C-fos is an early expression oncogene that can be stimulated by a variety of regulators. It is expressed by subsets of all pituitary cells, with increased expression seen in proestrous rats. However, in freshly dispersed pituitary cells studied during different stages of the cycle, there is limited expression of fos by luteinizing hormone (LH) cells and little basal expression by cells with follicle-stimulating hormone (FSH) antigens. Proestrus is a time during which pituitary gonadotropes express peak levels of receptors for gonadotropin-releasing hormone (GnRH) and epidermal growth factor (EGF). We hypothesized that if GnRH or EGF stimulated fos activity in gonadotropes they would be most effective during the peak expression of their receptors. Anterior pituitaries were removed, cut into small pieces, and stimulated for 30 min. Total RNA was then collected and analyzed by Northern analysis. Both EGF and GnRH caused an increase in c-fos mRNA levels in the anterior pituitary gland compared with unstimulated pituitary glands assayed immediately after removal from the pituitary. However, the stimulatory effects were no greater than those seen with medium alone. This suggested that fos expression could be stimulated by local factors either in the pituitary or the medium itself. The second phase of the study focused on pituitary cells plated for 1 hr and then stimulated with EGF and GnRH for 15 min. Dual immunocytochemistry was done to learn which cell types expressed the fos proteins. After 15 min, EGF and GnRH both increased the percentages of fos-bearing cells above levels seen in medium alone. EGF stimulated fos proteins in subsets of FSH, adrenocorticotropin (ACTH), and growth hormone (GH) cells. GnRH increased fos proteins in subsets of ACTH and GH cells. These results suggest that EGF and GnRH may regulate fos expression, but not necessarily in gonadotropes. They also highlight the need for carefully timed experiments because endogenous factors in the pituitary itself may stimulate immediate early gene expression. Planar carbon-based electronic devices, including metal/semiconductor junctions, transistors and interconnects, can now be formed from patterned sheets of graphene. Most simulations of charge transport within graphene-based electronic devices assume an energy band structure based on a nearest-neighbour tight binding analysis. In this paper, the energy band structure and conductance of graphene nanoribbons and metal/semiconductor junctions are obtained using a third nearest-neighbour tight binding analysis in conjunction with an efficient nonequilibrium Green's function formalism. We find significant differences in both the energy band structure and conductance obtained with the two approximations.  相似文献

Object

At high-field strengths (≥3T) inhomogeneity of the radio frequency (RF) field and RF power deposition become increasingly problematic. Parallel Transmission (PTx)—the use of segmented transmission arrays with independently driven elements—affords the ability to combat both of these issues. There are a variety of existing designs for PTx coils, ranging from systems with two channels to systems with eight or more. In this work, we have investigated the impact of the number of independent channels on the achievable results for both homogeneity improvement and power reduction in vivo.

Materials and methods

A 3T Philips Achieva MRI system fitted with an 8-channel PTx body coil was driven so as to emulate configurations with 1, 2 4 and 8 independent channels. RF shimming was used in two different anatomies in order to assess improvements in RF homogeneity.

Results

Significant homogeneity improvements were observed when increasing from 1 to 2, 2 to 4, and 4 to 8 channel configurations. Reductions in RF power requirements and local SAR were predicted for increasing numbers of channels.

Conclusion

Increasing the number of RF transmit channels adds extra degrees of freedom which can be used to benefit homogeneity improvement or power reduction for body imaging at 3T.  相似文献

Theory has emphasized the importance of both intrinsic factors such as host immunity and extrinsic drivers such as climate in determining disease dynamics. In particular, seasonality may lead to multi-annual cycles in prevalence, but the likelihood of this depends on the role of acquired immunity. Some diseases including malaria have immunity that falls between the classic susceptible–infectious–removed and susceptible–infectious–susceptible models. Here, we investigate the general conditions promoting the subharmonic resonance behaviour that may lead to multi-annual cycles in a general malaria dynamical model. Utilizing two complementary approaches to bifurcation analyses, we show that resonance is promoted by processes shortening the length of the infectious period and that subharmonic cycles are favoured in situations with strong seasonality in transmission but at intermediate levels of endemicity. We discuss the implications of our results for understanding prevalence patterns in long-term malaria datasets from Kenya that show multi-annual cycles and one from Thailand that does not and discuss the possible implications of treatment.  相似文献

Recent research into motivation and achievement behaviour in sport has focused on achievement goal theory. This theory states that two goal orientations manifest themselves in achievement contexts and impact on the motivation process. These two goals have been defined as 'task' and 'ego' goal orientations. This paper traces the development of the Perception of Success Questionnaire as a measure of achievement goals developed specifically for the sport context. The early development of the questionnaire is documented, in which the scale was shortened from the initial 29 to the current 12 question format. We demonstrate that task and ego goals are orthogonal, internal reliabilities for the orientations are high, with strong construct and concurrent validity. We conclude by reporting results from two recent confirmatory factor analyses that were conducted on the Children's and Adult versions of the questionnaire; these results show the Perception of Success Questionnaire to be a reliable and valid instrument to measure achievement goal orientations in sport.  相似文献
