Oral fibroblasts stimulated invasion of oral-carcinoma cells into the collagen matrix. The mechanisms of the fibroblast-induced stimulation of invasiveness was further investigated by examining cell motility and proteolytic activity of tumor cells, using mainly an adenoid-cystic-carcinoma cell line (ACCS) and normal fibroblasts from gingival tissues. Conditioned medium from the fibroblasts grown in serum-free medium was fractionated on a Superdex 200 pg column, and Peak 1 eluted at 200 to 300 kDa and Peak 2 eluted at 50 to 100 kDa were found to contain different specific activity. Treatment of ACCS cells with Peak 1 resulted in an increase in the production of proteolytic enzymes. Peak 2 stimulated both chemotaxis and chemokinesis of ACCS cells. A chemotactic factor was purified from the heparin-unbound fraction of Peak 2 by anion exchange and hydrophobic chromatography, and was named "fibroblast-derived motility factor (FDMF)". At 1 microg/ml, FDMF stimulated chemotaxis of ACCS cells by 4-fold compared with unstimulated controls. Characterization of the physicochemical properties of FDMF suggested that it might be different from any known motility factors. Exposure of ACCS cells to FDMF resulted in reduced amounts of actin stress fiber in the cytoplasm and induction of tyrosine phosphorylation of several cellular proteins detectable 30 to 60 min after treatment. These FDMF-induced changes were blocked by pre-treatment either with genistein or with pertussis toxin. These findings suggest that FDMF may be a novel protein which stimulates cell motility via a signaling pathway mediated by a pertussis-toxin-sensitive G protein and tyrosine phosphorylation. 相似文献
BACKGROUND & AIMS: Antineutrophil cytoplasmic antibodies (ANCA) have been consistently detected in a subgroup of patients with Crohn's disease (CD). This study was designed to determine whether serum ANCA expression in patients with CD characterizes an identifiable clinical subgroup. METHODS: The study population consisted of 69 consecutive patients with an established diagnosis of CD as determined by a combination of characteristic clinical, radiographic, endoscopic, and histopathologic criteria. Sera from the patients were analyzed for the presence of ANCAs using the fixed neutrophil enzyme-linked immunosorbent assay (ELISA) assay. Perinuclear ANCA (pANCA)-positive and cytoplasmic ANCA (cANCA)-positive results by ELISA were confirmed by indirect immunofluorescence staining. Clinical profiles of the ANCA-positive patients with CD were compared with those of patients with CD not expressing ANCA (ANCA-negative). RESULTS: pANCA-positive patients with CD have endoscopically and/or histopathologically documented left-sided colitis and symptoms of left-sided colonic inflammation, clinically reflected by rectal bleeding and mucus discharge, urgency, and treatment with topical agents. One hundred percent of patients with CD expressing pANCA had "UC-like" features. CONCLUSIONS: In patients with CD, serum pANCA expression characterizes a UC-like clinical phenotype. Stratification of CD by serum pANCA provides evidence of heterogeneity within CD and suggests a common intestinal mucosal inflammatory process among a definable subgroup of patients with CD and UC expressing this marker. 相似文献

"Zero-loop" of the molecular potential transformer of submitochondrial particles (SMP) is separated from the remaining electron transfer chain by rotenone, and its e.m.f. ET=0,003+RT/2F in [NADP X H] [NAD+]/[NADP+] [NAD X H] volts is used in the compensative method of measurement of the potential difference across the SMP membrane (delta USMP). The phospholipid membrane, measuring the concentration of the penetrating anions in the solution contained SMP, is used as "zero-indicators". This concentration drops monotonically with increase in delta USMP. The culture system for in vitro evaluation of "colony forming units - culture (CFU-c)" is briefly outlined. This method offers a new approach to studies of proliferation and differentiation of hemopoietic progenitor cells, especially in disorders of granulopoiesis. From available published data it is evident that quantitation of CFU-c is also an indicator of diagnostic and prognostic value for assessment of various types of leukemia. The CFU-c assay has furthermore been introduced to test the viability and proliferating capacity of cryopreserved bone marrow, especially with a view to possible transfusion of stored autologous bone marrow as an adjuvant to cytostatic therapy. 相似文献
The problem of reconstructing a pattern of an object from its approximate discrete orthogonal projections in a 2-dimensional grid, may have no solution because the inaccuracy in the measurements of the projections may generate an inconsistent problem. To attempt to overcome this difficulty, one seeks to reconstruct a pattern with projection values having possibly some bounded differences with the given projection values and minimizing the sum of the absolute differences.
This paper addresses the problem of reconstructing a pattern with a difference at most equal to +1 or −1 between each of its projection values and the corresponding given projection value. We deal with the case of patterns which have to be horizontally and vertically convex and the case of patterns which have to be moreover connected, the so-called convex polyominoes. We show that in both cases, the problem of reconstructing a pattern can be transformed into a Satisfiability (SAT) Problem. This is done in order to take advantage of the recent advances in the design of solvers for the SAT Problem. We show, experimentally, that by adding two important features to CSAT (an efficient SAT solver), optimal patterns can be found if there exist feasible ones. These two features are: first, a method that extracts in linear time an optimal pattern from a set of feasible patterns grouped in a generic pattern (obtaining a generic pattern may be exponential in the worst case) and second, a method that computes actively a lower bound of the sum of absolute differences that can be obtained from a partially defined pattern. This allows to prune the search tree if this lower bound exceeds the best sum of absolute differences found so far. 相似文献
An important issue in risk analysis is the distinction between epistemic and aleatory uncertainties. In this paper, the use of distinct representation formats for aleatory and epistemic uncertainties is advocated, the latter being modelled by sets of possible values. Modern uncertainty theories based on convex sets of probabilities are known to be instrumental for hybrid representations where aleatory and epistemic components of uncertainty remain distinct. Simple uncertainty representation techniques based on fuzzy intervals and p-boxes are used in practice. This paper outlines a risk analysis methodology from elicitation of knowledge about parameters to decision. It proposes an elicitation methodology where the chosen representation format depends on the nature and the amount of available information. Uncertainty propagation methods then blend Monte Carlo simulation and interval analysis techniques. Nevertheless, results provided by these techniques, often in terms of probability intervals, may be too complex to interpret for a decision-maker and we, therefore, propose to compute a unique indicator of the likelihood of risk, called confidence index. It explicitly accounts for the decision-maker's attitude in the face of ambiguity. This step takes place at the end of the risk analysis process, when no further collection of evidence is possible that might reduce the ambiguity due to epistemic uncertainty. This last feature stands in contrast with the Bayesian methodology, where epistemic uncertainties on input parameters are modelled by single subjective probabilities at the beginning of the risk analysis process. 相似文献
The analytical parameters for the measurement of mercury in air have been investigated for the integrated silver method. The ultimate sensitivity of the method and the reproducibility of the standard curve are discussed. The proper storage of the activated silver absorber is described, as well as the storage of a mercury-air sample on the silver element.The collection efficiency for mercury has been found to be quantitatively reversible, with respect to both capture and release. The collection process on silver is highly specific for mercury. Benzene, for example, is not retained on silver, though it can give a spurious response when sampled directly by a “continuous” ultraviolet photometer. Most commonly encountered organic vapors, which give a response at mercury absorption wavelengths, are not retained by the silver element and do not, therefore, interfere with subsequent ultraviolet measurement of the desorbed mercury vapor.For ambient air sampling, the optimum flow-rate was found to be 1.51/min. This also proved to be a suitable flow-rate for desorption and assay. Pure air, as carrier, has been found to be satisfactory, since no differences have been noticed when comparative measurements have been made using inert gases.Instrumental measurement by means of a digital integrator has been found to be superior to a recorder output using peak height. Peak area measurement, by integrator, is not affected by the rate of diffusion of the mercury vapor within the sampling system and optical train. 相似文献
Polymerization of β-butyrolactone has been studied in toluene with Al(OiPr)3 as an initiator. The ring-opening polyaddition proceeds through a coordination–insertion mechanism at a very low rate. Well defined α-isopropylester, ω-hydroxy poly(β-butyrolactone)s (PBL) are formed with a narrow molecular weight distribution at low monomer-to-initiator molar ratios, When this ratio is higher (ca. 170), a competition occurs between propagation and side reactions, i.e. elimination, inter- and intra-molecular transesterifications and thermal degradation, which is responsible for a loss of control of the PBL molecular charcteristics. The addition of a Lewis base (1 equivalent of nicotine/Al) to the Al-alkoxide initiator has no significant effect on the polymerization rate, although the chain microstructure is deeply affected since predominantly syndiotactic PBL chains are formed (63% syndio-diads) in contrast to a completely atactic polymer in the absence of nicotine. 相似文献
