We conducted an experiment to determine the proportion of the lysine requirement of lactating sows that can be met using L-lysine x HCl. A total of 247 Pig Improvement Company (PIC) sows (parity one to four) were randomly allotted to one of five experimental diets containing .79% apparently digestible lysine. The first four diets contained 0, .075, .150, and .225% L-lysine x HCl replacing the intact lysine, primarily derived from soybean meal. Dietary crude protein was reduced from 17.9 to 16.9, 15.8, and 14.8% respectively. The fifth diet contained .174% L-lysine x HCl (15.5% CP) with added synthetic methionine, threonine, and tryptophan to restore the ratios of these amino acids to lysine to those in the control diet with no synthetic amino acids. The average lactation length was 15.7 +/- .3 d. Diet did not affect ADFI, sow backfat loss, sow loin eye area loss, or weaning-to-mating interval. Sows consumed an average of 4.6 kg/d and were provided 36 g/d of digestible lysine. Replacing soybean meal with increasing levels of L-lysine x HCl did not affect sow weight change. The number of pigs weaned decreased and preweaning mortality increased linearly (P = .08) with increasing levels of L-lysine x HCl. Litters from sows fed the .174% L-lysine x HCl with added methionine, threonine, and tryptophan grew slower and had a higher mortality rate than litters from sows fed no synthetic amino acids (P < .05). The addition of synthetic methionine, threonine, and tryptophan to the .174% L-lysine x HCl diet did not improve litter growth rate, but it did increase preweaning mortality (P = .05) and decrease the number of pigs weaned (P = .06) compared to the .15% L-lysine x HCl with no additional synthetic amino acids. These additions also resulted in an increased sow weight loss (P = .10). These results suggest that when more than .075% L-lysine x HCl is used to meet the lysine requirement preweaning mortality is increased and the number of pigs weaned is decreased. Supplementation with methionine, threonine, and tryptophan failed to ameliorate the negative response associated with L-lysine x HCl, which suggests that other amino acids may be limiting.  相似文献

Rapid Viscoanalysis was investigated as a method off determining the malting quality of barley and malt, to define the conditions required to select progeny with high malting potential from a barley breeding programme. Studies were made on (i) varying physical parameters and chemical additions, (ii) the effect of modification, (iii) the prediction of malting potential from viscograms, (iv) the relationship of the viscometric characteristics of starch to malting quality and (v) the use of a Rapid Viscoanalyser (RVA) in a commercial meltings. The results will be presented as a series of papers. In this paper, physical and chemical factors were examined. Firstly, sample/liquid ratios, initial and final temperatures, speed off mixing and the fineness of grinding of the samples were varied. The effect of the time of standing of malt slurries was also investigated. The peak areas, peak viscosities and Stirring Numbers were shown to obey a relationship with the cube of the sample concentration under a wide range of conditions. Comparisons with the Amylograph and Falling Number Apparatus are reported. Conditions of grinding, temperature, speed of mixing and grist/liquid ratios for the routine examination of barleys and malts were defined. Secondly, calcium-complexing agents, acids, alkalies and the salts of heavy metals were tested for their effects on the Rapid Viscogram of malt. Unless substantial time was allowed for reaction to occur, calcium-complexing agents per se had no effect on viscograms: acidic radicles were necessary. Although both acids and alkalies affected viscograms, inhibition could be reversed by neutralisation. Silver nitrate is suggested as the most suitable chemical for inhibition of enzymes for the determination of the intrinsic viscosity of malts with an RVA.  相似文献

BACKGROUND: Thrombolysis to normal flow in patients with acute myocardial infarction preserves left ventricular function and decreases mortality. Failure of early reperfusion, reocclusion, or residual thrombus may be due to concurrent activation of the platelet-coagulation system. Thus, we hypothesized that the best concomitant antithrombotic therapy (recombinant [r]-hirudin, heparin, or aspirin) will maximally accelerate thrombolysis by r-tissue-type plasminogen activator (rTPA) and reduce residual thrombus. METHODS AND RESULTS: Occlusive thrombi were formed in the carotid arteries of 29 pigs (by balloon dilatation followed by endarterectomy at the site of injury-induced vasospasm) and matured for 30 minutes before rTPA was started, with or without antithrombotic therapy. Thrombolysis was assessed with the use of angiography and measurement of residual thrombus. Pigs were allocated to one of five treatments: placebo, rTPA, rTPA plus r-hirudin, rTPA plus heparin, or rTPA plus intravenous aspirin. No placebo-treated pig reperfused. Two of six animals treated with rTPA alone reperfused compared with seven of seven animals treated with rTPA plus r-hirudin (reperfusion time, 33 +/- 10 minutes), six of seven animals treated with rTPA plus heparin (reperfusion time, 110 +/- 31 minutes), and two of six animals with rTPA plus aspirin. The activated partial thromboplastin time was prolonged in only the rTPA plus r-hirudin group (25 +/- 0.1 times baseline) and the rTPA plus heparin group (5.3 +/- 0.2 times baseline). Residual 111In-platelet and 125I-fibrin(ogen) depositions were lower in the heparin-treated group and lowest in the r-hirudin-treated group (heparin versus hirudin, respectively; incidence of residual macroscopic thrombus was six of six animals versus two of seven [P = .01]; 125I-fibrin(ogen), 170 +/- 76 versus 48 +/- 6 x 10(6) molecules/cm2 [P = .02]; 111In-platelets, 47 +/- 15 versus 13 +/- 2 x 10(6)/cm2, P = .10). No pigs developed spontaneous bleeding. CONCLUSIONS: Thrombin inhibition with heparin or r-hirudin significantly accelerated thrombolysis of occlusive platelet-rich thrombosis, but only the best antithrombotic therapy (r-hirudin) eliminated or nearly eliminated residual thrombus.  相似文献

We evaluated the contribution of nitric oxide (NO) to morphine-induced rounding of spontaneously activated (mobile) ameboid human monocytes, granulocytes, or arterial endothelial cells and invertebrate immunocytes and microglia. Morphine induced significant rounding and inactivation of ameboid cells within 20 min except for arterial endothelial cells, which became rounded 24 h after morphine exposure. The effects of morphine on cell conformation were blocked in the presence of N-nitro-L-arginine, a nitric oxide synthase inhibitor. Treatment of cells with the NO donor, sodium nitroprusside, induced cell rounding similar to that observed following morphine exposure, suggesting that NO release may mediate morphine-induced changes in cell conformation. The contribution of NO release to morphine-induced cell rounding was determined by direct evaluation of NO concentration in real-time using a NO-specific amperometric probe. Significant increases in NO concentration were observed 2 min after morphine stimulation, whereas morphine-induced NO release was markedly impaired by pretreatment with N-nitro-L-arginine or the opiate alkaloid antagonist, naloxone. In contrast, opioid peptides failed to induce NO release, consistent with our previous observations that demonstrated the failure of opioid peptides to promote cell rounding. Taken together, these data suggest that morphine-induced NO release may be mediated by activation of the opiate alkaloid-selective, opioid peptide-insensitive micro3 receptor, and that functional coupling of morphine to NO production has been conserved during evolution and may modulate cellular activation.  相似文献

Cardiac L-carnitine content, essential for mitochondrial fatty acid transport and ATP-ADP exchange, decreases during ischemia. In animal models, administration of the natural derivative, L-propionylcarnitine, may reduce ischemia and improve cardiac function. To evaluate possible antiischemic effects of L-propionylcarnitine was compared with placebo in a randomized, double-blind, parallel design, in addition to preexisting therapy. Patients with > or = 2 anginal attacks per week and objective signs of ischemia with angina during bicycle exercise testing were included. After an initial 2-week, single-blind placebo phase, 37 patients received 500 mg L-propionylcarnitine tid, and 37 patients received placebo for 6 weeks. Both groups were comparable at baseline. Three patients discontinued the study while on placebo (two because of noncompliance, one because of palpitations) and one while on L-propionylcarnitine (noncompliance). Although heart rate, blood pressure at rest, and maximal exercise were not affected, L-propionylcarnitine increased the time to 0.1 mV ST-segment depression [44 +/- 3 vs. 8 +/- 2 seconds (mean +/- SEM) in the placebo group; p = 0.05], and exercise duration improved by 5% compared with placebo. Anginal attacks and the consumption of nitroglycerin were not affected in either group. Thus, following a 6 week treatment period, L-propionylcarnitine induced additional, albeit marginal, antiischemic effects in anginal patients who were still symptomatic despite maximal conventional antianginal therapy. It is questionable whether in these patients this form of metabolic treatment will achieve great benefit, although in some improvement can be expected.  相似文献

A double column and double pump HPLC switching system is described for the analysis of cefixime in human plasma and urine. The system used muBondapak C18 short pretreatment column for on-line sample clean-up and a Hitachi GEL 3056 (ODS) analytical column for separation. A mixed solution of 0.01 mol/L H3PO4-0.1 mol/L KH2PO4-H2O (20:1:79) was used as the pretreatment mobile phase and CH2CH-0.01 mol/L H3PO4-0.1 mol/L KH2PO4-H2O (13:20:1:66) was used as analytical mobile phase. The compound in plasma and urine is detected by ultraviolet absorption at 286 nm and 314 nm, respectively. The absolute recoveries of the method in plasma and urine were 99.1% and 98.6% respectively. The relative standard deviations of the method are 0.70-3.82% and 0.80-3.73% in plasma, 1.53-3.08% and 1.31-2.67% in urine between days and day-to-day. Linear calibration curve for cefixime was measured over the range of 0.1-3.2 micrograms/ml in plasma and 1.0-32.0 micrograms/ml in urine, and the correlation coefficients were all 0.9999. The detection limit was 0.05 micrograms/ml in plasma and 0.2 micrograms/ml in urine. The plasma and urine samples were diluted with water and injected directly onto the HPLC system. The operation is simple and the relative sensitivity is markedly increased because of higher recoveries and larger loading capacity of the sample.  相似文献

Increase in heterogeneous growth as a result of grouping in the Nile tilapia, Oreochromis niloticus (L.), is presumed to be partially promoted by the social stress imposed by the dominant fish on the subordinates. Such stress may decrease the energy available for growth. In this study, the effect of social stress on carbohydrate metabolism was studied in adult, growing males. All animals were deprived of food during the experimentation period and pairing was imposed for either 2 or 4 days. Glycemia was measured before and after pairing, and muscle and liver glycogen contents were determined only after pairing. Subordinate fish showed the highest consumption of carbohydrate reserves. This response was caused by the social stress imposed which corroborates the idea that metabolic differences promoted by social stress may be involved in the rouping effect on heterogeneous growth in the Nile tilapia.  相似文献

Magnetic resonance imaging is frequently complicated by the presence of motion and susceptibility gradients. Also, some biologic tissues have short T2s. These problems are particularly troublesome in fast spin-echo (FSE) imaging, in which T2 decay and motion between echoes result in image blurring and ghost artifacts. The authors reduced TE in conventional spin-echo (SE) imaging to 5 msec and echo spacing (E-space) in FSE imaging to 6 msec. All magnetic gradients (except readout) were kept at a maximum, with data sampling as fast as 125 kHz and only ramp waveforms used. Truncated sinc radio-frequency pulses and asymmetric echo sampling were also used in SE imaging. Short TE (5.8 msec) SE images of the upper abdomen were compared with conventional SE images (TE = 11 msec). Also, FSE images with short E-space were compared with conventional FSE images in multiple body sites. Short TE significantly improved the liver-spleen contrast-to-total noise ratio (C/N) (7.9 vs 4.1, n = 9, P < .01) on T1-weighted SE images, reduced the intensity of ghost artifacts (by 34%, P < .02), and increased the number of available imaging planes by 30%. It also improved delineation of cranial nerves and reduced susceptibility artifacts. On short E-space FSE images, spine, lung, upper abdomen, and musculoskeletal tissues appeared crisper and measured spleen-liver C/N increased significantly (6.9 vs 4.0, n = 12, P < .01). The delineation of tissues with short T2 (eg, cartilage) and motion artifact suppression were also improved. Short TE methods can improve image quality in both SE and FSE imaging and merit further clinical evaluation.  相似文献

In a coalfield of noted high-chlorine coals, seam profiles were taken from two collieries. Three seam profiles from each colliery were analysed for total chlorine to try to correlate chlorine level with position in the seam. Moisture contents and ash yields were determined for four of the seams. Total chlorine is highest in the coals, reaching 1 wt%, and low in the mudrocks, down to 0.08 wt%. Intermediate chlorine values are associated with dirty coals and carbonaceous mudrocks. Water centrifuged from an intraseam dirt band showed that the moisture held the total chlorine content. Similar element ratios were achieved from water centrifuged from coal. These results, together with National Coal Board groundwater data, suggest that the majority of chlorine in coal has been introduced by chloride-rich groundwaters which evolved during diagenesis, as has been suggested for Illinois and Nottinghamshire coals. There is a very significant positive correlation between coal moisture and total chlorine contents. Total chlorine levels are thus thought to be controlled by groundwater concentration levels and coal porosity, hence the positive relationship between organic carbon and chlorine concentrations. Total chlorine levels vary through the seams, but there is not a consistent change with position in the seam. There exists a correlation between coal type and total chlorine content. Bright coals hold significantly more chlorine than dull coals, owing to different porosities of the coal macerals. Departures from this relationship with coal type may be explained by mineral matter dilution or the ingress of recent groundwaters, overriding the original coal type-chlorine relationship.  相似文献
