Defective repair of oxidative damage in mitochondrial DNA in Down''s syndrome

BACKGROUND: Synthetic homopyrimidine peptide nucleic acids (PNAs) can bind complementary targets in double-stranded DNA, generating strand-displacement complexes, and so offering an opportunity to modulate specific gene expression. Several issues remain to be addressed before these attributes can be exploited in vivo, however. RESULTS: The kinetics of the interaction between a homopyrimidine PNA and a complementary homopurine target on double-stranded DNA were analyzed in the presence or absence of a preformed strand-displacement complex proximal to the target. The complex was established under low salt conditions by the binding of a different homopyrimidine PNA to a target situated adjacent to the first PNA target. These two targets were placed next to each other on opposite strands at distances of 0, 2, 4 and 8 base pairs apart. The presence of a preformed strand-displacement complex near the target accelerates the binding of PNA to double-stranded DNA in a salt-dependent manner. The influence of salt on the binding rates was also examined. The binding rate is increased by a factor of 1 x exp(70[NaCl]), that is, 16-fold at 40 mM NaCl and more than 10(4)-fold if extrapolated to 140 mM NaCl. This effect is significantly reduced if the two targets are 2 base pairs apart and completely absent if the distance is 4 base pairs or more. CONCLUSIONS: The perturbation of the DNA helix imposed by a PNA strand-displacement complex only propagates a few base pairs. It is therefore possible to target sites in the immediate vicinity of strand invasion complexes specifically. The results presented have implications for the mechanism of strand displacement and for the application of PNA in a genomic context.     

Tuberculosis cutis miliaris disseminata due to multidrug-resistant Mycobacterium tuberculosis in AIDS patients

Two patients with AIDS and disseminated tuberculosis characterized by cutaneous involvement are reported. They developed a maculopapular skin eruption, from which a multidrug-resistant Mycobacterium tuberculosis strain was isolated. In both cases the clinical course was rapidly fatal. Tuberculosis cutis miliaris disseminata should be differentiated from the skin lesions frequently seen in HIV-infected patients, especially from folliculitis. In patients with tuberculosis, the appearance of cutaneous lesions may be due to the haematogenous dissemination of mycobacteria. Therefore, early identification of the causative organism by use of optimal microbiological methods is fundamental.     

Familial essential hypertension and HLA

Ten families living in Shenyang city with multiple cases of essential hypertension were studied by HLA haplotype analysis. They were selected strictly in accordance with the methods of sibling pair analysis and lods analysis. The obtained ratio of HLA haplotype sharing among affected siblings were 28.6%(2): 52.4%(1): 19.0%(0), P > 0.9. It is indicated that the haplotype shared randomly among affected siblings. The results of Lods analysis were Q = 0.40, Lods = 0.046, Pr = 1.112. These results showed that there is no evidence of patients with essential hypertension susceptible to gene linked with HLA.     

Quantitative evaluation of somatotrophic cells (SC) in the adenohypophysis of normal and diabetic male Chinese hamsters

Free amino acid determinations in the cerebrospinal fluid of patients with Parkinson's disease or other extrapyramidal disorders showed that in both groups most amino acids were increased. Significantly increased amino acids were neutral and basic amino acids. Amino acid clusters were present and corresponded with the grouping of amino acids in accordance with amino acid transport systems. The amino acids within the clusters showed a remarkable degree of correlation, and there was a notable correlation between the clusters both in the parkinsonian group and in the group of extrapyramidal disorders, but not in a control group. Our data suggest that in a number of extrapyramidal disorders, Parkinson's disease included, a more generalized abnormality exists than neuronal loss only and, in addition, that patients with these neurologic conditions have a common biochemical defect in their amino acid transport systems.     

A Jordan control canonical form for singular systems

A canonical form for controllable singular systems modulo restricted system equivalence is presented. The canonical form respects the decomposition of the singular system into a regular and an impulsive part. The construction is based on Popov's control canonical form and on the Jordan canonical form recently developed by Hinrichsen and Prätzel-Wolters. Continuity properties are analysed and the cellular partition of the orbit space induced by the canonical form is briefly discussed.     

随机算法异步并行化的效率分析

随机算法的执行时间具有不确定性,这种不确定性为随机算法的异步并行提供了良好的基础,已有许多计算实验表明了随机算法的异步并行可以达到线性甚至超线性的加速.对于求解SAT问题的随机算法RDP,研究了异步并行效率与运行时间分布和处理器数目之间的关系.应用一种单峰分布──分段线性分布模型来模拟随机算法的运行时间分布.理论分析和计算结果均表明:当处理器数目k较小和单峰位于分布的前部时,随机算法的异步并行具有近线性加速.     

Use of the glycosylated hemoglobin test in outpatient practice

Haemoglobin glycosilate was evaluated using the bio-rad method in 129 diabetics during their periodic visits to outpatient departments. A significant correlation between this test and metabolic compensation was revealed in confirmation of results reported in most research so far carried out. The advantages of using this test in outpatients departments is also emphasised.     

Corneal endothelial cell measurements in megalocornea

A pedigree of megalocornea manifested many of the characteristic features as well as some less common features of the disorder. Endothelial specular microscopy of affected patients disclosed normal endothelial cell densities and morphologic characteristics and increased total endothelial cell populations, suggesting a process of total corneal hyperplasia. In contrast, unpublished data indicate diminished densities in congenital glaucoma, a finding more consistent with corneal distention. We conclude that specular microscopy may be of value in differentiating the two disorders, although larger series are necessary for confirmation.
