To better understand the nature of the mechanism involved in the membrane uptake of a vector peptide, the interactions between dioleoylphosphatidylcholine and a primary amphipathic peptide containing a signal peptide associated with a nuclear localization sequence have been studied by isotherms analysis of mixed monolayers spread at the air-water interface. The peptide and the lipid interact through strong hydrophobic interactions with expansion of the mean molecular area that resulted from a lipid-induced modification of the organization of the peptide at the interface. In addition, a phase separation occurs for peptide molar fraction ranging from about 0.08 to 0.4 Atomic force microscopy observations made on transferred monolayers confirm the existence of phase separation and further reveal that mixed lipid-peptide particles are formed, the size and shape of which depend on the peptide molar fraction. At low peptide contents, round-shaped particles are observed and an increase of the peptide amount, simultaneously to the lipidic phase separation, induces morphological changes from bowls to filamentous particles. Fourier transform infrared spectra (FTIR) obtained on transferred monolayers indicate that the peptide adopts a beta-like structure for high peptide molar fractions. Such an approach involving complementary methods allows us to conclude that the lipid and the peptide have a nonideal miscibility and form mixed particles which phase separate.  相似文献

Many enteroviruses, members of the family Picornaviridae, cause a rapid and drastic inhibition of host cell protein synthesis during infection, a process referred to as host cell shutoff. Poliovirus, one of the best-studied enteroviruses, causes marked inhibition of host cell translation while preferentially allowing translation of its own genomic mRNA. An abundance of experimental evidence has accumulated to indicate that cleavage of an essential translation initiation factor, eIF4G, during infection is responsible at least in part for this shutoff. However, evidence from inhibitors of viral replication suggests that an additional event is necessary for the complete translational shutoff observed during productive infection. This report examines the effect of poliovirus infection on a recently characterized 3' end translational stimulatory protein, poly(A)-binding protein (PABP). PABP is involved in stimulating translation initiation in lower eukaryotes by its interaction with the poly(A) tail on mRNAs and has been proposed to facilitate 5'-end-3'-end interactions in the context of the closed-loop translational model. Here, we show that PABP is specifically degraded during poliovirus infection and that it is cleaved in vitro by both poliovirus 2A and 3C proteases and coxsackievirus B3 2A protease. Further, PABP cleavage by 2A protease is accompanied by concurrent loss of translational activity in an in vitro-translation assay. Similar loss of translational activity also occurs simultaneously with partial 3C protease-mediated cleavage of PABP in translation assays. Further, PABP is not degraded during infections in the presence of guanidine-HCl, which blocks the complete development of host translation shutoff. These results provide preliminary evidence that cleavage of PABP may contribute to inhibition of host translation in infected HeLa cells, and they are consistent with the hypothesis that PABP plays a role in facilitating translation initiation in higher eukaryotes.  相似文献

Money laundering has been affecting the global economy for many years. Large sums of money are laundered every year, posing a threat to the global economy and its security. Money laundering encompasses illegal activities that are used to make illegally acquired funds appear legal and legitimate. This paper aims to provide a comprehensive survey of machine learning algorithms and methods applied to detect suspicious transactions. In particular, solutions of anti-money laundering typologies, link analysis, behavioural modelling, risk scoring, anomaly detection, and geographic capability have been identified and analysed. Key steps of data preparation, data transformation, and data analytics techniques have been discussed; existing machine learning algorithms and methods described in the literature have been categorised, summarised, and compared. Finally, what techniques were lacking or under-addressed in the existing research has been elaborated with the purpose of pinpointing future research directions.  相似文献

The aim of the present study was to evaluate the efficacy of different methods of sulphide microencapsulation in terms of inhibiting sulphide oxidation in roofing slate. For this, a preliminary test was carried out with ground pyrite and the methods that provided the best results were applied to samples of roofing slate. Protection against oxidation was measured by the amount of iron released after enforced oxidation with H2O2 in the tests with pyrite, and by thermal cycle alteration, SO2 and saline spray tests (EN 12326-2:2000 and EN 14147:2004) in the tests with the slate samples. The results indicate that treatments with potassium phosphate and potassium silicate, proposed for controlling acid mine drainage, were the most effective at protecting pyrite against oxidation, and that these methods are also effective when applied to slate.  相似文献

Abstract Software documentation for the novice user typically must try to achieve at least three goals: to support basic knowledge and skills development; to prevent or support the handling of mistakes, and to support the joint handling of manual, input device and screen. This paper concentrates on the latter goal. Novice users often experience split‐attention problem due to the need to (almost) simultaneously attend to different media. Existing research indicates that split‐attention problems can be prevented or reduced by the presence of screen images in the manual. Research is yet unclear about the optimal design of these pictures. This study examines three design styles. Forty‐eight novice users received one of the three manual based on these styles. The manuals were an introduction to Windows 95. The users of the most successful manual needed 25% less training time and had a 60% better retention. The most important characteristics of the design style of this manual were its use of full screen images (instead of partial ones) and a two‐column lay‐out in which the instructions and screen images were presented side‐by‐side in a left‐to‐right reading order. The discussion focuses on the tension that exists between theory and practice. Special attention is given to the contributions of a taxonomy of screen images and cognitive load theory.  相似文献

Using numerical continuation we show a new bifurcation scenario involving resonant periodic orbits in a parametrized four-dimensional autonomous system deriving from nonlinear rotordynamics. The scenario consists of a carefully orchestrated sequence of transcritical bifurcations in which branches of periodic solutions are exchanged. Collectively, the bifurcations resemble the action of a zipper. An underlying governing mechanism clearly exists but still has to be uncovered. For a range of parameter values the sequence of bifurcations forms a global connection between a Sil'nikov bifurcation and (partial) mode-locking. The homoclinic bifurcation is introduced into the system by a Takens-Bogdanov bifurcation. The system also features an interaction between two chaotic Sil'nikov bifurcations.  相似文献

The present work continues the series of experimental investigations undertaken in order to elucidate the mechanisms controlling elastic and anelastic properties of the β1′ martensitic phase of Cu-based shape memory alloys. The paper reports an attempt to distinguish between ‘dislocation’ and ‘interface’ mechanisms of the internal friction in the β1′ martensitic phase of Cu–Al–Ni single crystals. Two types of experiments have been performed. First, the ultrasonic strain amplitude-independent and amplitude-dependent internal friction (ADIF) of a monovariant specimen for temperatures 90–300 K is carefully re-examined. Second, in situ measurements of the ADIF and of the influence of ultrasonic oscillations on the plastic deformation (acoustoplastic effect) were carried out during quasistatic deformation of a quenched polyvariant specimen. Experimental results support a dislocation rather than an interface mechanism of anelasticity, at least at ultrasonic frequencies and moderate strain amplitudes.  相似文献
