Deletion of PHR1, a pH-regulated gene of Candida albicans, results in pH-conditional defects in growth, morphogenesis, and virulence evident at neutral to alkaline pH but absent at acidic pH. Consequently, we searched for a functional homolog of PHR1 active at low pH. This resulted in the isolation of a second pH-regulated gene, designated PHR2. The expression of PHR2 was inversely related to that of PHR1, being repressed at pH values above 6 and progressively induced at more acidic pH values. The predicted amino acid sequence of the PHR2 protein, Phr2p, was 54% identical to that of Phr1p. A PHR2 null mutant exhibited pH-conditional defects in growth and morphogenesis analogous to those of PHR1 mutants but manifest at acid rather than alkaline pH values. Engineered expression of PHR1 at acid pH in a PHR2 mutant strain and PHR2 at alkaline pH in a PHR1 mutant strain complemented the defects in the opposing mutant. Deletion of both PHR1 and PHR2 resulted in a strain with pH-independent, constitutive growth and morphological defects. These results indicate that PHR1 and PHR2 represent a novel pH-balanced system of functional homologs required for C. albicans to adapt to environments of diverse pH.  相似文献

A necessary and sufficient condition for the union of an arbitrary family of fuzzy subgroups of a group to be a fuzzy group has been proposed. The criterion which gives a cut of the unit interval is called the Schnittaxiom. A modified CIECAM02 colour appearance model, named CIECAM02‐m2, is proposed to enable CIECAM02 to predict the simultaneous contrast effect. The structure of the CIECAM02‐m2 is a development from CIECAM02, and contains two different procedures for modifying the reference white; one is for lightness and the other is for hue. The model was tested using a data set accumulated in this study and the LUTCHI data. The CV values for three colour attributes between predictions and experimental data were used to evaluate the accuracy of the model. The low CV values obtained show the performance of the CIECAM02‐m2 model to predict the simultaneous contrast effect satisfactorily. © 2007 Wiley Periodicals, Inc. The concentrations of several chemical metabolites in Cheddar cheese were monitored by various chromatographic techniques during the aging process to learn which metabolites were the best predictors of the glycolytic, lipolytic, and proteolytic age of the cheese. Pyruvic, lactic, acetic, and propionic acids were measured by ion-exchange high performance liquid chromatography; acetone, 2-butanone, ethanol, 2-pentanone, 2-butanol, and n-propanol were monitored by headspace gas chromatography; free fatty acids were quantitated (without derivatization) by gas chromatography; and free amino acids were determined as their o-phthaldehye derivatives by high performance liquid chromatography.The best predictors of the glycolytic age were propionic acid and acetic acid; the best predictors of lipolysis were the free fatty acids C10, C12, C14, and C16; and the best predictors of proteolysis were the free amino acids leucine, methionine, and glutamic acid. The volatile metabolites determined by headspace gas chromatography were not good indicators of aging; however, they did provide useful information related to flavor problems.Cheddar cheese aged at elevated temperatures produced propionic acid, acetic acid, and free amino acids at significantly faster rates than the other chemicals that were monitored.  相似文献

The purpose of this study was to compare probing depth resolution achieved by gingivectomy and periodontal flap techniques in the treatment of cyclosporine A- and nifedipine-induced gingival enlargement. Ten kidney transplant patients who were receiving cyclosporine A and nifedipine for at least 6 months participated in the study. Five patients were randomly assigned to the gingivectomy group and 5 patients to the periodontal flap group. Only anterior segments of the oral cavity (canine to canine) were surgically treated. Clinical measurements, including probing depths, plaque index, and gingival sulcus index, were taken at baseline, 6 weeks, 6 months, and 1 year. Results showed that probing depths, while similar for both groups in the first 6 weeks of the study, were significantly shallower for the periodontal flap group when compared to the gingivectomy group at 6 months (2.48 +/- 0.34 mm versus 4.87 +/- 0.79 mm, respectively) and 1 year (322 +/- 0.65 mm versus 6.40 +/- 1.02 mm, respectively). Within its limitations, this study suggests that the pocket reduction achieved by the periodontal flap may be sustained for longer periods of time than by the gingivectomy technique in the treatment of cyclosporine A- and nifedipine-induced gingival enlargement.  相似文献

We developed surface‐modified silica fillers by coating these with an acrylate monomer, trimethylolpropane triacrylate, or a silane coupling agent, triethoxyvinyl silane, followed by electron‐beam irradiation at room temperature. These were incorporated in an ethylene–octene copolymer rubber. Thermorheological studies of the unvulcanized ethylene–octene copolymer and its untreated and modified silica‐filled composites were done with a shear dynamic oscillating rheometer. Modification of the silica filler, especially via the silanization process followed by electron beam treatment, significantly reduced filler–filler networking as revealed from the log–log plots of storage modulus and complex shear viscosity, and its real component. The rheological complexity of the compositions was analyzed from a double logarithmic plot of the storage modulus and loss modulus. The results obtained from the master curves constructed on the basis of the time–temperature superposition principle and the activation energy calculated from the Arrhenius equation for the flow of above these compounds further supported these findings. © 2003 Wiley Periodicals, Inc. This paper formulates a self-organization algorithm to address the problem of global behavior supervision in engineered swarms of arbitrarily large population sizes. The swarms considered in this paper are assumed to be homogeneous collections of independent identical finite-state agents, each of which is modeled by an irreducible finite Markov chain. The proposed algorithm computes the necessary perturbations in the local agents' behavior, which guarantees convergence to the desired observed state of the swarm. The ergodicity property of the swarm, which is induced as a result of the irreducibility of the agent models, implies that while the local behavior of the agents converges to the desired behavior only in the time average, the overall swarm behavior converges to the specification and stays there at all times.
