane 500 mg (71 patients), both given i.m. every 2 weeks. In comparison with baseline, E2 serum levels decreased by an average of 40% after only 15 days and remained unchanged thereafter, with no difference being observed between the two doses. The values of 17-OHCS remained unchanged during treatment in both groups. Objective responses were 28% (19/69) in the 250 mg and 46% (31/68) in the 500 mg group. In conclusion, the two formestane doses were equally effective in reducing E2 levels without affecting adrenal function, and in inducing a considerable percentage of clinical responses.  相似文献
