We demonstrate a strategy employing collision-induced dissociation for phosphopeptide discovery, followed by targeted electron capture dissociation (ECD) for site localization. The high mass accuracy and low background noise of the ECD mass spectra allow facile sequencing of coeluting isobaric phosphopeptides, with up to two isobaric phosphopeptides sequenced from a single mass spectrum. In contrast to the previously described neutral loss dependent ECD method, targeted ECD allows analysis of both phosphotyrosine peptides and lower abundance phosphopeptides. The approach was applied to phosphorylation analysis of human Sprouty2, a regulator of receptor tyrosine kinase signaling. Fifteen sites of phosphorylation were identified, 11 of which are novel.  相似文献

Multiferroic materials displaying coupled ferroelectric and ferromagnetic order parameters could provide a means for data storage whereby bits could be written electrically and read magnetically, or vice versa. Thin films of Aurivillius phase Bi6Ti2.8Fe1.52Mn0.68O18, previously prepared by a chemical solution deposition (CSD) technique, are multiferroics demonstrating magnetoelectric coupling at room temperature. Here, we demonstrate the growth of a similar composition, Bi6Ti2.99Fe1.46Mn0.55O18, via the liquid injection chemical vapor deposition technique. High‐resolution magnetic measurements reveal a considerably higher in‐plane ferromagnetic signature than CSD grown films (MS=24.25 emu/g (215 emu/cm3), MR=9.916 emu/g (81.5 emu/cm3), HC=170 Oe). A statistical analysis of the results from a thorough microstructural examination of the samples, allows us to conclude that the ferromagnetic signature can be attributed to the Aurivillius phase, with a confidence level of 99.95%. In addition, we report the direct piezoresponse force microscopy visualization of ferroelectric switching while going through a full in‐plane magnetic field cycle, where increased volumes (8.6% to 14% compared with 4% to 7% for the CSD‐grown films) of the film engage in magnetoelectric coupling and demonstrate both irreversible and reversible magnetoelectric domain switching.  相似文献

Trehalose is a disaccharide produced by many organisms to better enable them to survive environmental stresses, including heat, cold, desiccation, and reactive oxygen species. Mammalian cells do not naturally biosynthesize trehalose; however, when introduced into mammalian cells, trehalose provides protection from damage associated with freezing and drying. One of the major difficulties in using trehalose as a cellular protectant for mammalian cells is the delivery of this disaccharide into the intracellular environment; mammalian cell membranes are impermeable to the hydrophilic sugar trehalose. A panel of cell‐permeable trehalose analogues, in which the hydrophilic hydroxyl groups of trehalose are masked as esters, have been synthesized and the ability of these analogues to load trehalose into mammalian cells has been evaluated. Two of these analogues deliver millimolar concentrations of free trehalose into a variety of mammalian cells. Critically, Jurkat cells incubated with these analogues show improved survival after heat shock, relative to untreated Jurkat cells. The method reported herein thus paves the way for the use of esterified analogues of trehalose as a facile means to deliver high concentrations of trehalose into mammalian cells for use as a cellular protectant.  相似文献

The ability to monitor supplier performance is a critical capability for maintaining strong buyer–supplier relationships. Monitoring type B suppliers is especially challenging as they are not as clearly defined as either type A, with strong strategic partnerships, or type C, with little partnership. This research develops a non-parametric multivariate Hoteling’s T 2 control chart to capture the in-control state of a dyadic relationship (Phase I), and show how it would be developed based on survey data of buyer–supplier relationship attributes. Modelling the satisfactory level of dyadic relationship performance is very useful for identifying when the relationship begins to move away from the desired state. We then use the designed control chart to monitor the relationship between dyads over time to determine if any unusual behaviour has occurred (Phase II) and illustrate its implementation through a case study from the auto industry. This research illustrates how supply chain managers can secure and improve their supply chain performance by monitoring and maintaining strong relationships with their partners. The proposed method extends the existing SPC tools to effectively manage Type B buyer–supplier relationships.  相似文献

SCOOP is a concurrent programming language with a new semantics for contracts that applies equally well in concurrent and sequential contexts. SCOOP eliminates race conditions and atomicity violations by construction. However, it is still vulnerable to deadlocks. In this paper we describe how far contracts can take us in verifying interesting properties of concurrent systems using modular Hoare rules and show how theorem proving methods developed for sequential Eiffel can be extended to the concurrent case. However, some safety and liveness properties depend upon the environment and cannot be proved using the Hoare rules. To deal with such system properties, we outline a SCOOP Virtual Machine (SVM) as a fair transition system. The SVM makes it feasible to use model-checking and theorem proving methods for checking global temporal logic properties of SCOOP programs. The SVM uses the Hoare rules where applicable to reduce the number of steps in a computation. P. J. Brooke, R. F. We present a method to design control charts such that in‐control and out‐of‐control run lengths are guaranteed with prespecified probabilities. We call this method the percentile‐based approach to control chart design. This method is an improvement over the classical and popular statistical design approach employing constraints on in‐control and out‐of‐control average run lengths since we can ensure with prespecified probability that the actual in‐control run length exceeds a desired magnitude. Similarly, we can ensure that the out‐of‐control run length is less than a desired magnitude with prespecified probability. Some numerical examples illustrate the efficacy of this design method.  相似文献

Pseudomonas aeruginosa (P. aeruginosa), one of the dangerous multidrug resistance pathogens, orchestrates virulence factors production through quorum sensing (QS). Since the exploration of QS inhibitors, targeting virulence to circumvent bacterial pathogenesis without causing significant growth inhibition is a promising approach to treat P. aeruginosa infections. The present study has evaluated the anti-QS and anti-infective activity of epigallocatechin-3-gallate (EGCG), a bioactive ingredient of the traditional green tea, against P. aeruginosa. EGCG showed significant inhibitory effects on the development of biofilm, protease, elastase activity, swimming, and swarming motility, which was positively related to the production of C4-AHL. The expression of QS-related and QS-regulated virulence factors genes was also evaluated. Quantitative PCR analysis showed that EGCG significantly reduced the expression of las, rhl, and PQS genes and was highly correlated with the alterations of C4-AHL production. In-vivo experiments demonstrated that EGCG treatment reduced P. aeruginosa pathogenicity in Caenorhabditis elegans (C. elegans). EGCG increased the survival of C. elegans by 23.25%, 30.04%, and 36.35% in a dose-dependent manner. The findings of this study strongly suggest that EGCG could be a potential candidate for QS inhibition as an anti-virulence compound against bacterial infection.  相似文献

This paper presents an energy demand model for a fleet of plug-in fuel cell vehicles and a medium-sized commercial/office building interfaced with a clean energy hub. The approach taken is to model the architecture and daily operation of every individual vehicle in the fleet. A simplified architecture model was developed, with daily operation divided into two periods: charging and travelling. During the charging period, the vehicle charges its batteries and refills its compressed hydrogen tanks. During the travelling period, the vehicle depletes the batteries and hydrogen tanks based on distance travelled. Daily travel distance is generated by a stochastic model. The modeling of the clean energy hub is also presented. The clean energy hub functions as an interface between electricity supply and the energy demand (i.e. hydrogen and electricity) of the vehicle fleet and the commercial building. Finally, a sample case is presented to demonstrate the use of the models.  相似文献
