Three-dimensional ultrastructural analysis of the Saccharomyces cerevisiae mitotic spindle

The three dimensional organization of microtubules in mitotic spindles of the yeast Saccharomyces cerevisiae has been determined by computer-aided reconstruction from electron micrographs of serially cross-sectioned spindles. Fifteen spindles ranging in length from 0.6-9.4 microns have been analyzed. Ordered microtubule packing is absent in spindles up to 0.8 micron, but the total number of microtubules is sufficient to allow one microtubule per kinetochore with a few additional microtubules that may form an interpolar spindle. An obvious bundle of about eight interpolar microtubules was found in spindles 1.3-1.6 microns long, and we suggest that the approximately 32 remaining microtubules act as kinetochore fibers. The relative lengths of the microtubules in these spindles suggest that they may be in an early stage of anaphase, even though these spindles are all situated in the mother cell, not in the isthmus between mother and bud. None of the reconstructed spindles exhibited the uniform populations of kinetochore microtubules characteristic of metaphase. Long spindles (2.7-9.4 microns), presumably in anaphase B, contained short remnants of a few presumed kinetochore microtubules clustered near the poles and a few long microtubules extending from each pole toward the spindle midplane, where they interdigitated with their counterparts from the other pole. Interpretation of these reconstructed spindles offers some insights into the mechanisms of mitosis in this yeast.     

A new distinctive variation of renal arterial vascularization

In addition to the usual renal arteries, a bilateral artery branching from the aorta was found connecting the aorta to both kidneys in an 82 year-old Caucasian man. By creating an additional blood supply to the kidneys this artery may have had an effect on renal perfusion.     

Factors affecting primary care physician participation in Medicare

OBJECTIVES: This study investigates the levels of participation and the relative association of economic and noneconomic factors on primary care physician participation in the Medicare program. METHODS: Demographic information, participation in Medicare, and attitudes toward both the Medicare program and Medicare patients were collected in a written survey mailed to half the primary care physicians in Iowa. Ordinary least squares and logistic regression analyses were conducted to determine factors associated with the percentage of Medicare patients in a practice and the acceptance of all new Medicare patients, respectively. RESULTS: Two thirds of physicians were accepting all new Medicare patients, whereas 16% were accepting no new Medicare patients. Factors associated with having a higher percentage of Medicare patients in a practice were as follows: (1) a larger proportion of Medicare recipients in the county, (2) practice as a general internal medicine physician, (3) more years in practice at the current location, (4) greater enjoyment treating elderly patients, (5) less concern about having too many Medicare patients, and (6) a stronger belief that the Medicare program respects their professional judgment. Physicians less concerned about having too many Medicare patients in their practice and physicians in counties with a higher percentage of Medicare patients were significantly more likely to accept all new Medicare patients. CONCLUSIONS: These results suggest that as Medicare reforms are discussed, careful consideration of the impact of these reforms on noneconomic issues is important to ensure adequate physician participation and access for elderly patients through the Medicare program.     

Pseudotaxonicity in MAMBAC and MAXCOV analyses of rating-scale data: Turning continua into classes by manipulating observer's expectations.

Taxometric procedures such as mean above minus below a cut and maximum covariance can determine whether a trait is distributed as a discrete latent class. These methods have been used to infer taxonic structure in several personality and psychopathology constructs, often from analyses of rating scale data. This is problematic given (a) well established biases in ratings, (b) the human tendency to think categorically, and (c) implicit typological models of personality and psychopathology among expert raters. Using an experimental method in which the cognitive sets of raters were manipulated as dimensional versus categorical, it is demonstrated that pseudotaxonicity can be created readily with rating scale measures. This suggests that researchers avoid an exclusive reliance on rating scales when conducting taxometrics investigations. (PsycINFO Database Record (c) 2010 APA, all rights reserved)     

In Search of a Common Core: A Content Analysis of Interventions to Promote Forgiveness.

This article reviews published methods for promoting forgiveness for a broad range of clinical issues. The review revealed a consensus among applied researchers regarding several broad types of interventions to promote forgiveness, namely, (a) defining forgiveness, (b) helping clients remember the hurt, (c) building empathy in clients for the perpetrator, (d) helping clients acknowledge their own past offenses, and (e) encouraging commitment to forgive the offender. Roughly half of the studies also prescribed interventions to help clients overcome unforgiveness (e.g., bitterness, vengefulness) without explicitly promoting forgiveness. Speculations about how to use forgiveness interventions in sensitive and client-supportive ways are advanced on the basis of the findings. Modeling of static concentrator modules incorporating lambertian or v-groove rear reflectors

Ray tracing modeling has been used to calculate the performance of static concentrator modules with a geometric concentration ratio of 2 and incorporating very narrow (1–2 mm), long and bifacial cells. The modules utilize either a v-groove or a lambertian rear reflector. It is shown that the use of very narrow cells allows a performance improvement of 5% or more compared to structures incorporating wider cells, in the case of v-groove reflectors. The averaged yearly performance for both types of reflectors is found to be rather similar, with expected light collection in the range 82–86% of that of a module with 100% cell coverage. Experimental measurements on modules with lambertian reflectors are shown to be in good agreement with the results of modeling.     

Systematic environmental, direct, and service sire effects on conception rate in artificially inseminated Holstein cows

Result of insemination was verified for 329,314 artificial inseminations by 882 service sires to 97,245 Holstein cows in 1,075 herds between May 1970 and December 1983. Estimates of systematic environmental and genetic components of cow and service sire components of conception rate were obtained preliminary to development of a fertility monitoring system. Conception rate was 2.9% higher in stanchion than loose housed herds and 2.3% higher in grade than registered cows. Fall months were superior to winter months, the greatest difference being 6.1% between October and January. Conception rate increased with herd milk production, decreased with both increased cow age, and increased herd size in mature and old cows. Regions and inseminators within regions were highly variable. Conception may be influenced by semen price; however, week day of insemination and duration of semen storage had no effect. Conception rate decreased for semen by bulls 8 yr and older, was lowest for semen harvested in June, but no season of collection effect was detected. No genetic trends for cow and service sire conception rate were found; heritability and repeatability were .08 and .06, respectively. Genetic correlations between cow and service sire conception rate and these components with first lactation production and semen output measures were all near zero. Therefore, the relationship between sire fertility and daughter fertility is near zero.     

Approach and avoidance strength during goal attainment: regulatory focus and the "goal looms larger" effect

Three studies tested the hypothesis that regulatory focus, both as a chronic person variable and as a manipulated situational variable, differentially affects the strength of participants' approach and avoidance strategic motivations as they get closer to the goal. In 2 studies, flexion and extension arm pressure were used as on-line measures of approach and avoidance intensity, respectively. As predicted, the approach gradient was steeper for participants with a promotion focus on aspirations and gains than for participants with a prevention focus on responsibilities and nonlosses, whereas the reverse was true for the avoidance gradient. In a third study, the same pattern of results was found on a persistence measure of motivational strength. Participants with a promotion focus worked longer on anagrams closer to the goal when they were approach means for goal attainment than when they were avoidance means, whereas the reverse was true for participants with a prevention focus.
