The Clinical Significance of Transfer RNAs Present in Extracellular Vesicles

Extracellular vesicles (EVs) are important for intercellular signalling in multi-cellular organisms. However, the role of mature transfer RNAs (tRNAs) and tRNA fragments in EVs has yet to be characterised. This systematic review aimed to identify up-to-date literature on tRNAs present within human EVs and explores their potential clinical significance in health and disease. A comprehensive and systematic literature search was performed, and the study was conducted in accordance with PRISMA guidelines. Electronic databases MEDLINE and EMBASE were searched up until 1 January 2022. From 685 papers, 60 studies were identified for analysis. The majority of papers reviewed focussed on the role of EV tRNAs in cancers (31.7%), with numerous other conditions represented. Blood and cell lines were the most common EV sources, representing 85.9% of protocols used. EV isolation methods included most known methods, precipitation being the most common (49.3%). The proportion of EV tRNAs was highly variable, ranging between 0.04% to >95% depending on tissue source. EV tRNAs are present in a multitude of sources and show promise as disease markers in breast cancer, gastrointestinal cancers, and other diseases. EV tRNA research is an emerging field, with increasing numbers of papers highlighting novel methodologies for tRNA and tRNA fragment discovery.     

Pial Vessel-Associated Microglia/Macrophages Increase in Female Dahl-SS/Jr Rats Independent of Pregnancy History

As the resident immune cells of the central nervous system, microglia have a wide range of functions such as surveillance, phagocytosis, and signaling through production of chemokines and cytokines. Recent studies have identified and characterized macrophages residing at the meninges, a series of layers surrounding the brain and spinal cord. While perivascular microglia within the brain parenchyma increase following chronic hypertension, there are no reports of changes at the meninges, and specifically, associated with the pial vasculature. Thus, we used female Sprague Dawley and Dahl salt-sensitive (SS/Jr) rat brains, stained for ionized calcium-binding adapter molecule (Iba1), and characterized microglia/macrophages associated with pial vessels in the posterior brain. Results indicate that Iba1⁺ pial vessel-associated microglia (PVAM) completely surrounded the vessels in brains from the Dahl-SS/Jr rats. PVAM density was significantly higher and distance between PVAMs lower in Dahl-SS/Jr compared to the Sprague Dawley rat brains. Pregnancy history did not affect these findings. While the functional role of these cells are not known, we contextualize our novel findings with that of other studies assessing or characterizing myeloid cells at the borders of the CNS (meninges and choroid plexus) and perivascular macrophages and propose their possible origin in the Dahl-SS/Jr model of chronic hypertension.     

Positive and negative opinion modeling: The influence of another's similarity and dissimilarity.

Modeling research that has focused on the effects of observing similar others appears to have underestimated the influence of observing dissimilar others. Two experiments demonstrated that observing a model express liking for a piece of music induced more favorable opinions of the music (positive modeling) when the model was similar to the participant observer in relevant opinions, and more negative opinions (negative modeling) when the model was dissimilar to the participant in relevant opinions. Of note, this pattern was more pronounced when participants also believed their general backgrounds were dissimilar rather than similar to that of the model. Optical Kerr switching technique for the production of a picosecond,multiwavelength CO2 laser pulse

A wavelength-independent method for optical gating, based on the optical Kerr effect, has been demonstrated. Using this method, we produced 100-ps, 10-kW, two-wavelength pulses (10.3 and 10.6 microm) with a signal-to-background ratio contrast of 10(5) by slicing a long CO2 pulse. The capability of gating consecutive pulses separated on a picosecond time scale with this method is also shown.     

An Integrated Framework for Implementing Quality of Network Concepts

This paper presents an integrated framework for describing and implementing quality of network (QoN) concepts. It focuses on identifying a systems engineering framework for better understanding how to specify and implement QoN concepts. We demonstrate, by means of several examples, that the quality of service interfaces between the end user and the underlying network are very important. Finally, we provide insights as to future development efforts needed to realize the goal of high quality networks.     

The modulation of cellular responses to poly(2-hydroxyethyl methacrylate) hydrogel surfaces: phosphorylation decreases macrophage collagenase production in vitro

We examined the regulation of collagenase production by the monocyte/macrophage THP-1 cell line when these cells were exposed to poly(2-hydroxyethyl methacrylate) (PHEMA) hydrogel surfaces with different chemistries and morphologies. Tissue culture modified polystyrene (TCP), used as a control surface, induced the maximum collagenase response. Copolymer hydrogels containing 2-ethoxyethyl methacrylate (EMA) or methyl methacrylate (MMA) also induced a high response, while PHEMA hydrogels induced a low level response and the phosphorylated hydrogel induced no response. This pattern was altered when the morphology of the hydrogels was changed to that of a sponge. The overall enzyme response to the sponge hydrogels was lower than that to the homogeneous hydrogels. Sponges containing EMA and MMA produced low level response relative to the TCP control. PHEMA and phosphorylated sponges produced little and no response respectively. The dramatically reduced enzyme response to phosphorylated surfaces was not a consequence of cell death, and may be a phenomenon related to changes in cell surface charge.     

Effect of individual, group, and intergroup relations on attitudes toward a management development program.

Studied the impact of a management development program in a medium-sized bank that was designed to increase the number of competent and aggressive young managers available to the organization. In this situation after a period of 10 yr., the training program had developed into a subsystem of the organization. 263 employees completed an attitude and job satisfaction questionnaire. Attitudes toward the program were a function of seniority, organizational rank, pay satisfaction, and perception of own career prospects. The kinds of attitudes also depended heavily on status as an employee, trainee, or officer, and on sex. Behavioral explanations of the attitude effects are presented.
