Genograms help reveal patterns of events and relationships in family histories. A variation of the standard genogram, the time-line genogram, uses a time scale as a vertical axis to show individuals and events directly in their proper temporal relationships (H. Friedman et al, 1988). After a brief training session, undergraduates with negligible experience in clinical matters recorded family data in the standard or time-line genogram formats or by taking notes. The 3 groups were equally successful in recording and interpreting family information. Although the time-line group reported more difficulty in recording, perhaps in reaction to the greater visual complexity of the time-line format, this group was best at recording events linked in time. When a complete graphic display of temporal patterns and relationships is needed, the time-line genogram may be a practical alternative to the standard format. Synthesis of the Vibrio fischeri autoinducer, a signal involved in the cell density-dependent activation of bioluminescence, is directed by the luxI gene product. The LuxI protein catalyzes the synthesis of N-acyl-homoserine lactones from S-adenosylmethionine and acylated-acyl carrier protein. We have gained an appreciation of the LuxI regions and amino acid residues involved in autoinducer synthesis by isolating and analyzing mutations generated by random and site-specific mutagenesis of luxI. By random mutagenesis we isolated 13 different single amino acid substitutions in the LuxI polypeptide. Eleven of these substitutions resulted in no detectable autoinducer synthase activity, while the remaining two amino acid substitutions resulted in reduced but detectable activity. The substitutions that resulted in no detectable autoinducer synthase activity mapped to two small regions of LuxI. In Escherichia coli, wild-type luxI showed dominance over all of the mutations. Because autoinducer synthesis has been proposed to involve formation of a covalent bond between an acyl group and an active-site cysteine, we constructed site-directed mutations that altered each of the three cysteine residues in LuxI. All of the cysteine mutants retained substantial activity as an autoinducer synthase in E. coli. Based on the analysis of random mutations we propose a model in which there are two critical regions of LuxI, at least one of which is an intimate part of an active site, and based on the analysis of site-directed mutations we conclude that an active-site cysteine is not essential for autoinducer synthase activity.  相似文献

A variable threshold voltage keeper circuit technique is proposed for simultaneous power reduction and speed enhancement of domino logic circuits. The threshold voltage of a keeper transistor is dynamically modified during circuit operation to reduce contention current without sacrificing noise immunity. The variable threshold voltage keeper circuit technique enhances circuit evaluation speed by up to 60% while reducing power dissipation by 35% as compared to a standard domino (SD) logic circuit. The keeper size can be increased with the proposed technique while preserving the same delay or power characteristics as compared to a SD circuit. The proposed domino logic circuit technique offers 14% higher noise immunity as compared to a SD circuit with the same evaluation delay characteristics. Forward body biasing the keeper transistor is also proposed for improved noise immunity as compared to a SD circuit with the same keeper size. It is shown that by applying forward and reverse body biased keeper circuit techniques, the noise immunity and evaluation speed of domino logic circuits are simultaneously enhanced.  相似文献

Flash photolysis experiments have been carried out for the first time on a hemoglobin ferrous cyanide adduct with an 8 ns laser pulse. A 95% nonexponential rebinding process occurs within 2 micros after full photolysis in ferrous cyanide dimeric Scapharca inaequivalvis hemoglobin (HbI), indicating that once photolyzed the cyanide anion is not able to escape from the protein matrix and rebinds to the heme iron. The resonance Raman spectrum of the 10 ns photoproduct is identical to that of the fully relaxed deoxy derivative, indicating that in the ferrous cyanide HbI adduct protein relaxation occurs within 10 ns after photolysis. This behavior is at variance with that of the carbonmonoxy HbI derivative in which very little geminate rebinding is observed and the photoproduct relaxes with a lifetime of 1 micros. The fast relaxation of the cyanide HbI photoproduct can be accounted for by the small perturbation of the heme structure induced by cyanide binding to ferrous HbI. This is consistent with a deoxy-like conformation of the HbI ferrous cyanide adduct and implies that the pathway for relaxation involves only minor local rearrangements of the heme moiety. Photolysis experiments carried out on ferrous cyanide horse myoglobin, which can be saturated only partially, show a qualitatively similar behavior in ligand rebinding, indicating that the geminate process of the cyanide anion is a general phenomenon in hemoproteins.  相似文献

Different types of databases available for health-related research, the data contained in these databases, and potential applications for pharmacists or researchers are discussed. Case studies that demonstrate uses for health databases are presented. Databases can be organized by facility, by health care provider, by disease or organ, or by sector. The types of data they contain include financial data, utilization data, demographic data, and outcomes data. Data can be obtained from the public sector, the private sector, or the researcher's own health system. The costs and time associated with using existing databases are often less than those required to collect data, but the quality and accessibility of the data must also be considered. The researcher's choice of database will depend on the research question. Health care databases can be used for health management and decision-making, quality review and evaluation, outcomes research, episode-of-illness studies, and evaluation of treatment protocols. Researchers must comply with patient-confidentiality and other agreements when accessing data. The format of the data needs to be matched with the hardware and software to be used in the analysis, and the data need to be loaded, verified, and cleaned before use. In deciding which of the many available data sources to use, researchers must determine the appropriate balance between external data and data available within their own health systems. The decision on whether to use existing data sources or to collect data prospectively will depend on the research question, the available resources, and the scope of the study.  相似文献

Multiple power supply voltages are often used in modern high-performance ICs, such as microprocessors, to decrease power consumption without affecting circuit speed. To maintain the impedance of a power distribution system below a specified level, multiple decoupling capacitors are placed at different levels of the power grid hierarchy. The system of decoupling capacitors used in power distribution systems with multiple power supplies is described in this paper. The noise at one power supply can propagate to the other power supply, causing power and signal integrity problems in the overall system. With the introduction of a second power supply, therefore, the interaction between the two power distribution networks should be considered. The dependence of the impedance and magnitude of the voltage transfer function on the parameters of the power distribution system is investigated. An antiresonance phenomenon is intuitively explained in this paper. It is shown that the magnitude of the voltage transfer function is strongly dependent on the parasitic inductance of the decoupling capacitors, decreasing with smaller inductance. Design techniques to cancel and shift antiresonant spikes out of range of the operating frequencies are presented. It is also shown that it is highly desirable to maintain the effective series inductance of the decoupling capacitors as low as possible to decrease the overshoots of the response of the dual-voltage power distribution system over a wide range of operating frequencies. A criterion for an overshoot-free voltage response is presented in this paper. It is noted that the frequency range of the overshoot-free voltage response can be traded off with the magnitude of the response.  相似文献

This article describes an intensive, inpatient behavioral rehabilitation program for patients with schizophrenia who have been considered "treatment-refractory" at state hospitals. The program is a public-private partnership involving state and private hospitals and community residence providers. The essential elements of this program are described, along with the conceptual and philosophical bases of its treatment and examples of staff behaviors critical to treatment success. Outcome data are then discussed to emphasize the point that when evidence-based psychological treatment is implemented with this population, outcomes can be positive in most cases, and therefore, the number of treatment-refractory patients is actually far less than is estimated on the basis of response to medication alone.
