The global dynamic response of floating wind turbines is commonly simulated using aero‐hydro‐servo‐elastic tools that consider numerically generated wind files as input. Based on the guidelines given in existing standards, two methods of generating such wind files are typically used: the Mann uniform shear model and the Kaimal spectral and exponential coherence model. Both models consider the Kaimal spectrum with similar frequency characteristics: The main difference between the approaches is related to the spatial coherence. The present work examines the consequences of using the two different wind file generation methods for estimating the global responses of representative spar, semi‐submersible, and tension leg platform (TLP) 5 MW wind turbines. Predictions of the standard deviation of low‐frequency responses in operational conditions, including motions at the natural frequency in surge and pitch as well as quasi‐static (spar, TLP) and resonant (semi‐submersible) motions in yaw, are seen to differ up to 30% to 40% depending on the wind field model. The differences in motion responses have important consequences for the design of the mooring system components. Proper orthogonal decomposition (POD) techniques are used to qualitatively explain the differing spatial coherence of the wind field. Simulations including a limited number of POD modes in the wind field highlight the importance of the low‐frequency, energy‐rich modes, suggesting that this type of visualization can be a useful tool. 相似文献

One promising strategy to combat antibiotic-resistant bacteria is to develop compounds that block bacterial defenses against antibacterial conditions produced by the innate immune system. Salmonella enterica, which causes food-borne gastroenteritis and typhoid fever, requires histidine kinases (HKs) to resist innate immune defenses such as cationic antimicrobial peptides (CAMPs). Herein, we report that 2-aminobenzothiazoles block histidine kinase-dependent phenotypes in Salmonella enterica serotype Typhimurium. We found that 2-aminobenzothiazoles inhibited growth under low Mg2+, a stressful condition that requires histidine kinase-mediated responses, and decreased expression of the virulence genes pagC and pagK. Furthermore, we discovered that 2-aminobenzothiazoles weaken Salmonella’s resistance to polymyxin B and polymyxin E, which are last-line antibiotics and models for host defense CAMPs. These findings raise the possibilities that 2-aminobenzothiazoles can block HK-mediated bacterial defenses and can be used in combination with polymyxins to treat infections caused by Salmonella. 相似文献

Conditions of chemical separation of Al macroamounts and U microamounts, followed by atomic emission spectroscopic determination of the isotope composition and gravimetric content of U, were studied. An algorithm was developed for constructing calibration plots to determine the 235U/238U isotope ratio in various samples from nuclear fuel reprocessing. Optimum conditions of the spectral analysis for the U content were found by mathematical design of the experiment: NaCl content 2%, current 18 А, and exposure time 40 s. Highly-filled polymer systems include color masterbatches, feedstocks for powder injection molding, and rigid sheets with high levels of flame retardants, but they have not been explored for flexible sheet. This work investigated the (a) selecting a polymer matrix with enough melt strength and flexibility to form a stable sheet with high filler loading, (b) the maximum achievable filler loading for the sheet, and (c) optimizing the process of extruding a highly-filled flexible polymer system. Extrusion grade low-density polyethylene (LDPE) provided sufficient flexibility and permitted a maximum filler loading of 36 vol% (~78 wt%). Good dispersion of the nanoparticle filler, however, required two passes through multiple screw extruders and a small reduction in the viscosity of the LDPE. Sheet with thickness of 415 μm, surface roughness of <1 μm, and sufficient flexibility was extruded continuously at a rate of 10 m/min., but it required a more traditional coat hanger manifold to prevent filler hang up in the sheet die. Enhanced understanding of neuropathologies has created a need for more advanced tools. Current neural implants result in extensive glial scarring and are not able to highly localize drug delivery due to their size. Smaller implants reduce surgical trauma and improve spatial resolution, but such a reduction requires improvements in device design to enable accurate and chronic implantation in subcortical structures. Flexible needle steering techniques offer improved control over implant placement, but often require complex closed‐loop control for accurate implantation. This study reports the development of steerable microinvasive neural implants (S‐MINIs) constructed from borosilicate capillaries (OD = 60 µm, ID = 20 µm) that do not require closed‐loop guidance or guide tubes. S‐MINIs reduce glial scarring 3.5‐fold compared to prior implants. Bevel steered needles are utilized for open‐loop targeting of deep‐brain structures. This study demonstrates a sinusoidal relationship between implant bevel angle and the trajectory radius of curvature both in vitro and ex vivo. This relationship allows for bevel‐tipped capillaries to be steered to a target with an average error of 0.23 mm ± 0.19 without closed‐loop control. Polished microcapillaries present a new microinvasive tool for chronic, predictable targeting of pathophysiological structures without the need for closed‐loop feedback and complex imaging. 相似文献

Alzheimer's disease is characterized in part by the aggregation of tau protein into filamentous inclusions. Because tau filaments form in brain regions associated with memory retention, and because their appearance correlates well with the degree of dementia, they have emerged as robust markers of disease progression. Yet the discovery that mutations in tau protein can lead directly to filament and tangle formation in humans, and that filament formation is linked to neurodegeneration in model biological systems, suggests that tau aggregation may also contribute directly to degeneration in affected neurons. In this context, the mechanism of tau filament formation and its modulation by mutation and posttranslational modification is of fundamental importance. Here, recent progress on the molecular mechanisms underlying tau aggregation deduced from in vivo and in vitro experimentation is reviewed and a model rationalizing the effect of posttranslational and other structural modifications on assembly kinetics and thermodynamics is presented. We hypothesize that tau aggregation can be described as a heterogeneous nucleation reaction, where exogenous effectors, tau gene mutations, or other modifications that stabilize assembly-competent conformations of tau act to trigger the fibrillization reaction. In contrast, those that modulate postnuclear equilibria can enhance fibrillization by increasing the free energy difference between polymers and unincorporated monomers, resulting in stabilization of filaments at low bulk protein concentrations. 相似文献

Breeding birds and anurans (frogs and toads) in coastal wetlands of Green Bay, Lake Michigan vary dynamically with changing water levels, habitat type, and geography. We describe species assemblages over a seven-year period (2011–2017) beginning with historic low water levels followed by an increase in average lake level of 0.85?m. In general, species richness and abundance of marsh-obligate species responded positively to increasing water levels, although several species of shallow wetlands (sandhill crane, sedge wren, swamp sparrow, and American toad) showed the opposite trend. Anuran assemblages were more diverse in the middle and upper bay, coinciding with a well-established nutrient gradient from the hypereutrophic lower bay to more oligotrophic waters of the upper bay. Three marsh-obligate bird species (black tern, sandhill crane, and sedge wren) were significantly more abundant in the middle or upper bay while sora, American coot, and common gallinule were more abundant in the eutrophic lower bay. Our findings have several important implications for conservation. Inland wetlands near the coast (including diked wetlands) might play a key ecological role by providing refugia for some species during low water years. However, the importance of shallow coastal wetlands and nearshore gradients of wetland habitat might be overlooked during low water years; when high water returns, these areas can become extremely productive and species-rich. 相似文献
