Oxidized low-density lipoprotein (ox-LDL) is the most harmful form of cholesterol associated with vascular atherosclerosis and hepatic injury, mainly due to inflammatory cell infiltration and subsequent severe tissue injury. Lox-1 is the central ox-LDL receptor expressed in endothelial and immune cells, its activation regulating inflammatory cytokines and chemotactic factor secretion. Recently, a Lox-1 truncated protein isoform lacking the ox-LDL binding domain named LOXIN has been described. We have previously shown that LOXIN overexpression blocked Lox-1-mediated ox-LDL internalization in human endothelial progenitor cells in vitro. However, the functional role of LOXIN in targeting inflammation or tissue injury in vivo remains unknown. In this study, we investigate whether LOXIN modulated the expression of Lox-1 and reduced the inflammatory response in a high-fat-diet mice model. Results indicate that human LOXIN blocks Lox-1 mediated uptake of ox-LDL in H4-II-E-C3 cells. Furthermore, in vivo experiments showed that overexpression of LOXIN reduced both fatty streak lesions in the aorta and inflammation and fibrosis in the liver. These findings were associated with the down-regulation of Lox-1 in endothelial cells. Then, LOXIN prevents hepatic and aortic tissue damage in vivo associated with reduced Lox-1 expression in endothelial cells. We encourage future research to understand better the underlying molecular mechanisms and potential therapeutic use of LOXIN.  相似文献

We presentthe results of a research work targeted to understanding thedomains and consequences of CASE tools usage in Nokia. We aimto evaluate the importance of the various CASE tools features,as rated by our developers, and how well such features are implementedin currently available CASE tools. A structured questionnairewas sent to our most experienced developers and CASE users. Fromthis survey, it emerged that CASE tools support is reputed mostuseful for the following functions: graphical drawing, automaticdocumentation generation and storage of diagrams. The resultshint to a mismatch between the features required by the developersand those offered by CASE products. Further research is neededbefore more definite conclusions can be drawn.  相似文献

A new algorithm for the integration of partially overlapping range images into a triangular mesh is presented. The algorithm consists of three main steps: it locates the intersections between the range surfaces and a reference grid chosen by the user, then merges all nearly coincident and redundant intersections according to a proximity criterion, and, finally, reconstructs the merged surface(s) from the filtered intersection set. Compared with previous methods, which adopt a volumetric approach, our algorithm shows lower computational costs and improves the accuracy of the surfaces produced. It takes into account the quality of the input measurements and is able to patch small holes corresponding to the parts of the 3D scanned object that were not observed by the acquisition device. The algorithm has been tested on several datasets of range maps; graphical and numeric results are reported.  相似文献

Varicocele (VC) is the most common abnormality identified in men evaluated for hypofertility. Increased levels of reactive oxygen species (ROS) and reduced antioxidants concentrations are key contributors in varicocele-mediated hypofertility. Moreover, inflammation and alterations in testicular immunity negatively impact male fertility. In particular, NLRP3 inflammasome activation was hypothesized to lead to seminal inflammation, in which the levels of specific cytokines, such as IL-1β and IL-18, are overexpressed. In this review, we described the role played by oxidative stress (OS), inflammation, and NLRP3 inflammasome activation in VC disease. The consequences of ROS overproduction in testis, including inflammation, lipid peroxidation, mitochondrial dysfunction, chromatin damage, and sperm DNA fragmentation, leading to abnormal testicular function and failed spermatogenesis, were highlighted. Finally, we described some therapeutic antioxidant strategies, with recognized beneficial effects in counteracting OS and inflammation in testes, as possible therapeutic drugs against varicocele-mediated hypofertility.  相似文献

A short survey of cellular automata based models for corrosion and passivation phenomena is given. Results of simulations based on the model of spatially separated anodic and cathodic reactions are presented for a cavity development from a point-like damage of a protective layer and from an initially flat unprotected surface. We show some new peculiar examples of the symmetry breaking in the cavity development. The results for the initially flat surface show roughening of the surface at the beginning of the corrosion process. After that, pit merging causes a resmoothing of the surface. An oscillatory behavior of the surface roughness is observed caused by a peninsula formation with subsequent island detachment. These results are obtained in 2D because of computational limitations. We plan simulations in 3D and point out the problems we encounter in their realization.  相似文献

Basin and petroleum systems modelling is a powerful tool in petroleum exploration, but uncertainties remain in terms of the evaluation of a petroleum accumulation's size and quality, even when the petroleum system is well known and the latest modelling technology is applied. In order to interpret the results of a modelling exercise, it is necessary to understand the advantages and disadvantages of the various possible approaches used to assess petroleum migration and accumulation. This paper attempts to compare the influence of different migration modelling techniques – the flowpath, Darcy “hybrid” and invasion percolation approaches – on basin‐wide mass balance calculations for a temperature‐ and pressure‐calibrated, numerical four‐dimensional basin model. The study was performed using PetroMod® software. The study area is the well‐known Jeanne d'Arc Basin located offshore Newfoundland, eastern Canada. Model predictions were verified against pre‐existing data including the quantity and quality of discovered hydrocarbons in the basin. Modelling results showed that the Darcy method produced substantially different results compared to the other migration techniques and this was due to the high accumulation efficiency. The flowpath method, and a combination of flowpath and Darcy methods (referred to as the “hybrid” method) yielded similar results; furthermore, the hybrid method predicted the petroleum composition quite accurately. The invasion percolation method gave similar results to the hybrid approach, but little or no variation in API and GOR across the basin was predicted. The adsorption model initially applied did not adequately reproduce the natural behaviour of source rocks with respect to petroleum expulsion efficiency. Therefore a revised model was implemented in which the adsorption capacity was reduced with increasing maturity. This revised adsorption model led to more realistic volumes of hydrocarbons being retained within the source rock. The application of this approach had only a minor impact on the volume and quality of the petroleum present in the reservoir units.  相似文献
