Lung function and immunologic studies have been performed in 9 patients with bird breeder's hypersensitivity pneumonitis at the time of diagnosis and at intervals from 8 to 30 months after they ceased exposure to the provoking antigen. All 9 patients had evidence of small airway obstruction at the time of diagnosis. Complete recovery of lung function occurred in 4 patients. The other 5 all had evidence of interstitial damage; 3 of them had progressive increase in the degree of airway obstruction, and one had progressive loss of elastic recoil. The patients were nonsmokers, and these progressive changes could not be explained by occult exposure to antigen because the tests of immunologic sensitivity to relevant avian antigens became negative. Neither the nature or degree of lung function abnormality nor the form of clinical presentation was related to the development of residual damage. The period of exposure to antigen after symptoms developed and the age of the patient appeared to be the most important factors determining recovery of lung function. 相似文献

The effects of curing on removal of solvent and the development of chemical crosslinks were studied in a preimidized, intrinsically photosensitive polyimide. The polymer, Probimide 412, was in solution of γ-butyrolactone (GBL), and spun cast or doctor bladed to form samples for analysis. The films were systematically studied from soft (100°C) to hard bake (400°C), as the effects of cure environment (i.e., air vs. nitrogen) and UV exposure were monitored. The samples were characterized by thermogravimetric analysis (TGA), Fourier transform infrared spectroscopy (FTIR), and dynamic mechanical analysis (DMA). The index of refraction was measured by waveguide propagation mode determination. TGA results show that there is a distribution of solvent removal temperatures, probably due to polymer-solvent binding. Solvent is removed from the polymer as it is thermally cured from ∼150°C to 300°C, and UV curing aids in the removal of solvent. Chemical crosslinks induced by both thermal and UV curing increase the refractive index and darken the material. Crosslinking broadens the glass transition and reinforces the rubbery modulus. FTIR results show that thermally induced crosslinking occurs in the polymer at 400°C in nitrogen and at 300°C in air but does not occur appreciably at 300°C in nitrogen or at any lower temperature. Ultraviolet curing also crosslinks the polymer and is caused by hydrogen abstraction from alkyl substituents and subsequent crosslinking at the benzophenone carbonyl group. During thermal curing, although there is evidence from FTIR spectroscopy that a similar reaction may occur, there are probably multiple reactions simultaneously taking place. 相似文献

BACKGROUND: To determine the effects of anatomic site on the presentation and diagnosis of malignant peripheral nerve sheath tumors (MPNSTs) and on the treatment and outcomes of the patients, the authors initiated a study of these tumors at different sites. An earlier report described MPNSTs of the buttock and lower extremity, and the current series analyzes those presenting at intrathoracic (IT) and subdiaphragmatic (SD) paraspinal sites. METHODS: The authors reviewed data on patients with paraspinal MPNSTs who were seen at Memorial Hospital during the period 1960-1995 and for whom histologic slides were available. Various clinicopathologic parameters and their effects on patient outcomes were examined. RESULTS: Twenty-five patients with 26 tumors were evaluated. Seven tumors were IT and 19 were SD; 60% of the patients had neurofibromatosis type 1 (NF1). Most patients presented with pain, and a diagnostic delay (of 3 months to 2 years) was often noted. Mean tumor sizes for SD and IT tumors were 14.3 cm and 6.6 cm, respectively. Most MPNSTs were composed of spindle cells in fascicles. Twenty-seven percent exhibited divergent differentiation. Twenty-four tumors were high grade, and a low grade component was identified in 8 tumors. Surgical resection was attempted for 23 tumors (88%), but complete resection was achieved in only 6 cases (23%). Eighty percent of the patients died of their tumors, 2-year and 5-year survival rates were 35% and 16%, and median survival was 8.5 months. Significant prognostic factors were tumor size <5 cm, the presence of a low grade component, and complete tumor resection. CONCLUSIONS: Paraspinal MPNSTs have more aggressive behavior than peripherally located tumors, mainly because of the difficulty encountered in resecting them completely. Prognoses of patients with MPNST at this site appear to be affected by resection status, tumor size, and tumor grade. 相似文献

The cytologically active secondary lipid peroxidation products, malondialdehyde (MDA) and 4-hydroxy-2-nonenal (HNE) have been detected as their 2,4-dinitrophenylhydrazone (DNP) derivatives in plant tissue cultures using LC-MS. This paper reports, for the first time, the use of LC-MS methodology to definitively identify 4-hydroxy-2-nonenal in plants. Limits of detection for the two derivatives are approximately 5 pmol (1.2 x 10(-9) g; 1 microM) and 0.1 pmol (3 x 10(-11) g; 20 nM) respectively. Mass spectrometer response was linear in the range from 2-200 microM DNP-MDA and 0.02-10 microM DNP-HNE. This methodology has been used to assess the formation of aldehydic secondary lipid peroxidation products in dedifferentiated callus cultures of Daucus carota. The finding that profiles of MDA and HNE can be correlated with embryogenic competence is of considerable interest as oxidative status has already been implicated as a regulatory factor in animal development. 相似文献

An in-situ corrosion sensor based on electrochemical impedance spectroscopy (EIS) has been used to detect moisture ingress into aluminum-aluminum and aluminum-composite adhesive bonds. Both wedge tests and tensile button tests (aluminum-aluminum bonds only) were performed. Upon moisture absorption, the impedance spectra change shape with the low-frequency region becoming resistive. The low-frequency impedance decreases by several orders of magnitude, depending on the adhesive and the experimental conditions. For bonds with stable interfaces, such as phosphoric acid anodized (PAA) aluminum, the absorbed moisture causes an initial weakening of the adhesive resulting in reduced strength or small crack propagation. A substantial incubation time prior to substrate hydration and bond degradation allows warning of potential joint deterioration and enables condition-based maintenance. For bonds with smooth interfaces with little or no physical bonding (mechanical interlocking), crack propagation can proceed interfacially with minimal moisture absorption. A comparison of the incubation times for Forest Products Laboratory (FPL, or sulfuric acid-sodium dichromate) etched surfaces, both bonded to epoxy adhesives and freely exposed to water or humidity at different temperatures, shows that hydration occurs with the same activation energy and, hence, the same mechanism, independent of whether or not the surface is covered with adhesive. However, the pre-exponential factor in the rate constant is dependent on the concentration of free moisture at the interface so that the hydration rate varies by several orders of magnitude. 相似文献

A number of targeted cytotoxic agents have been developed that selectively kill malignant or otherwise pathological cells. These engineered proteins consist of a potent cytotoxic element connected to a ligand domain that binds to specific molecules on the surface of the target cell. Several of these agents have shown promise in clinical trials and one is currently administered to patients. A significant technical obstacle that has impeded the development of some of these toxins is the difficulty of preparing certain recombinant proteins in properly folded forms. These fusion proteins have generally been produced in bacteria requiring them to be denatured and renatured in vitro. For some proteins this is an efficient process whereas for others it is not. We describe here a system to produce fusion toxins rapidly and efficiently by engineering mammalian cells to secrete them as properly folded molecules which can be purified in native form from cell culture medium. We have used this system to produce highly active preparations of DAB(389)-IL7, a molecule consisting of the catalytic and transmembrane domains of diphtheria toxin fused to interleukin 7. This system is generalizable and can be used to produce and evaluate rapidly fusion toxins incorporating novel or uncharacterized ligands. 相似文献

Laboratory studies were performed on the stabilization of waste activated sludge solids in a deposit formed at the bottom of an aerated water column. Conclusions reached were as follows: (1) As long as dissolved oxygen is maintained in the water column, the benthal process is stable and odor free. (2) At temperatures within the range of 20 ± 1°C, an average benthal stabilization rate of 79.4 g m−2 d−1 (biodegradable solids) can be attained if solids are not limiting. (3) Under conditions stated in conclusion (2), as much as 63% of total carbon stabilization can occur via methane formation.
