Intensity discrimination for precedence effect stimuli

When two identical stimuli are presented from two loudspeakers with a brief delay between them, a single image is heard near the source of the leading sound. The delayed sound or echo appears to be suppressed whereas the preceding sound determines perceived location, hence the name, the precedence effect. This study investigated normal-hearing listeners' sensitivity to changes in the intensity of the lagging sound. Pairs of 2-ms white noise bursts, with a 2-ms delay between the onsets of lead and lag, were presented from two loudspeakers 45 degrees left and right of midline in an anechoic chamber. A 2AFC procedure was used to test discrimination of intensity changes in the lead, lag, and both sounds together. The untreated results showed discrimination to be poorest for changes in the lag stimulus. However, when the intensity differences were transformed into predictions of equivalent monaural level based on KEMAR measurements and binaural loudness summation, discrimination for the lag was equal to the other two conditions. A follow-up experiment found that listeners were highly sensitive to the presence of the lag, more sensitive than would be predicted from loudness changes. It is concluded that the precedence effect does not consist of a general suppression or attenuation of the lagging sound, but rather that suppression may be limited to directionality cues.

The adjoint of CMAQ

An adjoint model for the internationally used Community Multiscale Air Quality (CMAQ) modeling platform of the U.S. EPA is developed. The adjoint version for CMAQ (CMAQ-ADJ) provides the user community with forward (decoupled direct method or DDM) and backward (adjoint) sensitivity analysis capabilities. Current implementation is for gas-phase processes. Discrete adjoints are implemented for all processes with the exception of horizontal advection, for which, because of inherent discontinuities in the advection scheme, the continuous approach is superior. The adjoint of chemistry is constructed by interfacing CMAQ with the kinetic pre-processor, which provides for increased flexibility in the choice of chemical solver and facilitates the implementation of new chemical mechanisms. The adjoint implementation is evaluated both on a process-by-process basis and for the full model. In general, adjoint results show good agreement with brute-force and DDM sensitivities. As expected for a continuous adjoint implementation in a nonlinear scheme, the agreement is not perfect for horizontal transport. Sensitivities of various air quality, public health, and environmental metrics with respect to emissions are calculated using the adjoint method. In order to show applicability to regional climate studies, as an example, the sensitivities of these metrics with respect to local temperatures are calculated.     

Activation of p38 mitogen-activated protein kinase by lipopolysaccharide in human neutrophils requires nitric oxide-dependent cGMP accumulation

This study examined the signal transduction pathway(s) leading to phosphorylation of p38 in human neutrophils stimulated with lipopolysaccharide and formyl peptides. Blockade of the nitric oxide (NO) pathway in neutrophils with the NO synthase inhibitor N-nitro-L-arginine methyl ester or by treatment with the NO scavenger 2-phenyl-tetramethylimidazoline-1-oxyl-3-oxide attenuated phosphorylation of the mitogen-activated protein kinase p38 in response to lipopolysaccharide but not fMet-Leu-Phe. Using the NO releasing agents S-nitroso-N-acetylpenicillamine and sodium nitroprusside it was determined that nitric oxide is sufficient to cause an increase in phosphorylation of p38. Increasing cellular cGMP with phosphodiesterase inhibitors, by stimulation of soluble guanylyl cyclase with YC-1 or with exogenous dibutyryl cGMP resulted in mitogen-activated protein kinase/extracellular signal-regulated kinase kinase 3,6 (MEK3,6) activation and phosphorylation of p38. This phenomenon was specific for MEK3,6, because these agents had no effect on the phosphorylation state of MEK1,2. A role for protein kinase G but not protein kinase A downstream of lipopolysaccharide but not formylmethionylleucylphenylalanine was shown using the specific inhibitors KT5823 and H89, respectively. These data indicate that activation of p38 by fMet-Leu-Phe and lipopolysaccharide involve different mechanisms, and that activation of protein kinase G by NO-dependent stimulation of guanylyl cyclase is necessary and sufficient for phosphorylation of p38 downstream of lipopolysaccharide.     

Cryopreservation of bovine buffy coat leukocytes for use in immunologic studies

A simple cryogenic technique for preserving bovine buffy coat leukocytes was developed. This was coupled with a variation of the standard discontinuous gradient technique to purify mononuclear cells that retained immunologic function. The total number of mononuclear cells recovered from cyropreserved samples were only 87 to 42% of those recovered from freshly obtained blood samples. However, the functional capabilities of mononuclear cells from cyopreserved buffy coat preparations were retained. Polyclonal proliferative responses to 3 mitogens were measured, using a titration of mitogen concentrations, and were found to be normal, compared with those of cells isolated from fresh blood. Blood samples collected after vaccination with Brucella abortus contained leukocytes that responded to irradiated B abortus. These antigen-specific responses were also retained through cyopreservation. Cell surface expression of T-lymphocyte antigens, CD2, CD4, and CD8, and cell-surface IgM on B lymphocytes was also evaluated. Flow cytometric analysis of fresh and cryopreserved mononuclear cell preparations indicated that the relative proportions of different subpopulations were not altered. The technical simplicity of our cryopreservation system will allow processing of large numbers of samples with the ability to assay various immune functions at a later time.
