The wound is induced by several mechanical and metabolic factors. In the etiology of the wound recovery, excessive oxidative stress, calcium ion (Ca2+) influx, and apoptosis have important roles. Ca2+-permeable TRPM2 channel is activated by oxidative stress. Protective roles of Hypericum perforatum extract (HP) on the mechanical nerve injury-induced apoptosis and oxidative toxicity through regulation of TRPM2 in the experimental animals were recently reported. The potential protective roles in HP treatment were evaluated on the TRPM2-mediated cellular oxidative toxicity in the renal epithelium (MPK) cells. The cells were divided into three groups as control, wound, and wound + HP treatment (75 µM for 72 h). Wound diameters were more importantly decreased in the wound+HP group than in the wound group. In addition, the results of laser confocal microscopy analyses indicated protective roles of HP and TRPM2 antagonists (N-(p-Amylcinnamoyl) anthranilic acid and 2-aminoethyl diphenylborinate) against the wound-induced increase of Ca2+ influx and mitochondrial ROS production. The wound-induced increase of early (annexin V-FITC) apoptosis and late (propidium iodide) apoptosis were also decreased in the cells by the HP treatment. In conclusion, HP treatment acted protective effects against wound-mediated oxidative cell toxicity and apoptosis through TRPM2 inhibition. These effects may be attributed to their potent antioxidant effect.  相似文献

The long-underestimated role of extracellular vesicles in cancer is now reconsidered worldwide by basic and clinical scientists, who recently highlighted novel and crucial activities of these moieties. Extracellular vesicles are now considered as king transporters of specific cargoes, including molecular components of parent cells, thus mediating a wide variety of cellular activities both in normal and neoplastic tissues. Here, we discuss the multifunctional activities and underlying mechanisms of extracellular vesicles in neuroblastoma, the most frequent common extra-cranial tumor in childhood. The ability of extracellular vesicles to cross-talk with different cells in the tumor microenvironment and to modulate an anti-tumor immune response, tumorigenesis, tumor growth, metastasis and drug resistance will be pinpointed in detail. The results obtained on the role of extracellular vesicles may represent a panel of suggestions potentially useful in practice, due to their involvement in the response to chemotherapy, and, moreover, their ability to predict resistance to standard therapies—all issues of clinical relevance.  相似文献

Nanoporous microtubes of a nickel-copper alloy were obtained from a Cu-44Ni-1Mn (wt%) commercial wire (200 μm diameter). A new synthesis method was established through three steps: 1) partial oxidation of the wire at 1173 K in air, 2) removal of the inner unoxidized core by chemical etching, 3) reduction in 10 bar hydrogen atmosphere. During oxidation, the segregation of Cu and Ni occurred because of their different diffusion coefficients in the corresponding oxides. As a consequence, pores were formed by Kirkendall effect and due to selective chemical etching of the different oxides. Additional porosity formed because of volume contraction during reduction with hydrogen. After reduction, the microtube shows a composition gradient from the inner wall (almost pure nickel) to the outer wall (almost pure copper). The process allowed to obtain microtubes with tuneable wall thickness and inner pores around 180 ± 80 nm. The morphological features developed suggest improved capillarity properties for applications in MEMS.  相似文献

A new and simple solid‐phase extraction method for the determination of trace levels of sunset yellow has been developed on Diaion HP 2MG column system. The adsorbed sunset yellow on the resin was eluted with 3 mL of methanol and determined by UV–Vis spectrophotometry at 484 nm. Various experimental parameters such as pH, amount of sunset yellow, flow rates of sample and eluent solutions, resin amount, sample volumes, etc. were investigated. The influences of some alkali, alkali earth and transition metals on the recoveries of sunset yellow were investigated. The recovery values for sunset yellow were higher than 95%. Preconcentration factor was found as 150. The detection limit (three sigma) of the reagent blank for sunset yellow was 5.2 μg L?1. The relative standard deviations (RSD) of the procedure were found as 3.9%. The validation of the presented procedure was performed by the method of standard addition. The procedure was successfully applied to the determination of trace levels of sunset yellow in powdered beverage and confectionery products.  相似文献
