AIM: To investigate the variation and biological properties of HPV16 E7 isolated from cervical cancer biopsy samples from highest incidence area in HuBei province of China. METHODS: HVP16 E7 sequences isolated from the cervical cancer biopsies of 10 local patients were amplified, sequenced and compared with prototype E7 gene. Then the variant gene was cloned into different vectors to study the antigenicity, expression and immunogenicity of its protein by Western blot, immunofluorescence and genetic immunization in vitro or in vivo. RESULTS: The results showed that 7 of 10 samples had the same mutations which led to a nonsense mutation at codon 43 of E7 sequence. The truncated E7 protein could be recognized by standard E7 monoclonal antibody in Western blot and expressed in NIH3T3 cells. In the blood sera of mice immunized intramuscularly by the plasmid DNA expressing the variant E7 gene specific E7 antibodies could be detected at week 2, 3, 5 and 6 after inoculation. However, no specific lymphoproliferation after E7 protein stimulation in vitro was detected by MTT colorimetric assay in comparison to the prototype E7 protein. CONCLUSION: HPV16 E7 gene may show variation in China and the variant protein could be expressed and induce host humoral immune response, but could not elicit special cellular-immune response against it. We construct a novel nested linear cavity configuration to generate supercontinuum (SC) under continuous-wave pump. With this structure, stimulated Raman scattering and four-wave mixing can affect the whole region of the SC spectrum while the pump line is removed outside of the continuum, and a symmetrical spectrum can be obtained. The properties of the structure are demonstrated by using 3.5-km single-mode fiber, and an SC centered at 1315 nm with the bandwith of 102 nm and the output power of 1.3 W is achieved. By tuning the feedback fiber Bragg grating, we can easily adjust the spectral shape of SC to fit different requirement. 相似文献

Mode dependent directional transform (MDDT) can improve the coding efficiency of H.264/AVC but it also brings high computation complexity. In this paper we present a new design for implementing fast MDDT transform through integer lifting steps. We first approximate the optimal MDDT by a proper transform matrix that can be implemented with butterfly-style operation. We further factorize the butterfly-style transform into a series of integer lifting steps to eliminate the need of multiplications. Experimental results show that the proposed fast MDDT can significantly reduce the computation complexity while introducing negligible loss in the coding efficiency. We demonstrate efficient inverted polymer solar cells (PSCs) based on poly(3-hexylthiophene) (P3HT) and [6,6]-phenyl-C61-butyric acid methyl ester (PCBM) by using solution-processed titanium(IV) oxide bis(2,4-pentanedionate) (TOPD) as electron collection layer (ECL) between the indium tin oxide (ITO) electrode and photoactive layer. The TOPD buffer layer was prepared by spin-coating isopropanol solution of TOPD on ITO and then baked at 140 °C for 5 min. The power conversion efficiency (PCE) of the inverted PSC with TOPD buffer layer reaches 4% under the illumination of AM1.5G, 100 mW/cm2, which is increased by 76% in comparison with that (2.27%) of the inverted device without TOPD ECL. The results indicate that TOPD is a promising electron collection layer for inverted PSCs. 相似文献
