The response to implantation of novel apatite glass–ceramics was evaluated using a weight bearing in vivo bone implant model. Five novel glasses with varying calcium to phosphate ratios were cast as short rods and heat-treated to crystallize principally apatite. One glass ceramic had an apatite stoichiometry (Ca : P=1.67); three were phosphate-rich and one calcium-rich. One of the phosphate-rich glasses was also tested in its glassy state to determine the effect of crystallization on the biological response. Rods were implanted into the midshaft of rat femurs and left for 28 days. The femurs were then harvested and processed for scanning electron microscopy, energy dispersive X-ray microanalysis and conventional histology as ground and polished sections. Four of the materials exhibited evidence of osseointegration and osteoconduction. However, there was a marked inflammatory response to one of the phosphate-rich glass–ceramics, and to the non-crystallized glass. Crystallization of the latter significantly improved the bone tissue response. The glass–ceramic with an apatite stoichiometry elicited the most favorable response and merited further study as an osteoconductive bone substitute in maxillofacial and orthopedic surgery. 相似文献

The advantage of using the genotype of a quantitative trait locus (QTL) in selection schemes of dairy cattle was quantified using stochastic simulation. Three selection plans were studied. In the first plan, young bulls waited for 3 yr until their sisters completed a lactation and then were evaluated and selected based on an animal model. In a second plan, young bulls waited for 5 yr until their daughters completed a lactation. An intermediate 4-yr waiting plan was also studied. Simulation was for 16 yr with overlapping generations. Population and model parameters were proportional to the U.S. Holstein population. The advantage of using a QTL was quantified as the percentage of superiority of QTL-assisted over QTL-free selection using cumulative genetic response. Percentage of superiority was reported for four selection pathways: active sires, young bulls, bull dams, and first lactation cows. A general trend was observed: low superiority in early years of selection that increased to a plateau in later years and then decreased. The superiority of the QTL information was greatest in the 3-yr waiting plan and least in the 4-yr waiting plan. Superiority at plateau for selection pathways ranged from 16 to 26% for the 3-yr waiting plan, from 3 to 12% for the 4-yr waiting plan, and from 5 to 13% for the 5-yr waiting plan. The contribution to selection response attributed to the QTL and the polygenes was quantified. The rate at which the favorable allele approached fixation and the accuracy of predicting breeding values on the percentage of superiority were studied. 相似文献

In this article, I describe the design of an early 1950s attempt to build a relatively low-cost computer, using a mechanical desktop calculator to perform the arithmetic operations. The resulting computer-one of the earliest attempts at building a "minicomputer"-could perform stored-program addition, subtraction, multiplication, and division, as well as carry out a simple decision operation based on a number comparison. The management of risks in projects is a growing area of concern. Both the identification and analysis phases of the risk management process are considered the most important, for they can have a big effect on the precision of the risk assessment exercise. Currently, it is assumed that project managers rely largely on experience to identify project risks. These decisions, influenced by individual perception and attitudes, are made primarily under conditions of uncertainty. Understanding how individuals respond to uncertain situations, therefore, requires an understanding of how individuals intuitively assess the situation they perceive, before expressing a response. The Project Risk Identification (Pro-RIde) project interviewed 51 project managers using active information search (AIS) as a data collection method and cognitive mapping as a data-capturing tool. Our results suggest that the role of experience in the risk identification process is much less significant than it is commonly assumed to be. By contrast, information search style, level of education and risk management training do play a significant role in risk identification performance. This paper builds on previous work in which simple structure ILC algorithms were experimentally evaluated on a non-minimum phase plant. The phase-lead update law, the most promising of those implemented, is examined using an established performance criterion. The use of a forgetting factor is found to overcome the problem of instability, but at the expense of increased final error. This is verified through experimentation. The phase-lead algorithm is improved using additional phase-leads to remove the instability and improve convergence and final error. This technique is generalized to produce an optimization routine which leads to greatly improved results. The learning law utilizing the plant adjoint is found to fit naturally into this framework and practical results are presented to compare their respective performance. This algorithm, which requires a model of the plant, is reformulated into one which does not. Results are presented using this technique and practical guidelines are produced and tested to improve its performance. A simple method of increasing the learning at higher frequencies is proposed and practical limitations are addressed and verified experimentally. All the algorithms tested are compared with respect to their performance, practicality and robustness. 相似文献

OBJECTIVE: The audiologic presentation of vestibular schwannoma (VS) associated with neurofibromatosis type 2 (NF2) has not been well characterized. The goal of this study was to investigate the audiologic features of NF2-associated VS and to determine their relationship to the size of the tumor. STUDY DESIGN: A retrospective case review. SETTING: Quaternary governmental medical research institute evaluating patients fitting specific criteria for ongoing clinical studies. PATIENTS: Audiologic and magnetic resonance imaging data were available for 40 patients (25 males, 15 females), with an average age of 32 years, who had been recruited for ongoing clinical and genetic studies of NF2. MAIN OUTCOME MEASURES: The audiologic profile and magnetic resonance imaging characteristic of tumor were retrospectively reviewed. RESULTS: The average size of the tumor at presentation was 7.26 +/- 16.58 cm3 and measured 1.2, 1.6, and 1.1 cm in the anterior/posterior, lateral/medial, and superior/inferior dimensions, respectively. An increase in lateral/medial size of the tumor most significantly correlated with deterioration in mid- (1,000-2,000 Hz) and high- (4,000-8,000 Hz) frequency hearing levels, elevated speech reception threshold, and prolonged auditory brain stem response waves III and V latency. CONCLUSIONS: Patients with NF2 demonstrate a more predictable audiologic profile for a given size tumor than has been previously described with spontaneous or sporadic VS. 相似文献

Juvenile rats are more susceptible to the acute toxicity of the phosphorothionate insecticides parathion and chlorpyrifos than are adult rats. Developmental changes in brain acetylcholinesterase and hepatic aliesterase (carboxylesterase), cytochrome P450, and the P450-mediated metabolism of these two phosphorothionate insecticides were investigated in male Sprague-Dawley rats. Specific activities of acetylcholinesterase in cerebral cortex, but not medulla oblongata, and of liver aliesterases increased with age, indicating the presence of both more target esterases and more protective esterases, respectively, in the adult compared to the juvenile animal. Sensitivity of the brain acetylcholinesterase to inhibition by paraoxon and chlorpyrifosoxon, as measured by IC50 values, did not change significantly with age, whereas the hepatic aliesterase sensitivity to inhibition decreased with age. Progressive increases in activities of P450-mediated activation (desulfuration) (6- to 14-fold) and detoxication (dearylation) (2- to 4-fold), as well as concentrations of P450 (7-fold) and protein (2-fold), were observed between neonate and adult hepatic microsomes. Microsomal pentoxyresorufin O-dealkylase activity followed a developmental pattern similar to desulfuration and dearylation, displaying a 16-fold increase between neonates and adults. However, microsomal ethoxyresorufin O-deethylase activity increased until 21 days of age, displaying a 16-fold increase, then decreased in adulthood to a level 10-fold higher than neonates. These results indicate that target enzyme sensitivity is not responsible for age-related toxicity differences, nor is the potential for hepatic bioactivation, whereas lower levels of hepatic aliesterase-mediated protection and P450-mediated dearylation probably contribute significantly to the greater sensitivity of juveniles to phosphorothionate toxicity. 相似文献

In most patients with atherosclerosis, the underlying metabolic derangement remains undefined. Animal experiments have suggested that the ability to produce and excrete large amounts of bile acids may be an adaptation mechanism to cholesterol overload protecting against the atherogenic effects of cholesterol. However, there are very few data on bile acid excretion in human atherosclerosis. In the present study, we have investigated fecal bile acid secretion in subjects with and without coronary artery disease. The target group consisted of 30 patients with proven coronary artery disease and the control group consisted of 27 matched subjects without clinical or laboratory evidence of coronary atherosclerosis. Fecal bile acids were measured by gas-liquid chromatography from 24-hr stool collections under a controlled diet. The patients excreted significantly less bile acids than the controls (325+/-135 vs. 592+/-223 mg/day, respectively, p < 0.0001). The difference was primarily due to a reduced excretion of secondary bile acids. Less than 50% of deoxycholate was excreted by patients (180+/-81 mg/day) as compared to controls (367+/-168 mg/day, p < 0.0002), while lithocholic acid excretion was 111+/-62 mg/day in patients vs. 190 +/-70 mg/day in controls (p < 0.005). The fecal output of the two primary bile acids, cholic and chenodeoxycholic acid, did not differ significantly between patients and controls. The fecal output of total bile acids correlated with that of both secondary bile acids in patients as well as in controls. These findings suggest that patients with coronary heart disease are unable to excrete adequate amounts of bile acids to rid themselves of excess cholesterol, even if they are able to maintain a plasma cholesterol level comparable to that of healthy controls. 相似文献
