This report shows (a) that the Core Conflictual Relationship Theme (CCRT) can be rated reliably either from therapy sessions or from a special pretreatment interview in a group of depressed patients, and (b) that the CCRTs obtained before treatment are similar to the CCRTs extracted from the early sessions of brief dynamic psychotherapy. The data suggest that, at least in the early sessions of treatment, the therapist's influence did not significantly alter the patients' CCRTs. The chemical basis of feeding responses to the acceptable nonhost plantVigna sinensis (cowpea) by larvae ofManduca sexta was investigated using chemical isolation techniques directed by a novel chemosensory-based bioassay. The presence of feeding stimulatory and inhibitory compounds in leaves or leaf extracts was determined in a two-choice preference test using leaf disks or glass fiber filter paper disks laced with leaf extract as test substrate and filter paper disks laced with water as control. Larvae strongly prefer the control disks over leaf disks, indicating the presence of feeding inhibitory compounds in the leaf. An ethanol extract of both fresh and dried leaves neither stimulated nor inhibited feeding. The cause of this inactivity was examined by using larvae that respond strongly to either feeding stimulatory or inhibitory compounds due to selective chemosensory deprivation. Larvae having chemosensory organs remaining only on the maxillary palps are stimulated to feed by whole leaf disks and by the ethanol extracts. In contrast, larvae having only the medial and lateral maxillary sensilla styloconica and the epipharyngeal sensilla remaining are strongly inhibited by whole leaf disks and the ethanol extract of fresh leaves. Thus, the ethanol extract contains both feeding stimulatory and inhibitory compounds, which elicit opposite behavioral effects in unoperated larvae, therefore nullifying any stimulatory and inhibitory activity. These compounds can only be demonstrated by using discrimination-enhanced larvae in the choice tests. Further isolation of the feeding stimulatory principle inV. sinensis yielded two separate fractions of neutral compounds, suggesting at least two different chemicals belonging to two different classes: nonpolar and polar lipids. Feeding inhibitory chemicals have apparently polar properties because strong activity was found in the ethyl acetate and methanol extracts of dried leaves. The role of feeding stimulatory and inhibitory compounds in food selection ofM. sexta larvae is discussed.  相似文献

Prenatal cocaine and opiate exposure are thought to subtly compromise social and emotional development. The authors observed a large sample of 236 cocaine-exposed and 459 nonexposed infants (49 were opiate exposed and 646 nonexposed) with their mothers in the face-to-face still-face paradigm. Infant and maternal behaviors were microanalytically coded. No opiate-exposure effects were detected. However, mothers of cocaine-exposed infants showed more negative engagement than other mothers. The cocaine-exposed dyads also showed higher overall levels of mismatched engagement states than other dyads, including more negative engagement when the infants were in states of neutral engagement. Infants exposed to heavier levels of cocaine showed more passive-withdrawn negative engagement and engaged in more negative affective matching with their mothers than other infants. Although effect sizes were small, cocaine exposure, especially heavy cocaine exposure, was associated with subtly negative interchanges, which may have a cumulative impact on infants' later development and their relationships with their mothers. Sphingolipids are normally necessary for growth of Saccharomyces cerevisiae cells, but mutant strains that bypass the need for sphingolipids have been identified. Such bypass mutants fail to grow under stressful conditions, including low pH (pH 4.1), when they lack sphingolipids. To begin to understand why sphingolipids seem to be necessary for coping with low-pH stress, we screened a genomic library and selected a suppressor gene, CWP2 (cell wall protein 2), that when present in multiple copies partially compensates for the lack of sphingolipids and enhances survival at low pH. To explain these results, we present evidence that sphingolipids are required for a normal rate of transport of glycosylphosphatidylinositol (GPI)-anchored proteins, including Cwp2 and Gas1/Gpg1, from the endoplasmic reticulum (ER) to the Golgi apparatus. The effect of sphingolipids is specific for transport of GPI-anchored proteins because no effect on the rate of transport of carboxypeptidase Y, a non-GPI-anchored protein, was observed. Since the Gasl protein accumulated in the ER with a GPI anchor in cells lacking sphingolipids, we conclude that sphingolipids are not necessary for anchor attachment. Therefore, sphingolipids must be necessary for a step in formation of COPII vesicles or for their transport to the Golgi apparatus. Our data identify the Cwp2 protein as a vital component in protecting cells from the stress of low pH.  相似文献

We investigate a mechanism causing shorting of large area GaSb diodes grown on GaSb substrates using molecular beam epitaxy (MBE). The source of these shorts is determined to be large crystallographic defects on the surface of the diodes that are formed around droplets of gallium ejected from the gallium Knudsen cells during MBE. The gallium droplets cause defects in the crystal structure, and, as the epitaxy continues, the gallium is incorporated into the surrounding material. The shape of the defects is pyramidal with a central void extending from the epi-surface to the gallium core. Processing a GaSb diode with these surface defects results in the top-side contact metal migrating into the defect and shorting the diode. This prevents realization of large area diodes that are critical to applications such as photovoltaics and detectors. The diodes in this study are electrically characterized and the defect formation mechanism is investigated using cross-section transmission electron microscopy and electron dispersive spectroscopy.  相似文献
