A 17 amino acid peptide containing the arginine-rich region of the HIV Rev protein binds specifically to Rev response element (RRE) RNA. Even though it is highly charged, the peptide forms an alpha helix in solution, but only when its N- and C-termini are modified to provide favorable electrostatic interactions with the helix macrodipole. Binding affinity for IIB RNA (the primary binding site within the RRE) increases with alpha helix content, whereas nonspecific binding affinity is independent of helix content. Binding of mutant peptides demonstrates that one threonine, one asparagine, and four arginine side chains are important for sequence-specific recognition. Transactivation of the HIV LTR using Tat-Rev peptide hybrids and the RRE IIB site indicates that the peptide adopts an alpha-helical conformation in vivo. The results suggest that interactions with the RNA backbone may help to orient the alpha helix in the major groove of RNA.  相似文献

This paper deals with how the Hydrogen Energy may play a crucial role in taking care of the environmental scenario/climate change. The R&D efforts, at the Hydrogen Energy Center, Banaras Hindu University have been described and discussed to elucidate that hydrogen is the best option for taking care of the environmental/climate changes. All three important ingredients for hydrogen economy, i.e., production, storage and application of hydrogen have been dealt with. As regards hydrogen production, solar routes consisting of photoelectrochemical electrolysis of water have been described and discussed. Nanostructured TiO2 films used as photoanodes have been synthesized through hydrolysis of Ti[OCH(CH3)2]4. Modular designs of TiO2 photoelectrode-based PEC cells have been fabricated to get high hydrogen production rate (10.35 lh−1 m−2). However, hydrogen storage is a key issue in the success and realization of hydrogen technology and economy. Metal hydrides are the promising candidates due to their safety advantage with high volume efficient storage capacity for on-board applications. As regards storage, we have discussed the storage of hydrogen in intermetallics as well as lightweight complex hydride systems. For intermetallic systems, we have dealt with material tailoring of LaNi5 through Fe substitution. The La(Nil  xFex)5 (x = 0.16) has been found to yield a high storage capacity of 2.40 wt%. We have also discussed how CNT admixing helps to improve the hydrogen desorption rate of NaAlH4. CNT (8 mol%) admixed NaAlH4 is found to be optimum for faster desorption (3.3 wt% H2 within 2 h). From an applications point of view, we have focused on the use of hydrogen (stored in intermetallic La–Ni–Fe system) as fuel for Internal Combustion (IC) engine-based vehicular transport, particularly two and three-wheelers. It is shown that hydrogen used as a fuel is the most effective alternative fuel for circumventing climate change.  相似文献

BACKGROUND: To provide information concerning the geographical distribution of selenium (Se) in the soils of Scotland, we analysed 47 arable soils selected on the basis of their parent rock, which were expected to have relatively high, low or unclassified Se concentrations. To investigate relationships between the actual minerals in the soils and the aqua regia extractable Se concentration of the soil, soil minerals were quantified by X‐ray diffraction. RESULTS: The aqua regia extractable Se concentrations of the soils were between 0.19 and 1.46 mg kg?1. No simple correlation between the aqua regia extractable Se concentrations of the soil and the parent rock classification estimated by soil survey was evident. Partial least squares analysis revealed that the aqua regia extractable Se concentration of the soils was positively related to loss on ignition (LOI) or C concentration and negatively related to the K‐feldspar concentration, with other minerals being less important. CONCLUSION: The Se concentration of arable topsoils from Scotland is more related to LOI or carbon concentration, with parent material being less important. In situ analyses with a laser ionization mass spectrometer have shown that a large fraction of aerosols in the bottom few kilometers of the stratosphere contain small amounts of mercury (1). Electron microscopy of particles collected near the tropopause has also detected mercury. The distribution of mercury onto many particles, including those less than 20 nm in diameter, indicates that the mercury is from local condensation of mercury compounds onto particles rather than transport of mercury-rich aerosols from surface sources. Although the results are only semiquantitative, they suggest that most of the mercury in the lower stratosphere is converted into the particulate phase. Mercury-containing particles were present at both middle latitudes and the tropics in two seasons. There is therefore good reason to believe that particulate mercury above the tropopause is global and could affect the atmospheric lifetime of mercury. There are indications that bromine and/ or iodine may be involved in the conversion of mercury from the gas to particle phase. Measurements at altitudes below 5 km did not find mercury in any particles despite sampling some particles that clearly originated in the stratosphere. This indicates that the particulate mercury from the lower stratosphere may be volatile enough to evaporate or decompose once particles reach warmer temperatures.  相似文献

Graphene oxide (GO) has been prepared by employing modified Staudenmaier's method through thermal exfoliation of graphite oxide. High pressure hydrogen sorption isotherms up to 50 bar of GO, reduced by thermal reduction (TR-GO), chemical reduction (CR-GO) and graphene sheets decorated with Fe nanoclusters (Fe-GS) have been investigated. Thermal reduction of GO at 623 K under high vacuum yields TR-GO. Chemical reduction of GO using hydrazine forms CR-GO. Fe-GS was synthesized through arc-discharge between the ends of two graphite rods with one rod carrying Fe nanoparticles. The surface areas of these graphene samples were determined from the nitrogen adsorption isotherm employing Brunauer, Emmett and Teller (BET) method. Kelvin's equation was used to determine the pore size distribution of all graphene based samples. Hydrogen pressure-composition isotherms (PCI) were determined at 300 K and at 77 K, between 0.1 and 50 bar. Further, in this paper, we present a comparative adsorption isotherm analysis of hydrogen and helium on TR-GO. This reveals that the volume of hydrogen and helium adsorbed by TR-GO is nearly equal. The similar uptake volume determined for both hydrogen and helium indicates the possibility of monolayer adsorption of hydrogen and also nearly similar binding energy between TR-GO and H2/He.  相似文献

Although Concentrated Animal Feeding Operations (CAFOs) have been identified as potentially important sources for the release of estrogens into the environment, information is lacking on the concentrations of estrogens in whole lagoon effluents (including suspended solids) which are used for land application. Lagoons associated with swine, poultry, and cattle operations were sampled at three locations each for direct analysis for estrogens by GC/ MS/MS and estrogen conjugates by LC/MS/MS. Estrogen conjugates were also analyzed indirectly by first subjecting the same samples to enzyme hydrolysis. Solids from centrifuged samples were extracted for free estrogens to estimate total estrogen load. Total free estrogen levels (estrone, 17alpha-estradiol, 17beta-estradiol, estriol) were generally higher in swine primary (1000-21000 ng/L), followed by poultry primary (1800-4000 ng/L), dairy secondary (370-550 ng/L), and beef secondary (22-24 ng/L) whole lagoon samples. Swine and poultry lagoons contained levels of 17(alpha-estradiol comparable to those of 17beta-estradiol. Confirmed estrogen conjugates included estrone-3-sulfate (2-91 ng/L), 17beta-estradiol-3-sulfate (8-44 ng/L), 17alpha-estradiol-3-sulfate (141-182 ng/L), and 17beta-estradiol-17-sulfate (72-84 ng/L) in some lagoons. Enzymatic hydrolysis indicated the presence of additional unidentified estrogen conjugates not detected bythe LC/MS/MS method. In most cases estrogen conjugates accounted for at least a third of the total estrogen equivalents. Collectively, these methods can be used to better determine estrogen loads from CAFO operations, and this research shows that estrogen conjugates contribute significantly to the overall estrogen load, even in different types of CAFO lagoons.  相似文献
