The ability of the adenovirus type 5 E1B 55-kDa mutants dl1520 and dl338 to replicate efficiently and independently of the cell cycle, to synthesis viral DNA, and to lyse infected cells did not correlate with the status of p53 in seven cell lines examined. Rather, cell cycle-independent replication and virus-induced cell killing correlated with permissivity to viral replication. This correlation extended to S-phase HeLa cells, which were more susceptible to virus-induced cell killing by the E1B 55-kDa mutant virus than HeLa cells infected during G1. Wild-type p53 had only a modest effect on E1B mutant virus yields in H1299 cells expressing a temperature-sensitive p53 allele. The defect in E1B 55-kDa mutant virus replication resulting from reduced temperature was as much as 10-fold greater than the defect due to p53 function. At 39 degreesC, the E1B 55-kDa mutant viruses produced wild-type yields of virus and replicated independently of the cell cycle. In addition, the E1B 55-kDa mutant viruses directed the synthesis of late viral proteins to levels equivalent to the wild-type virus level at 39 degreesC. We have previously shown that the defect in mutant virus replication can also be overcome by infecting HeLa cells during S phase. Taken together, these results indicate that the capacity of the E1B 55-kDa mutant virus to replicate independently of the cell cycle does not correlate with the status of p53 but is determined by yet unidentified mechanisms. The cold-sensitive nature of the defect of the E1B 55-kDa mutant virus in both late gene expression and cell cycle-independent replication leads us to speculate that these functions of the E1B 55-kDa protein may be linked.  相似文献

Myofibroblastoma (MF) is an uncommon, usually benign, mesenchymal tumor infrequently described in soft tissues. We report here on the clinicopathologic findings of a soft tissue MF (STMF) presenting in the neck of a 90-year-old man as a slowly growing and non-painful nodule, 4 cm in greatest diameter. Histology revealed a circumscribed lesion constituted of monomorphous bipolar spindle cells arranged in swirling fascicles with intervening broad bands of hyalinized collagen and well formed "amianthoid" fibers. Immunohistochemistry showed the spindle cells to be immunoreactive for vimentin, smooth muscle actin and muscle specific actin and, focally, for desmin; immunostaining for cytokeratin, epithelial membrane antigen, S-100 protein, factor VIII-related antigen, and CD34 was negative. Based on the present case and on those previously reported in the literature, STMF is characterized by: 1) exclusive incidence in the male sex; 2) variable immunoreactivity of the neoplastic cells for desmin, probably reflecting an origin from a peculiar subset of myofibroblasts, or, alternatively, a further myoid differentiation; 3) variable abundance of (hyalinized) collagen; 4) presence of amianthoid fibers. The combination of desmin immunoreactivity, frequently observed in MF of the breast, and amianthoid fibers, the main feature of MF of the lymph nodes, has never been observed in soft tissue MF. It is important to recognize STMF as a specific clinicopathologic entity to avoid confusion with other types of spindle cell proliferation and to differentiate it from other types of myofibromatosis.  相似文献

The interaction of human plasma high density lipoproteins (HDL3) with discoidal complexes of apolipoprotein A-I (apoA-I) and dimyristoyl phosphatidylcholine (DMPC) containing 0, 10, 20 or 30 mol % cholesterol was investigated. Discoidal complexes containing various amounts of cholesterol were prepared by incubating apoA-I and DMPC-cholesterol liposomes for 12 hr at 25 C; the protein-lipid complexes were isolated by gel filtration chromatography on Bio-Gel A15m. Increasing the cholesterol content from 0 to 30 mol % caused a decrease in the fluidity of the discoidal complexes as determined by fluorescence polarization with 1,6-diphenyl-1,3,5-hexatriene; a reduced phase-transition amplitude; a decrease in the ratio of apoA-I to DMPC; and an increase in the width of the discoidal complexes as determined by electron microscopy after negative staining. Incubation of the apoA-I-lipid complexes with HDL3 resulted in a complete breakdown of the discoidal structures and a transfer of DMPC and cholesterol to HDL3. As a result of lipid transfer, there was an increase in the size of HDL3. These in vitro results may be of significance as they relate to the interconversion of HDL subfractions during lipoprotein-lipase-induced lipolysis of triglyceride-rich lipoproteins.  相似文献

Contamination of chemicals or reaction mixtures with iron or stainless steel is likely to take place during chemical processing. If energetic and thermally unstable chemicals are involved in a manufacturing process, contamination with iron or stainless steel can impact the decomposition characteristics of these chemicals and, subsequently, the safety of the processes, and should be investigated. The goal of this project was to undertake a systematic approach to study the impact of iron or stainless steel contamination on the decomposition characteristics of different chemical classes. Differential scanning calorimetry (DSC) was used to study the decomposition reaction by testing each chemical pure, and in mixtures with iron and stainless steel. The following classes of energetic chemicals were investigated: nitrobenzenes, tetrazoles, hydrazines, hydroxylamines and oximes, sulfonic acid derivatives and monomers. The following non-energetic groups were investigated for contributing effects: halogens, hydroxyls, amines, amides, nitriles, sulfonic acid esters, carbonyl halides and salts of hydrochloric acid. Based on the results obtained, conclusions were drawn regarding the sensitivity of the decomposition reaction to contamination with iron and stainless steel for the chemical classes listed above. It was demonstrated that the most sensitive classes are hydrazines and hydroxylamines/oximes. Contamination of these chemicals with iron or stainless steel not only destabilizes them, leading to decomposition at significantly lower temperatures, but also sometimes causes increased severity of the decomposition. The sensitivity of nitrobenzenes to contamination with iron or stainless steel depended upon the presence of other contributing groups: the presence of such groups as acid chlorides or chlorine/fluorine significantly increased the effect of contamination on decomposition characteristics of nitrobenzenes. The decomposition of sulfonic acid derivatives and tetrazoles was not impacted by presence of iron or stainless steel.  相似文献

Early identification of alcohol-related problems is important because these problems are prevalent, pose serious health risks to patients and their families, and are amenable to intervention. Physicians may be able to help patients change their drinking behaviors. The most effective tool for screening is a thorough history of the patient's drinking behavior, designed to identify patterns of alcohol-related difficulties with physical and mental health, family life, legal authorities and employment. Alcohol drinkers can be categorized as at-risk, problem or alcohol dependent, according to a protocol developed by the National Institute on Alcohol Abuse and Alcoholism. The severity of the alcohol problem and the patient's readiness to change should determine the intervention selected by the family physician.  相似文献

Handwritten word recognition is a field of study that has largely been neglected in the psychological literature, despite its prevalence in society. Whereas studies of spoken word recognition almost exclusively employ natural, human voices as stimuli, studies of visual word recognition use synthetic typefaces, thus simplifying the process of word recognition. The current study examined the effects of handwriting on a series of lexical variables thought to influence bottom-up and top-down processing, including word frequency, regularity, bidirectional consistency, and imageability. The results suggest that the natural physical ambiguity of handwritten stimuli forces a greater reliance on top-down processes, because almost all effects were magnified, relative to conditions with computer print. These findings suggest that processes of word perception naturally adapt to handwriting, compensating for physical ambiguity by increasing top-down feedback. OBJECTIVES: The goal of this research is to assess the degree to which the recent growth in the rate of Supplemental Security Income (SSI) usage is concentrated among recently arrived elderly immigrants or among earlier arriving immigrants who have "aged in place" and thus become eligible for benefits. METHODS: We use 1980 and 1990 Census data and 1997 Current Population Survey (CPS) data to examine whether the growth in the elderly noncitizen caseload during the 1980s and 1990s may be attributed to increases in rates of receipt among newly arrived elderly immigrants, to increases in rates of receipt among "settled" immigrants who have aged into categories that allow them to obtain SSI benefits, or to increases in the number of persons in each of these groups. RESULTS: We find that the major contribution to the growth in the noncitizen elderly SSI caseload has been the significant increase in the rate of receipt among those who have lived in the United States for more than 10 years (a smaller increase occurred among recent arrivals). This factor accounts for about half of the total growth in the caseload and cannot be explained by increases in poverty among noncitizens. DISCUSSION: The idea that the availability of SSI for elderly immigrants has acted as a magnet for poor elderly immigrants, thereby accounting for the growth in the elderly immigrant SSI caseloads during the 1980s and 1990s, does not receive much support in the findings of this research.  相似文献

Two groups of Yorkshire gilts (110 d of age) were maintained in two light regimens. Both light regimens consisted of 14 h of light and 10 h of darkness, but were 180 degrees out of phase. Gilts in Group 1 received light from 1200 to 0200 and gilts in Group 2 from 2400 to 1400. At approximately 140 d of age each group was divided into four subgroups of eight gilts each (1A, 1B, 1C, 1D or 2A, 2B, 2C, 2D). All gilts were blood sampled at 2-h intervals for 5 d commencing on d 142. The four subgroups received a single injection of estradiol (15 micrograms/kg body weight) on d 143 at either 2400 (A), 0600 (B), 1200 (C), or 1800 (D). For pigs in Groups 1A and 1D, the injection of estradiol coincided with the animals' "subjective day" and the injections given to Groups 1B and 1C with their "subjective night." When estradiol-17 beta (E2) was administered to the gilts during their subjective day the LH profile showed one peak, whereas when E2 was administered during dark hours the profile exhibited two peaks (P < .0001). In Group 2 for which the light cycle was reversed, the well-defined spikes of LH were found to coincide with the injections of estradiol administered during the dark hours. The impact of childhood victimization and other premilitary factors on warzone abusive violence was examined with 177 Vietnam combat veteran inpatients. Premilitary and military variables were also examined in relationship to postmilitary variables, including violence and PTSD. Statistical analyses showed that none of the premilitary variables predicted warzone violence. High combat exposure did, however, predict warzone abusive violence and PTSD. In addition, participation in warzone violence predicted postmilitary violence to self, spouse, and others. Although high rates of childhood victimization and high levels of combat exposure were found, neither predicted postmilitary violence, criminal activities, drug/alcohol problems, or suicide attempts. Low childhood adjustment ratings and school suspensions predicted adult alcohol abuse and drug abuse, respectively. These findings and their implication for treatment are discussed.  相似文献
